The bone and muscle tissues share several catabolic pathways that lead to variable degrees of disability in SCI patients. In this review article, we provide a comprehensive characterization of the available treatment options targeting the skeleton and the bone in the setting of SCI. Among the pharmacological intervention, bisphosphonates, anti-sclerostin monoclonal antibodies, hydrogen sulfide, parathyroid hormone, and RANKL pathway inhibitors represent valuable options for treating bone alterations. Loss phenomena at the level of the muscle can be counteracted with testosterone, anabolic-androgenic steroids, and selective androgen receptor modulators. Exercise and physical therapy are valuable strategies to increase bone and muscle mass. Nutritional interventions could enhance SCI treatment, particularly in the setting of synergistic and multidisciplinary interventions, but there are no specific guidelines available to date. The development of multidisciplinary recommendations is required for a proper clinical management of SCI patients. Here, we evaluated the influence of thyroid dysfunction on gestational anemia via published epidemiological articles and a new prospective study conducted by our team, respectively. Methods We searched studies on the PubMed, Embase, MEDLINE, and Cochrane databases as of November 2019, and conducted a prospective study in which participants underwent thyroid function and blood routine testing throughout pregnancy. Results The meta-analysis showed that pregnancies with overt hypothyroidism [OH; odds ratio (OR) = 3.74, 95% confidence interval (CI) 1.95-7.15] or that were thyroid peroxidase antibody (TPOAb)-positive (OR = 1.97, 95%CI 1.19-3.26) had increased anemia risk, but similar results were not found in pregnancies with subclinical hypothyroidism (SCH) and hyperthyroidism. In the prospective study from our new data, the hypothyroid group had significant reductions in hemoglobin (Hb) (P = 0.048) and increased anemia risk (OR = 6.384, 95%CI 2.498-16.311) during the second half of pregnancy. From the first to second half of pregnancy, the longitudinal reductions in Hb, erythrocyte (RBC), and hematocrit (Hct) levels were significantly increased in hypothyroid group. Conclusions Our meta-analysis indicates that untreated OH or TPOAb-positive pregnant women have increased risk of anemia. In addition, our new data showed that treated hypothyroidism is also a risk factor for anemia in the second half of pregnancy rather than in the first half. The results may guide strengthening of Hb monitoring in pregnancies with thyroid dysfunction. In this review, we present the advantages and adverse effects of SGLT2 inhibitors plus insulin therapy as a treatment regimen for patients with type 2 diabetes (T2D). Compared with placebo, SGLT2 inhibitors plus insulin therapy could significantly decrease fasting blood glucose and HbA1c, thereby reducing the daily required dose of insulin. Beige fat is unique due to its origin and its development in white fat. Subsequently, both brown fat and beige fat have become viable targets to combat obesity. Over the last few decades, most therapeutic strategies have been focused on the canonical pathway of thermogenic fat activation via the β3-adrenergic receptor (AR). Notwithstanding, administering β3-AR agonists often leads to side effects including hypertension and particularly cardiovascular disease. It is thus imperative to search for alternative therapeutic approaches to combat obesity. In this review, we discuss the current challenges in the field with respect to stimulating brown/beige fat thermogenesis. Additionally, we include a summary of other newly discovered pathways, including non-AR signaling- and non-UCP1-dependent mechanisms, which could be potential targets for the treatment of obesity and its related metabolic diseases.
