A population-based shape model is required to generate accurate and accessible musculoskeletal geometry in a paediatric population. The pelvis, femur, and tibia/fibula were segmented from 333 CT scans of children aged 4-18 years. Bone morphology variation was captured using principal component analysis (PCA). Subsequently, a shape model was developed to predict bone geometry from demographic and linear bone measurements and validated using a leave one out analysis. The shape model was compared to linear scaling of adult and paediatric bone geometry. The PCA captured growth-related changes in bone geometry. The shape model predicted bone geometry with root mean squared error (RMSE) of 2.91 ± 0.99 mm in the pelvis, 2.01 ± 0.62 mm in the femur, and 1.85 ± 0.54 mm in the tibia/fibula. Linear scaling of an adult mesh produced RMSE of 4.79 ± 1.39 mm in the pelvis, 4.38 ± 0.72 mm in the femur, and 4.39 ± 0.86 mm in the tibia/fibula. CD8+-T-cells, CD4+-T-cells, Treg, NK cells and macrophages were individually depleted through either direct administration of anti-marker antibodies/reagents or using DTR (diphtheria toxin receptor) knock-in mice, for some syngeneic tumors, where specific subsets were depleted following diphtheria toxin (DT) administration. These LOF experiments revealed distinctive intrinsic tumor immunity and thus different MOAs in their responses to anti-PD-1 antibody among different syngeneic tumors. Despite recent efforts towards classifying structural variants (SVs), more robust statistical frames are needed to better classify these variants and isolate those that derive from specific molecular mechanisms. We present a new statistical approach to analyze SVs patterns from 2392 tumor samples from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium and identify significant recurrence, which can inform relevant mechanisms involved in the biology of tumors. The method is based on recursive KDE clustering of 152,926 SVs, randomization methods, graph mining techniques and statistical measures. The proposed methodology was able not only to identify complex patterns across different cancer types but also to prove them as not random occurrences. Furthermore, a new class of pattern that was not previously described has been identified.It remains unclear how tropical cyclones (TCs) decay from their ocean lifetime maximum intensity (LMI) to landfall intensity (LI), yet this stage is of fundamental importance governing the socio-economic impact of TCs. Here we show that TCs decay on average by 25% from LMI to LI. A logistic decay model of energy production by ocean enthalpy input and surface dissipation by frictional drag, can physically connect the LMI to LI. The logistic model fits the observed intensity decay as well as an empirically exponential decay does, but with a clear physical foundation. The distance between locations of LMI and TC landfall is found to dominate the variability of the decay from the LMI to LI, whereas environmental conditions are generally less important. A major TC at landfall typically has a very large LMI close to land. The LMI depends on the heating by ocean warming, but the LMI location is also important to future landfall TC intensity changes which are of socio-economic importance.Fishery-dependent data are frequently used to inform management decisions. However, inferences about stock development based on commercial data such as Catch-Per-Unit-Effort (CPUE) can be severely biased due to a phenomenon known as technological creep, where fishing technology improves over time. Here we show how trap improvement over nine decades has driven technological creep in a European lobster (Homarus gammarus) fishery. We combined fishing data, experimental fishing with contemporary and older trap types, and information on depletion effects during fishing seasons. The resulting standardized CPUE time series indicates a 92% decline in lobster abundance between 1928 and 2019 compared to 70% if technological creep is not corrected for. Differences are most pronounced within the last 40 years when the most substantial shift in gear technology occurred an uncorrected CPUE index suggests an 8% increase in lobster abundance during this period, while the corrected CPUE index declined by 57%. We conclude that technological creep has masked a continuous stock decline, particularly in recent decades and largely driven by the shift from one- to two-chambered traps, as well as the ability of newer trap designs to capture larger lobsters.
