KEGG pathway analysis revealed that more DEGs were associated with the pathways of the two-component system and pyruvate metabolism. Moreover, the gene SE1634 code for staphylococcal toxin was significantly upregulated in S. epidermidis strains of the endophthalmitis, which might be directly responsible for the pathogenesis of endophthalmitis. In conclusion, this research is helpful for further investigations on genes or pathways related with the pathogenesis and therapeutic targets of S. epidermidis endophthalmitis.Flavonols play key roles in many plant defense mechanisms, consequently they are frequently investigated as stress sensitive factors in relation to several oxidative processes. It is well known that grapevine (Vitis vinifera L.) can synthesize various flavonol glycosides in the leaves, however, very little information is available regarding their distribution along the cane at different leaf levels. In this work, taking into consideration of leaf position, the main flavonol glycosides of a red grapevine cultivar (Cabernet Sauvignon) were profiled and quantified by HPLC-DAD analysis. It was found that amount of four flavonol glycosides, namely, quercetin-3-O-galactoside, quercetin-3-O-glucoside, kaempferol-3-O-glucoside and kaempferol-3-O-glucuronide decreased towards the shoot tip. Since leaf age also decreases towards the shoot tip, the obtained results suggest that these compounds continuously formed by leaf aging, resulting in their accumulation in the older leaves. KU-0063794 mTOR inhibitor In contrast, quercetin-3-O-glucuronide (predominant form) and quercetin-3-O-rutinoside were not accumulated significantly by aging. We also pointed out that grapevine boosted the flavonol biosynthesis in September, and flavonol profile differed significantly in the two seasons. Our results contribute to the better understanding of the role of flavonols in the antioxidant defense system of grapevine.Development of spermatozoa requires remodelling and formation of particular structures. In elongating spermatids, the transient microtubular manchette contributes to the formation of the head-tail coupling apparatus (HTCA) and the sperm tail. The HTCA derives from the centrosome in that the proximal centriole inserts into the nuclear indentation and the distal centriole gives rise to the sperm flagellum. Although impairments in the formation of HTCA and sperm tail cause male infertility their molecular constituents are only partially known. The WD40-protein CFAP52 is implicated in motile cilia, but its relevance for male germ cell differentiation is not known. Here we show that CFAP52 is widespread expressed and localizes to a subset of microtubular structures. In male germ cells, CFAP52 is a component of the transient manchette and the sperm tail. However, expression of Cfap52 is not restricted to motile cilia-bearing cells. In NIH3T3 cells, CFAP52 localizes to the centrosome, the basal body, and the mitotic spindle poles, but not to the primary cilium. Our results demonstrate that CFAP52 is not restricted to motile cilia but instead most likely functions in constituting the centrosome/basal body matrix and the sperm tail.We construct an experimental setup, consisting of conical refraction transformation in two biaxial cascade crystals and 4f-system, to realize Unitary transformation of light beam and the manipulation of Poincaré beams on the different parts of Poincaré sphere. The spatial structure of the polarization can be controlled by changing the polarization of the incident beam or rotating the angle between these two crystals. The beams with different SoPs covering the full-Poincaré sphere, part-Poincaré sphere and one point on the sphere are generated for the different angles between crystals. The Unitary transformation of light beam is proposed in the experiment with the invariant intensity distribution. Subsequently, the spin angular momentum is derived from the distribution of polarization measured in our experiment. Moreover, the conversion between orbital angular momentum and spin angular momentum of light beam is obtained by changing the angle between crystals. And the conversion progress can also be influenced by the polarization of incident beam. We realized the continuous control of the spatial structure of the angular momentum density, which has potential in the manipulation of optical trapping systems and polarization-multiplexed free-space optical communication. In depth studies of quantitative trait loci (QTL) can provide insights to the genetic architectures of complex traits. A major effect QTL at the distal end of chicken chromosome 1 has been associated with growth traits in multiple populations. This locus was fine-mapped in a fifteen-generation chicken advanced intercross population including 1119 birds and explored in further detail using 222 sequenced genomes from 10 high/low body weight chicken stocks. We detected this QTL that, in total, contributed 14.4% of the genetic variance for growth. Further, nine mosaic precise intervals (Kb level) which contain ancestral regulatory variants were fine-mapped and we chose one of them to demonstrate the key regulatory role in the duodenum. This is the first study to break down the detail genetic architectures for the well-known QTL in chicken and provides a good example of the fine-mapping of various of quantitative traits in any species.Agar have long been used as a growth media for plants. Here, we made agar media with embedded fluidic channels to study the effect of exposure to nutrient solution on root growth and pull-out force. Black Eye bean (Vigna Unguiculata) and Mung bean (Vigna Radiata) were used in this study due to their rapid root development. Agar media were fabricated using casting process with removable cores to form channels which were subsequently filled with nutrient solution. Upon germination, beans were transplanted onto the agar media and allowed to grow. Pull-out force was determined at 96, 120 and 144 h after germination by applying a force on the hypocotyl above the gel surface. The effect of nutrients was investigated by comparing corresponding data obtained from control plants which have not been exposed to nutrient solution. Pull-out force of Black Eye bean plantlets grown in agar with nutrient solution in channels was greater than those grown in gel without nutrients and was 110% greater after 144 h of germination.
