In this review, we introduced and summarized the cellular processes that govern hypoxia-driven regulation of histone methylation in the context of cancer biology. We also discussed the emerging roles of histone methyltransferases and demethylases in epigenetic response to hypoxia.Tumors can be classified as cold or hot according to the degree of immune cell infiltration into tumor tissues; cold tumors are insensitive to either chemotherapy or immunotherapy and are associated with poor prognosis. Recent studies have shown that STAT3 signaling molecules hinder the conversion of cold to hot tumors by regulating immunosuppressive molecule secretion and immunosuppressive cell functions. This review aims to present the most recent studies on how STAT3 regulates cold tumor formation and discuss its research status in cancer therapy. We also present insight for designing new therapeutic strategies to "heat" tumors and provide a reference for tumor immunotherapy.Background Acral melanoma (AM) is an epidemiologically and molecularly distinct entity that is underrepresented in clinical trials on immunotherapy in melanoma. We aimed to analyze the real-world efficacy of anti-PD-1 antibodies in advanced AM. Patients and methods We retrospectively evaluated unresectable stage III or stage IV AM patients treated with an anti-PD-1 antibody in any line at 21 Japanese institutions between 2014 and 2018. The clinicobiologic characteristics, objective response rate (ORR, Response Evaluation Criteria in Solid Tumors), survival estimated using Kaplan-Meier analysis, and toxicity (Common Terminology Criteria for Adverse Events 4.0.) were analyzed to estimate the efficacy of the anti-PD-1 antibodies. Results In total, 193 patients (nail apparatus, 70; palm and sole, 123) were included in the study. Anti-PD-1 antibody was used as first-line therapy in 143 patients (74.1%). Baseline lactate dehydrogenase (LDH) was within normal level in 102 patients (52.8%). The ORR of all patients was 16.6% (complete response, 3.1%; partial response, 13.5%), and the median overall survival (OS) was 18.1 months. Normal LDH levels showed a significantly stronger association with better OS than abnormal levels (median OS 24.9 vs 10.7 months; P less then 0.001). Although baseline characteristics were similar between the nail apparatus and the palm and sole groups, ORR was significantly lower in the nail apparatus group (6/70 patients [8.6%] vs 26/123 patients [21.1%]; P=0.026). Moreover, the median OS in this group was significantly poorer (12.8 vs 22.3 months; P=0.03). Conclusions Anti-PD-1 antibodies have limited efficacy in AM patients. Notably, patients with nail apparatus melanoma had poorer response and survival, making nail apparatus melanoma a strong candidate for further research on the efficacy of novel combination therapies with immune checkpoint inhibitors.The publisher regrets that this article has been temporarily removed. Methods Forty acid-etched dentin surfaces were randomly allocated into five groups, and pretreated with one of the primers 0.1, 1.0, and 10 mmol/L DMA/ dimethyl sulfoxide (DMSO) solutions, pure DMSO, and deionized water (control) before bonding. The bonded teeth of each group were randomly distributed into an immediate subgroup (water storage at 37 °C for 24 h) and an aged subgroup (thermocycling, 5-55 °C, 10,000 times). The teeth were sectioned into 0.9 mm slabs; two middle slabs were used for nanoleakage evaluation, while the remaining slabs were sectioned into beams for micro-tensile bond strength (MTBS) test. Fractured mode and nanoleakage were observed under SEM. selleck FTIR was used to assess the effect of primers on the degree of conversion (DC) of adhesive. Results The MTBS was significantly affected by primers (p 0.05). Dopamine methacrylamide might strengthen the integrity of hybrid layer by serving as a bridge connecting the demineralized collagen fibrils and adhesive polymer network.Objective This cross-sectional study aims to examine if cognitive impairment, depressive symptoms, and their combination are associated with biases between self-evaluated and measured masticatory performance in community-dwelling older Japanese adults. Methods The sample constituted of 804 community-dwelling elderly Japanese individuals aged ≥70 years. Cognitive impairment and depressive symptoms were assessed using the Mini-Mental State Examination and Geriatric Depression Scale, respectively. Self-evaluated masticatory performance (SMP) was assessed for 10 food items. Measured masticatory performance (MMP) was assessed as the maximum occlusal force, which is a surrogate indicator. Multivariate logistic regression was applied to examine the associations of cognitive impairment, depressive symptoms, and their combination with the deviation between SMP and MMP, SMP overestimation, and SMP underestimation, with adjustment for age, sex, body mass index, smoking, alcohol consumption, duration of education, medicastimate their self-evaluated masticatory performance. Conversely, the participants with depression symptoms tend to underestimate their self-evaluated masticatory performance.Objectives This study explored hydrophilic dyes as photosensitizers for application in dental adhesives. The goal was to identify dyes that enhance the degree of conversion (DC) of the hydrophilic-rich phase without impairing polymerization of the hydrophobic-rich phase. Methods Properties that were investigated included the molar extinction coefficient at 480 nm, relative normalized photon absorption efficiency (PAE), rate of polymerization and degree of conversion (DC). The following hydrophilic dyes Bromophenol blue sodium salt, Rosebengal sodium salt, Erythrosin B, New Fuchsin and Victoria blue B were identified as suitable photosensitizers. Results In this study it was observed that dyes such as Bromophenol blue sodium salt, New Fuchsin, Victoria blue B and Rosebengal sodium salt were suitable candidates for dental adhesive photopolymerization, leading to substantial degree of conversion to both the hydrophilic-rich phase and the hydrophobic-rich phase. Conclusions In addition to the ability of the photosensitizer to absorb light in the visible range and transition to an excited state as a result of the absorbed energy, other factors such as the efficiency of the photosensitizer/light curing unit (LCU) combination, stability/efficiency of the excited state of the photosensitizer and/or initiating reactive species play an important role in the photopolymerization of the dental adhesive.
