With the development of basic research, some genetic-based methods have been found to treat Duchenne muscular dystrophy (DMD) with large deletion mutations and nonsense mutations. Appropriate therapeutic approaches for repairing multiple duplications are limited. We used the CRISPR (clustered regularly interspaced short palindromic repeat)/Cas9 system with patient-derived primary myoblasts to correct multiple duplications of the dystrophin gene. Muscle tissues from a patient carrying duplications of dystrophin were obtained, and tissue-derived primary cells were cultured. Myoblasts were purified with an immunomagnetic sorting system using CD56 microbeads. After transduction by lentivirus with a designed single guide RNA (sgRNA) targeting a duplicated region, myoblasts were allowed to differentiate for 7 days. Copy number variations in the exons of the patient's myotubes were quantified by real-time PCR before and after genetic editing. Western blot analysis was performed to detect the full-length dystrophin pngth dystrophin. All experimental results from LC-MS, FT-IR, 1H and 13C-NMR characterizations support the molecular structure of Oleuropein. Electrochemical and gravimetric tests were used to evaluate the corrosion inhibition capabilities of Oleuropein. According to polarization investigation, Oleuropein is a mixed-type inhibitor. Oleuropein's inhibitory efficacy increases with concentration, attaining an optimum value (98.92%) at 100 mg L-1. At high temperatures, Oleuropein can be considered an efficient inhibitor. Thermodynamic variables for the activation operation and copper dissolution were computed and addressed as well. This in-vitro study sheds light on a missing link in the formation of E. faecalis biofilm in which the Ca2+ in Ca(OH)2.We report on unusual regimes of operation of a laser with a gain medium with a large Raman scattering cross-section, which is often inherent in new types of gain media such as colloidal and epitaxial quantum dots and perovskite materials. These media are characterized by a strong electron-phonon coupling. Using the Fröhlich Hamiltonian to describe the electron-phonon coupling in such media, we analyze the operation of the system above the lasing threshold. We show that below a critical value of the Fröhlich constant, the laser can only operate in the conventional regime namely, there are coherent cavity photons but no coherent phonons. Above the critical value, a new pump rate threshold appears. Above this threshold, either joint self-oscillations of coherent phonons in the gain medium and photons in a cavity or a chaotic regime are established. We also find a range of the values of the Fröhlich constant, the pump rate, and the resonator eigenfrequency, in which more than one dynamical regime of the system is stable. In this case the laser dynamics is determined by the initial values of the resonator field, the active medium polarization, the population inversion, and phonon amplitude.Dactylopius opuntiae (Cockerell) (Hemiptera Dactylopiidae) or prickly pear cochineal, is the most damaging pest on cactus species with heavy economic losses worldwide. The efficacy of two Moroccan EPN isolates; Steinernema feltiae (Filipjev) (Rhabditida Steinernematidae) and Heterorhabditis bacteriophora (Poinar) (Rhabditida Heterorhabditidae) (applied at 25, 50, and 75 IJs cm-2) against D. opuntiae nymphs and young females were evaluated under both laboratory bioassays and field conditions. Results showed that S. feltiae was more effective, causing higher mortality of nymphs and adult females (98.8% and 97.5%, respectively) after 8 days of exposure, resulting in an LT50 value of 5.9 days (nymph) and 6.0 days (young female). While, H. bacteriophora had lower mortalities (83.8% for nymph and 81.3% for adult females). For the cochineal nymphs and adult females, no significant difference was observed among S. feltiae at 25, 50, and 75 IJs cm-2, and the positive control, D-limonene applied at 0.5 g/L which was used due to its high effectiveness against nymphs and females of D. opuntiae. Thus, in addition to D-limonene, both Moroccan EPN isolates S. feltiae, and H. bacteriophora could be used as part of the integrated pest management strategy against D. opuntiae. Assessing the coordination level between genes from single-cell transcriptomic data, without a priori knowledge of the map of gene regulatory interactions, is a challenge. A 'top-down' approach has recently been developed to analyze single-cell transcriptomic data by evaluating the global coordination level between genes (called GCL). Here, we systematically analyze the performance of the GCL in typical scenarios of single-cell RNA sequencing (scRNA-seq) data. We show that an individual anomalous cell can have a disproportionate effect on the GCL calculated over a cohort of cells. In addition, we demonstrate how the GCL is affected by the presence of clusters, which are very common in scRNA-seq data. Finally, we analyze the effect of the sampling size of the Jackknife procedure on the GCL statistics. The manuscript is accompanied by a description of a custom-built Python package for calculating the GCL. These results provide practical guidelines for properly pre-processing and applying the GCL measure in transcriptional data.The neurobiological basis of learning is reflected in adaptations of brain structure, network organization and energy metabolism. However, it is still unknown how different neuroplastic mechanisms act together and if cognitive advancements relate to general or task-specific changes. Therefore, we tested how hierarchical network interactions contribute to improvements in the performance of a visuo-spatial processing task by employing simultaneous PET/MR neuroimaging before and after a 4-week learning period. We combined functional PET and metabolic connectivity mapping (MCM) to infer directional interactions across brain regions. Learning altered the top-down regulation of the salience network onto the occipital cortex, with increases in MCM at resting-state and decreases during task execution. Accordingly, a higher divergence between resting-state and task-specific effects was associated with better cognitive performance, indicating that these adaptations are complementary and both required for successful visuo-spatial skill learning. Simulations further showed that changes at resting-state were dependent on glucose metabolism, whereas those during task performance were driven by functional connectivity between salience and visual networks. Referring to previous work, we suggest that learning establishes a metabolically expensive skill engram at rest, whose retrieval serves for efficient task execution by minimizing prediction errors between neuronal representations of brain regions on different hierarchical levels.The Indian and Pacific Oceans surround the Maritime Continent (MC). Major modes of sea surface temperature variability in both oceans, including the Indian Ocean Dipole (IOD) and El Niño-Southern Oscillation (ENSO), can strongly affect precipitation on the MC. The prevalence of fires in the MC is closely associated with precipitation amount and terrestrial water storage in September and October. Precipitation and terrestrial water storage, which is a measurement of hydrological drought conditions, are significantly modulated by Indian Ocean Dipole (IOD) and El Niño events. We utilize long-term datasets to study the combined effects of ENSO and the IOD on MC precipitation during the past 100 years (1900-2019) and find that the reductions in MC precipitation and terrestrial water storage are more pronounced during years when El Niño and a positive phase of the IOD (pIOD) coincided. The combined negative effects are produced mainly through an enhanced reduction of upward motion over the MC. Coincident El Niño-pIOD events have occurred more frequently after 1965. However, climate models do not project a higher occurrence of coincident El Niño-pIOD events in a severely warming condition, implying that not the global warming but the natural variability might be the leading cause of this phenomenon.This meta-analysis compared the outcomes of transradial access (TRA) and transfemoral access (TFA) in chronic total occlusion (CTO) percutaneous coronary intervention (PCI) in recent decades. We searched multiple databases for articles published between January 1, 2015, and December 31, 2020. Six observational studies with 11,736 patients were analyzed. Data included baseline demographics, Japan-chronic total occlusion (J-CTO) score, sheath size, PCI vessel, retrograde method, procedural time, fluoroscopy time, and contrast volume. The more prevalent target CTO vessel was the left coronary artery in the TRA group and the right coronary artery in the TFA group. Higher J-CTO score, longer procedural time, and more contrast volume were seen in the TFA group. In comparison, the TRA group had better procedural success rate (odds ratio (OR), 0.846; 95% confidence interval (CI) 0.749-0.956) and less vascular complications (OR, 0.323; 95% CI 0.203-0.515), but similar retrograde success rate (OR, 0.965; 95% CI 0.382-2.
