2% and a linearity of less than 200 ppm, which indicates an overall superior performance compared to representative comparable CTs based on alternative technologies. In addition to the areas of application mentioned above, the new type of DC-CT can be used for general purpose metering, measurement instrumentation, and high power DC and AC systems. The endogenous PP2A inhibitor SET Nuclear Proto-Oncogene (SET) has been reported to play oncogenic roles and determines poor outcomes in colorectal cancer (CRC). Our group previously showed that miR-199b is deregulated in metastatic CRC, and reduced the cell viability and enhanced the sensitivity of CRC cells to standard induction chemotherapy drugs, mainly through direct negative SET regulation. Clinically, miR-199b downregulation was identified as the molecular mechanism responsible for SET overexpression in around half of metastatic CRC patients. However, the potential clinical value of miR-199b in early-stage CRC remains totally unknown. Thus, here we explored the expression levels of this microRNA in a cohort of 171 early-stage CRC patients using real-time polymerase chain reactions. MiR-199b downregulation was found in 21.6% of cases (37 out of 171) and was significantly associated with those patients with a worse Eastern Cooperative Oncology Group (ECOG) status (p = 0.045). Moreover, miR-199b downregulation predicted shorter overall (p less then 0.001) and progression-free survival (p = 0.015). As expected, we next immunohistochemically analyzed SET, observing that it was significantly associated with miR-199b in our cohort. However, multivariate analyses showed that miR-199b was an independent biomarker of poor outcomes in early-stage CRC with a predictive value stronger than SET. In conclusion, our results highlight the potential clinical usefulness of miR-199b and suggest that it could represent a novel molecular target in this disease.Fluorescent probes have been employed for more than half a century to study the structure and dynamics of model and biological membranes, using spectroscopic and/or microscopic experimental approaches. While their utilization has led to tremendous progress in our knowledge of membrane biophysics and physiology, in some respects the behavior of bilayer-inserted membrane probes has long remained inscrutable. check details The location, orientation and interaction of fluorophores with lipid and/or water molecules are often not well known, and they are crucial for understanding what the probe is actually reporting. Moreover, because the probe is an extraneous inclusion, it may perturb the properties of the host membrane system, altering the very properties it is supposed to measure. For these reasons, the need for independent methodologies to assess the behavior of bilayer-inserted fluorescence probes has been recognized for a long time. Because of recent improvements in computational tools, molecular dynamics (MD) simulations have become a popular means of obtaining this important information. The present review addresses MD studies of all major classes of fluorescent membrane probes, focusing in the period between 2011 and 2020, during which such work has undergone a dramatic surge in both the number of studies and the variety of probes and properties accessed.Globally, a substantial proportion of existing indigenous and naturalized foods are often neglected thereby narrowing the food-base available to humans. The current study explored the use and contribution of indigenous and naturalized plants to the households' food-pool among 12 communities in the North West Province of South Africa. An ethnobotanical survey was conducted among 133 rural households across the 12 selected communities from the four district municipalities in North West Province, South Africa. We analyzed the utilization patterns for 31 selected indigenous and naturalized plants (grains, fruits, and vegetables) among the 133 households using two ethnobotanical indices. In reference to the checklist of 31 plants, the sampled households utilize approximately 94% (29) as staple foods, beverages, fruits and fodder. Sorghum bicolor, Vigna unguiculata, Amaranthus sp., Sclerocarya birrea, Persea americana, and Mimusops zeyheri were among the top-six plants based on the Relative Frequency of Citation (RFC, 40-83%). In terms of the different crop-types, Sorghum bicolor (grain), Amaranthus sp. (vegetable), and Sclerocarya birrea (fruit) were the top-ranked plants based on the Species Popularity Index (SPI, 0.53-0.83) among the participants. Overall, there is a need for a renaissance of indigenous and naturalized plants, which has the potential to encourage rural farmers to further embrace the cultivation of these plants on a larger scale so as to enhance food security in the rural communities.Acellular particles (extracellular vesicles and membraneless condensates) have important research, drug discovery, and therapeutic implications. However, their isolation and retrieval have faced enormous challenges, impeding their use. Here, a novel size-guided particle purification liquid chromatography (PPLC) is integrated into a turbidimetry-enabled system for dye-free isolation, online characterization, and retrieval of intact acellular particles from biofluids. The chromatographic separation of particles from different biofluids-semen, blood, urine, milk, and cell culture supernatants-is achieved using a first-in-class gradient size exclusion column (gSEC). Purified particles are collected using a fraction collector. Online UV-Vis monitoring reveals biofluid-dependent particle spectral differences, with semen being the most complex. Turbidimetry provides the accurate physical characterization of seminal particle (Sp) lipid contents, sizes, and concentrations, validated by a nanoparticle tracking analysis, transmission electron microscopy, and naphthopyrene assay. Furthermore, different fractions of purified Sps contain distinct DNA, RNA species, and protein compositions. The integration of Sp physical and compositional properties identifies two archetypal membrane-encased seminal extracellular vesicles (SEV)-notably SEV large (SEVL), SEV small (SEVS), and a novel nonarchetypalμμembraneless Sps, herein named membraneless condensates (MCs). This study demonstrates a comprehensive yet affordable platform for isolating, collecting, and analyzing acellular particles to facilitate extracellular particle research and applications in drug delivery and therapeutics. Ongoing efforts focus on increased resolution by tailoring bead/column chemistry for each biofluid type.
