Chinese herbs are a useful resource bank for natural drug development, and have attracted considerable attention to exploit quorum sensing inhibitors (QSIs). This study was designed to screen QSIs from raw Chinese herb materials. Of the 38 common herbs examined, the ethanol extract of Campsis grandiflora flower had the strongest QSI activity. The C. grandiflora flower ethanol extract (CFEE) was purified by HPD600, and the QSI activities were examined in further detail. CFEE inhibited violacein production of Chromobacterium violaceum 026 in a dose-dependent manner, and inhibit the swarming abilities of Escherichia coli K-12 and Pseudomonas aeruginosa PAO1. Furthermore, CFEE could inhibited biofilm formation and destroyed mature biofilms of E. coli K-12 and P. aeruginosa PAO1. The composition of CFEE was determined by UPLC-MS/MS to distinguish active QSI compounds, and 21 compounds were identified. In addition to gallic acid and caffeic acid, two organic acids, malic acid and succinic acid, were confirmed for the first time to have autoinducer type 1 QSI activities. Therefore, CFEE is a potential QSI that could be used as a novel antimicrobial agent and should be considered for medicinal development.Short day length is associated with reduced milk production in dairy ruminants. Dairy ruminants have been kept in lit sheds during winter to extend the day length and stimulate milk production. However, there studies are few on the effect of an extended photoperiod on the ensuing reproductive performance of dairy goats. The aim of this study was to examine the effect of long day photoperiod (LDPP) and exposure to bucks on milk production and plasma progesterone and prolactin in dairy goats. The study was conducted in 122 non-pregnant lactating dairy goats over 18 weeks from April to August (late autumn and winter in the Southern Hemisphere). The goats were kept in open sided sheds in which the control treatment received ambient lighting while the LDPP treatment received 16 h of light, including artificial lighting. In June, July and August synchronised does were randomly assigned each month to the presence or absence of a buck and ovulatory activity determined from plasma progesterone. Plasma progesterone concentrations were reduced (0.73 vs. 0.46 pmol, p less then 0.001) while prolactin concentrations were increased (0.095 vs. 1.33 ng/mL, p less then 0.001) in LDPP goats. The former response was most marked in late winter (0.58 vs. 0.004 pmol, p less then 0.001) indicating a lack of functional corpora lutea. Also, persistency of lactation was greater in LDPP goats with fewer goats drying off (13 vs. 0%, p less then 0.05). These findings suggest that LDPP can increase milk production and persistence while decreasing ovulatory activity in dairy goats.Astilbin and neoastilbin are two flavonoid stereoisomers. In the present study, their solubility, stability, and bioavailability were compared in a rat. The results revealed that the water solubility of astilbin and neoastilbin was 132.72 μg/mL and 217.16 μg/mL, respectively. The oil-water distribution coefficient (log P) of astilbin and neoastilbin in simulated gastric fluid (SGF) was 1.57 and 1.39, and in simulated intestinal fluid (SIF) was 1.09 and 0.98, respectively. In SIF, about 78.6% astilbin remained after 4 h of incubation at 37 °C, while this value was 88.3% for neoastilbin. Most of the degraded astilbin and neoastilbin were isomerized into their cis-trans-isomer, namely neoisoastilbin and isoastilbin, respectively, and the decomposed parts were rare. For bioavailability comparison in a rat, an HPLC method for trace amounts of astilbin and neoastilbin determination in plasma was developed, and the pretreatment of plasma was optimized. EVs are cell derived particles found in body liquids, especially blood, from which they are isolated for further analysis. However, techniques for their isolation are not fully standardized and require further improvement. Herein modification of polypropylene (PP) tubes by cold Atmospheric Pressure Plasma Jet (APPJ) is suggested to minimize the EVs to surface binding and thus increase EVs isolation yields. The influence of gaseous plasma treatment on surface morphology was studied by Atomic Force Microscopy (AFM), changes in surface wettability by measuring the Water Contact Angle (WCA), while surface chemical changes were analyzed by X-Ray Photoelectron Spectroscopy (XPS). Moreover, PP tubes from different manufacturers were compared. The final isolation yields of EVs were evaluated by flow cytometry. The results of this study suggest that gaseous plasma treatment is an intriguing technique to uniformly alter surface properties of PP tubes and improve EVs isolation yields up to 42%.
