To selectively target telomeric G-quadruplex (G4) DNA, monomeric and dimeric naphthalene diimides (NDIs) were investigated as binders of multimeric G4 structures able to discriminate duplex DNA. These NDIs were analysed by the affinity chromatography-based screening G4-CPG (G-quadruplex on Controlled Pore Glass), using the sequence d[AGGG(TTAGGG)7] (tel46), folding into two consecutive G4s, as model of the human telomeric G4 multimer. In parallel, a telomeric G4 monomer (tel26) and a duplex structure (ds27) were used as controls. According to G4-CPG screening, NDI-5 proved to be the best ligand in terms of dimeric G4 vs. duplex DNA selectivity and was analysed by circular dichroism (CD), gel electrophoresis, isothermal titration calorimetry (ITC) and fluorescence spectroscopy in its interactions with tel46. NDI-5 strongly binds and stabilizes tel46 G4, favouring a hybrid folding in K+-containing buffer. Under these conditions, the binding process comprises a first event involving three molecules of NDI-5 and a second one in which other six molecules bind to the DNA. In a metal cation-free system, NDI-5 induces tel46 G4 folding, as indicated by CD and PAGE, favouring an antiparallel structuring. This review highlights experimental evidences supporting that the carbohydrate polymers containing phenolic polysaccharides may become promising drug candidate for the prevention of aging and age related diseases.As a heparin analogue, sulfonated chitosan (SCS) has been confirmed to have similar structure and properties to heparin which is shown to be a linker molecule having specific binding sites with collagen fibrils. In this study, the effects of a varying concentration of SCS on the self-assembly process of type I collagen were investigated. The study on intermolecular interaction between collagen and SCS was carried out via using ultraviolet-visible (UV-vis) spectrophotometry and circular dichroism (CD) spectroscopy. The addition of SCS did not disrupt the triple helix conformation of collagen. However, the decreased value of Rpn showed that the SCS, to some extent, influenced the percentage of triple helix conformation. The turbidity measurements revealed that the self-assembly rate was increased in the presence of a low concentration of SCS whereas decreased with further increasing the SCS concentration. The observation of microstructure via scanning electron microscopy (SEM) and atomic force microscopy (AFM) exhibited the characteristic D-periodicity, indicating that the presence of SCS did not disrupt the self-assembly nature of collagen. Moreover, the addition of SCS facilitated the lateral aggregation of fibrils, leading to the formation of larger fibrils. The rheological analysis showed that the gelation time of collagen was prolonged with increasing the concentration of SCS, in support of a longer lag-phase duration detected in turbidimetric measurements. We expect that valuable data would be provided in this study for further developing of ECM analogues, and propitious performances could be endowed to these biomimetic materials after SCS incorporation.

Development of a tissue-engineered construct for hepatic regeneration remains a challenging task due to the lack of an optimum environment that support the growth of hepatocytes. Hydrogel systems possess many similarities with tissues and have the potential to provide the microenvironment essential for the cells to grow, proliferate, and remain functionally active.

In this work, fibrin (FIB) incorporated injectable alginate dialdehyde (ADA) - gelatin (G) hydrogel was explored as a matrix for liver tissue engineering. ADA was prepared by periodate oxidation of sodium alginate. An injectable formulation of ADA-G-FIB hydrogel was prepared and characterized by FTIR spectroscopy, Scanning Electron Microscopy, and Micro-Computed Tomography. HepG2 cells were cultured on the hydrogel system; cellular growth and functions were analyzed using various functional markers.

FTIR spectra of ADA-G-FIB depicted the formation of Schiff's base at 1608.53cm

with a gelation time of 3 min. ADA-G-FIB depicted a 3D surface topography with a pore size in the range of 100-200μm.
