A study was conducted at the Escuela Superior Politècnica de Chimborazo, Ecuador, to evaluate the influence of litter size of guinea pigs (Cavia porcellus) on their development and to establish the economic profitability of the production system. Forty-eight animals were used, distributed into litters of two, three, and four rodents per litter, with a balanced diet and green fresh alfalfa for the weaning, growth, and fattening stage, the rodents and litters were randomly selected, applying the statistical model completely randomly and evaluating different variables across 120 days. The litters of three guinea pigs obtained the best productive responses and economic profitability. With respect to sex, the males presented better productive behavior, greater economic increase, and less cost, evidencing that mixed feeding influences the number of guinea pigs per birth in terms of growth and development. The results serve to improve guinea pig meat production for the rural population.Ball burnishing treatment, using the Ecoroll system, of steel samples was conducted. In the experiment, the burnishing pressure was changed. After the treatments, measurements of the surface topographies of disc samples were conducted using a white light interferometer Talysurf CCI Lite. Tribological tests were carried out in a ball-on-disc configuration. After these tests, measurements of surface topographies of disc samples were repeated. Worn surfaces were also analyzed with a scanning electron microscope. It was found that as the result of burnishing, surface topography height of tested samples decreased. Ball burnishing led to a reduction in the frictional resistance. The highest friction reduction of more than 40% was achieved with a burnishing pressure of 20 MPa.The low efficiency of currently-used anti-cancer therapies poses a serious challenge, especially in the case of malignant melanoma, a cancer characterized by elevated invasiveness and relatively high mortality rate. The role of the tumor microenvironment in the progression of melanoma and its acquisition of resistance to treatment seems to be the main focus of recent studies. One of the factors that, in normal conditions, aids the organism in its fight against the cancer and, following the malignant transformation, adapts to facilitate the development of the tumor is the immune system. A variety of cell types, i.e., T and B lymphocytes, macrophages, and dendritic and natural killer cells, as well as neutrophils, support the growth and invasiveness of melanoma cells, utilizing a plethora of mechanisms, including secretion of pro-inflammatory molecules, induction of inhibitory receptors expression, or depletion of essential nutrients. This review provides a comprehensive summary of the processes regulated by tumor-associated cells that promote the immune escape of melanoma cells. The described mechanisms offer potential new targets for anti-cancer treatment and should be further studied to improve currently-employed therapies.Epithelial ovarian cancer (EOC) remains a lethal disease for the majority of women diagnosed with it worldwide. For the majority of patients, diagnosis occurs late, in the advanced setting. Disease-induced as well as treatment-related adverse events can negatively impact quality of life (QoL). Research to date has captured these data through use of patient-related outcomes (PROs) and, increasingly, has become an area of increased attention and focus in clinical trial reporting. QoL/PRO measurements in EOC clinical trials at different transition points in a patient's journey are increasingly being recognized by patients, clinicians and regulatory agencies as the key determinants of treatment benefit. Various context-specific PROs and PRO endpoints have been described for clinical trials in EOC. Standardized approaches and checklists for incorporating PRO endpoints in clinical trials have been proposed. In a real-world clinical practice setting, PRO/QoL measures, which are meaningful, valid, reliable, feasible and acceptable to patients and clinicians, need to be implemented and used. These may assist by serving as screening tools; helping with the identification of patient preferences to aid in decision making; improving patient-provider communication; facilitating shared decision making. Importantly, they may also improve quality of care through an increasingly patient-centered approach. Potential areas of future research include assessment of anxiety, depression and other mental health issues. In good prognostic groups, such as maintenance clinical trials, following patients beyond progression will capture possible downstream effects related to delaying the psychological trauma of relapse, symptoms due to disease progression and side-effects of subsequent chemotherapy. Identifying PRO endpoints in next-generation-targeted therapies (including immunotherapies) also warrants investigation.Heterodichogamous reproduction in plants involves two flowering morphs, reciprocal in their timing of male and female sexual functions. The degree of synchrony in floral sex phase, within and between individuals of each morph, determines the flowers' potential fertilization partners. Complete within-morph synchrony enables across-morph mating alone, whereas unsynchronized floral sex phases may allow fertilization within a plant individual (geitonogamy) or within a morph. We documented the disruption of flowering synchrony in the heterodichogamous Ziziphus spina-christi towards the end of its seven-month flowering season. This desert tree has self-incompatible, protandrous, short-lived (2-day) flowers that open before dawn ('Early' morph) or around noon ('Late' morph). We counted flowers in the male and female phase on flowering branches that were sampled monthly during the 2016-2018 flowering seasons. In 2018, we also tagged flowers and followed their sex-phase distributions over two days at the start, middle, and end of the season. The switch to the female phase was delayed at the end-season (November-December), and 74% of the flowers did not develop beyond their male phase. Differences in male-phase duration resulted in asynchrony among flowers within each tree and among trees of both flowering morphs. Consequently, fertilization between trees of the same morph becomes potentially possible during the end-season. In controlled hand-pollination assays, some within-morph fertilizations set fruit. The end-season breakdown of synchronous flowering generates variability within morphs and populations. We suggest that this variability may potentially enable new mating combinations in a population and enhance its genetic diversity.Bacillus phage φ29 and its relatives have been considered as one of the most important model organisms for DNA replication, transcription, morphogenesis, DNA packaging studies, and nanotechnology applications. Here, we isolated and characterized a new member of the φ29-like phage, named Bacillus cereus phage vB_BceP-DLc1. This phage, with a unique inserted gene cluster, has the largest genome among known φ29-like phages. DLc1 can use the surface carbohydrate structures of the host cell as receptors and only infects the most related B. cereus strains, showing high host-specificity. The adsorption rate constant and life cycle of DLc1 under experimental conditions were also determined. Not only stable under temperatures below 55 °C and pH range from 5 to 11, the new phage also showed tolerance to high concentrations of NaCl, 75% ethanol, chloroform, and mechanical vortex, which is preferable for practical use in the food and pharmaceutical industries.Liver cirrhosis (LC), contributing to more than 1 million of deaths annually, is a major healthcare concern worldwide. Hepatitis B virus (HBV) is a major LC etiological factor, and 15% of patients with chronic HBV infection (CHB) develop LC within 5 years. Recently, novel host genetic determinants were shown to influence HBV lifecycle and CHB course. DNA repair enzymes can affect dynamics of liver damage and are involved in HBV covalently closed circular DNA (cccDNA) formation, an essential step for viral replication. This study aimed to evaluate the possible role of genes representing key DNA-repair pathways in HBV-induced liver damage. MALDI-TOF MS genotyping platform was applied to evaluate variations within XRCC1, XRCC4, ERCC2, ERCC5, RAD52, Mre11, and NBN genes. Apart from older age (p less then 0.001), female sex (p = 0.021), portal hypertension (p less then 0.001), thrombocytopenia (p less then 0.001), high HBV DNA (p = 0.001), and high aspartate aminotransferase (AST) (p less then 0.001), we found that G allele at rs238406 (ERCC2, p = 0.025), T allele at rs25487 (XRCC1, p = 0.012), rs13181 GG genotype (ERCC2, p = 0.034), and C allele at rs2735383 (NBN, p = 0.042) were also LC risk factors. The multivariate logistic regression model showed that rs25487 CC (p = 0.005) and rs238406 TT (p = 0.027) were independently associated with lower risk of LC. This study provides evidence for the impact of functional and potentially functional variations in key DNA-repair genes XRCC1 and ERCC2 in HBV-induced liver damage in a Caucasian population.Mycoplasma synoviae (MS) and Mycoplasma gallisepticum (MG) strains were isolated at 39.5 °C to rule out temperature-sensitive strains (ts+) and identified using random amplification of polymorphic DNA. selleck compound Then, their minimum inhibitory concentrations (MIC100) were calculated in isolated strains from broiler breeders and laying hens vaccinated with ts+ MS-H and ts+ MG TS-11 vaccines in Mexico. We sampled 631 lots of hens. A total of 28 of the 123 MS isolates and 12 of the 23 MG isolates were analyzed using random amplification of polymorphic DNA, of which 24 and 3 matched the DNA banding patterns of the MS-H and MG-F strains, respectively. The isolated MS and MG strains were sensitive to tiamulin and tylosin and showed intermediate sensitivity or resistance to lincomycin, florfenicol, erythromycin, enrofloxacin, and curcumin. Although both the MS and MG strains were sensitive to the same antibiotics (MIC100 lower than 1 mg mL-1), the MG strains were 5 to 10 times more sensitive than the MS strains. This work describes the use of an amperometric cytosensor (i.e., a whole cell-based amperometric biosensoristic device) for studying mitochondrial interferences of BPA and BPB (5-100 µg/mL) in the yeast Saccharomyces cerevisiae model following long-term (24 h) exposure (acute toxicity). Percentage interference (%ρ) on yeast aerobic mitochondrial catabolism was calculated after comparison of aerobic respiration of exposed and control S. cerevisiae cell suspensions. Results suggested the hypothesis of a dose-dependent co-action of two mechanisms, namely uncoupling of oxidative phosphorylation and oxidative stress. These mechanisms respectively matched with opposite effects of hyperstimulation and inhibition of cellular respiration. While uncoupling of oxidative phosphorylation and oxidative stress have been previously described as separate effects from in vitro BPA exposure using other biochemical endpoints and biological systems, effects of BPB on cellular aerobic respiration are here reported for the first time.
