These findings illuminate a molecular mechanism linking the PKD2-mediated PERK-eIF2ɑ-ATF4 pathway and amino acid metabolic reprogramming in ADPKD.We explored the effect of baicalein on the ferroptosis of melanocytes in vitiligo. Melanocytes were treated with single RSL3 or combined RSL3 with FAC for 24 h and the effect of baicalein on RSL3 toxicity was further evaluated. Cell viability was examined by CCK8 assay. The mitochondrial membrane potential and the level of iron ion were measured by assay kit. Intracellular and lipid ROS production was detected by flow cytometry. The results indicated that RSL3 induced cell death, mitochondrial dysfunction, ROS production, and iron ion accumulation in melanocytes, which was aggravated by the addition of FAC. The damage induced by RSL3 was significantly relieved by baicalein treatment. Besides, baicalein up-regulated GPX4 and reduced TFR1 level in melanocytes treated with RSL3+FAC. Baicalein protected melanocytes against ferroptosis through up-regulating GPX4. Although dopaminergic system in amygdala-medial prefrontal cortex (mPFC) circuit is hyper-responsive to stress-induced anxiety, the mechanisms that control anxiety still remains unanswered. Here, the acute restraint stress model(ARS) was established to develop anxiety-like behavior. The D2-dopamine receptor (D2R) availability in amygdala and mPFC was assessed using [18F]-fallypride positron emission tomography(PET) and immunohistochemical assay. We revealed that ARS paradigm was successfully established, as evidenced by elevated plus-maze test(EPM) and increased corticosterone release. Moreover, PET imaging displayed elevated D2R availability in the amygdala and mPFC in ARS as compared to that in the naives. PET imaging combined with immunohistochemical assay confirmed that amygdaloid D2R was significantly implicated in stress-induced anxiety. Our findings delivered valuable insights into neuromechanism of amygdaloid D2R underlying stress-induced anxiety and might have important implications for developing therapeutics for anxiety by targeting amygdaloid D2R.

The present work focuses on assessment of Parkinson's disease (PD), including both PD subtype identification (unsupervised task) and prediction (supervised task). We specifically investigate optimal feature selection and machine learning algorithms for these tasks.

We selected 885 PD subjects as derived from longitudinal datasets (years 0-4; Parkinson's Progressive Marker Initiative), and investigated 981 features including motor, non-motor, and imaging features (SPECT-based radiomics features extracted using our standardized SERA software). Two different hybrid machine learning systems (HMLS) were constructed and applied to the data in order to select optimal combinations in both tasks (i) identification of subtypes in PD (unsupervised-clustering), and (ii) prediction of these subtypes in year 4 (supervised-classification). From the original data based on years 0 (baseline) and 1, we created new datasets as inputs to the prediction task (i,ii) CSD0 and CSD01 cross-sectional datasets from year 0 only and obust identification of subtypes as well as appropriate prediction of these subtypes in PD patients. Moreover, use of timeless dataset, beyond cross-sectional datasets, enabled predictive accuracies over 90%. Overall, we showed that radiomics features extracted from SPECT images are important in clustering as well as prediction of PD subtypes.

In this context, the present study aimed to characterize the molecular basis of neonicotinoid resistance in the mosquito Aedes aegypti. Resistance mechanisms were studied by combining transcriptomic and genomic data obtained from a laboratory strain selected at the larval stage after 30 generations of exposure to imidacloprid (Imida-R line). After thirty generations of selection, larvae of the Imida-R line showed an 8-fold increased resistance to imidacloprid and a significant cross-tolerance to the pyrethroids permethrin and deltamethrin. Cross-resistance to pyrethroids was only observed in adults when larvae were previously exposed to imidacloprid suggesting a low but inducible expression of resistance alleles at the adult stage. Resistance of the Imida-R line was associated with a slower larval development time in females. Multiple detoxification enzymes were over-transcribed in larvae in association with resistance including the P450s CYP6BB2, CYP9M9 and CYP6M11 previously associated with pyrethroid resistance.
