Our results suggest that framing matters when communicating to the public.Plasticity and evolution are two processes allowing populations to respond to environmental changes, but how both are related and impact each other remains controversial. We studied plastic and evolutionary responses in gene expression of Tribolium castaneum after exposure of the beetles to new environments that differed from ancestral conditions in temperature, humidity or both. Using experimental evolution with 10 replicated lines per condition, we were able to demonstrate adaptation after 20 generations. We measured whole-transcriptome gene expression with RNA-sequencing to infer evolutionary and plastic changes. selleck chemical We found more evidence for changes in mean expression (shift in the intercept of reaction norms) in adapted lines than for changes in plasticity (shifts in slopes). Plasticity was mainly preserved in selected lines and was responsible for a large part of the phenotypic divergence in expression between ancestral and new conditions. However, we found that genes with the largest evolutionary changes in expression also evolved reduced plasticity and often showed expression levels closer to the ancestral stage. Results obtained in the three different conditions were similar, suggesting that restoration of ancestral expression levels during adaptation is a general evolutionary pattern. With a larger sample in the most stressful condition, we were able to detect a positive correlation between the proportion of genes with reversion of the ancestral plastic response and mean fitness per selection line.Vitamin U (Vit U) is a novel free-radical scavenger. The protective effect of Vit U on valproic acid (VPA)-induced lung damage was examined. Rats were divided into four groups control rats; rats given Vit U (50 mg/kg/d, by gavage) for 15 days; rats treated with VPA (500 mg/kg/d, intraperitoneally) for 15 days; and rats were given VPA + Vit U (in same dose and time). On the 16th day of the experiment, the lungs were collected from rats. Lung structure, pulmonary oxidant/antioxidant parameters and Nrf2, α-SMA, and collagen-1 were evaluated by microscopic and biochemical analysis. Additionally, it was determined the interactions of Vit U with Nrf2 and Keap1 by in silico analysis. VPA administration increased lipid peroxidation and the activity of lactate dehydrogenase and myeloperoxidase. However, it decreased the glutathione level, and the activities of glutathione peroxidase, glutathione-S-transferase, catalase, and superoxide dismutase. VPA-mediated oxidative stress prompted structural distortion and fibrotic alterations in the lung. Vit U supplementation reversed structural and biochemical alterations, induced antioxidant system through Nrf2 activation, and attenuated fibrosis by reducing collagen expression in VPA-administered rats. However, Vit U pretreatment was unable to reduce α-SMA levels in the lung of VPA-treated rats. Molecular docking analysis showed the binding of Vit U to ETGE motif leads to dissociation of Nrf2 from the Nrf2/Keap1 complex and its transfer to nuclei. In conclusion, Vit U attenuated VPA-induced tissue damage by restoring antioxidative systems through amelioration of Nrf2 activity in the lung under oxidative stress.

This study was to evaluate the potential of long non-coding RNA maternally expressed gene 3 (lncRNA MEG3) in predicting acute respiratory distress syndrome (ARDS) risk and its correlation with prognosis in sepsis patients.

The plasma samples were obtained from 112 sepsis patients within 24hours after admission and 100 healthy controls (HCs) at enrollment. The lncRNA MEG3 expression in plasma samples was determined by RT-qPCR. In sepsis patients, ARDS occurrence was assessed based on Berlin definition of ARDS and 28-day mortality risk was evaluated.

LncRNA MEG3 expression was increased in sepsis patients compared with HCs. During 28-day duration, 30 sepsis patients occurred ARDS and 82 sepsis patients did not occur ARDS. LncRNA MEG3 expression was elevated in ARDS sepsis patients compared with non-ARDS sepsis patients, then the following receiver-operating characteristic (ROC) curve analysis disclosed that lncRNA MEG3 predicted ARDS risk (area under the curve (AUC)=0.775), which was further validated as an independent risk factor by multivariate logistic regression. Furthermore, lncRNA MEG3 was positively correlated with chronic obstructive pulmonary disease, respiratory infection, acute physiology and chronic health evaluation II score, sequential organ failure assessment score, white blood cell, and C-reactive protein, while negatively correlated with albumin in sepsis patients. Additionally, lncRNA MEG3 was elevated in 28-day deaths compared with 28-day survivors, and it predicted 28-day mortality risk in sepsis patients (AUC=0.708) by ROC curve analysis.

LncRNA MEG3 might represent as a valuable biomarker for individualizing prevention strategies against ARDS and improving prognosis in sepsis.

Treatment outcome varies with diagnosis, but little is known about the influence of sex, psychosocial adjustment and aftercare. To assess these variables, we interviewed male and female patients discharged from three psychiatric security hospitals in Germany in the years 2010-2017. Participants were interviewed at discharge (n = 609) and 1 year later (n = 366) about reoffending, readmissions, substance use and psychosocial adjustment. Among patients with substance use disorder (SUD), 14% reoffended, 20% were re-hospitalized and 60% maintained abstinence. Among patients with severe mental disorder, 5% reoffended and 13% were re-hospitalized. Significant sex differences were found in offenders with SUD. The results suggest that sociodemographic and disorder-related risk factors are associated with treatment success and that female patients with SUD might need a specific treatment approach.
