Using two 28-day microcosm studies involving two different photoperiods (light/dark cycle of 16/8 vs 8/16), the present study assessed the effects of four increasing nickel (Ni) concentrations (0-6 μM) on two natural biofilm communities collected at different seasons (summer and winter). The two communities were characterized by different structural profiles and showed significant differences in Ni accumulated content for each treatment. For instance, the biofilm metal content was four times higher in the case of summer biofilm at the highest Ni treatment and after 28 days of exposure. Biomarkers examined targeted both heterotrophic and autotrophic organisms. For heterotrophs, the β-glucosidase and β-glucosaminidase showed no marked effects of Ni exposure and were globally similar between the two communities suggesting low toxicity. However, the photosynthetic yield confirmed the toxicity of Ni on autotrophs with maximum inhibition of 81 ± 7% and 60 ± 1% respectively for the summer and winter biofilms. Furthermore, biofilms previously exposed to the highest long-term Ni concentration ([Ni2+] = 6 μM) revealed no acute effects in subsequent toxicity based on the PSII yield, suggesting a tolerance acquisition by the phototrophic community. Taken together, the results suggest that the biofilm response to Ni exposure was dependent of the function considered and that descriptors such as biofilm metal content could be seasonally dependent, information of great importance in a context of biomonitoring.Microplastics (MPs) provide a stable and protective habitat for diverse wastewater bacteria, including pathogenic and antibiotic-resistant species. Therefore, MPs may potentially transport these bacteria through wastewater treatment steps to the environment and far distances. This study investigated bacterial communities of MP-associated bacteria from different stages of municipal wastewater treatment processes to evaluate the potential negative effect of these biofilms on the environment. The results showed a high diversity of bacteria that were strongly attached to MPs. After all treatment steps, the core bacterial groups remained attached to MPs and escaped from the wastewater treatment plant with effluent water. Several pathogenic bacteria were identified in MP samples from all treatment steps, and most of them were found in effluent water. These data provide new insights into the possible impacts of wastewater-derived MPs on the environment. MP-associated biofilms were proved to be important sources of pathogens and antibiotic-resistant genes in natural waters.Ecotoxicological effects of photolytic degradation mixtures of the two brominated flame retardants PolymericFR and Tetrabromobisphenol A-bis (2,3-dibrom-2-methyl-propyl) Ether (TBBPA-BDBMPE) have been studied in vitro and in vivo. Both substances were experimentally degraded separately by exposure to artificial UV-light and the resulting degradation mixtures from different time points during the UV-exposure were applied in ecotoxicological tests. The in vitro investigation showed no effects of the degraded flame retardants on the estrogenic and androgenic receptors via the CALUX (chemically activated luciferase gene expression) assay. In this study, we created a systematic dataset of cooking emission factors (CEFs) and applied it with a new data source, cooking-related point of interest (POI) data, to build up highly spatial resolved cooking emission inventories from the city scale. Averaged CEFs of six particulate and gaseous species (PM, OC, EC, NMHC, OVOCs, VOCs) were 5.92 ± 6.28, 4.10 ± 5.50, 0.05 ± 0.05, 22.54 ± 20.48, 1.56 ± 1.44, and 7.94 ± 6.27 g/h normalized in every cook stove, respectively. A three-field CEF index containing activity and emission factor species was created to identify and further build a connection with cooking-related POI data. A total of 95,034 cooking point sources were extracted from Beijing, as a study city. In downtown areas, four POI types were overlapped in the central part of the city and radiated into eight distinct directions from south to north. Estimated PM/VOC emissions caused by cooking activities in Beijing were 4.81/9.85 t per day. A 3D emission map showed an extremely unbalanced emission density in the Beijing region. Emission hotspots were seen in Central Business District (CBD), Sanlitun, and Wangjing in Chaoyang District and Willow and Zhongguancun in Haidian District. Quantitative real-time reverse transcription-polymerase chain reaction was performed to assess miR-328a-3p levels 0, 1, 4, 7 and 14 days post-sciatic nerve damage. The results showed miR-328a-3p was upregulated after nerve damage. CCK8 and EdU assays revealed elevated miR-328a-3p amounts suppressed Schwann cell viability and proliferation, respectively. Next, the migratory potential of cells was assessed by the Transwell chamber and wound healing assays. We found elevated miR-328a-3p amounts also suppressed Schwann cell migration. Conversely, low miR-328a-3p amounts promoted Schwann cell migration. The possible miR-328a-3p targets were predicted by bioinformatics. The 15 target genes retrieved provided insights into miR-328a-3p's effects on Schwann cells and expanded the understanding of miR-328a-3p's biological functions in the peripheral nervous system. In the present study, we investigated chronic sickness responses in TRPM8 knockout (KO) mice during lipopolysaccharide (LPS)-induced sepsis. The intraperitoneal administration of 5 mg/kg LPS generated longer-lasting hypothermia in TRPM8 KO mice than in wild-type (WT) mice. TRPM8 KO mice also exhibited longer-lasting declines in locomotor activity, body weight, and food and water intakes than WT mice upon LPS administration. In addition, LPS-induced decreases in the numbers of leucocytes and lymphocytes that persisted for a longer time in TRPM8 KO mice than in WT mice. However, the function of IRFs in invertebrates is unclear. Here, the full-length cDNA of an IRF-encoding gene (CfIRF1) in the Zhikong scallop (Chlamys farreri) comprising 2007 bp with an open reading frame of 1053 bp that encoded 350 amino acids was characterized, and its immune function was studied. The CfIRF1 protein contained a typical IRF domain at its N-terminus. CfIRF1 was clustered with other proteins of the IRF1 subfamily, implying that they were closely related. CfIRF1 mRNA transcripts could be detected in all tested scallop tissues, with the highest expression observed in the gills and hepatopancreas. CfIRF1 expression was significantly induced by the polyinosinic-polycytidylic acid and acute viral necrosis virus challenge. CfIRF1 could directly interact with myeloid differentiation primary response protein 88 (MyD88), the key adaptor molecule of the toll-like receptor signaling pathway. CfIRF1 did not interact with scallop IKK1 (IKKα/β family protein), IKK2, IKK3 (IKKε/TBK1 family protein), or with other IRF family proteins (IRF2 or IRF3). However, β-aryl ethers are mostly cleaved to form carbon-carbon bonds under the acidic condition. In this work, lignin isolated with methanolic hydrochloric acid (MHL) was compared with conventional milled wood lignin (MWL). The methanol incorporated into lignin as α-OCH3 to prevent the cleavage of β-aryl ethers. As a result, the yield of MHL was over 12 times that of MWL. The MHL appeared a regular granular structure with an average diameter of 100 nm. Analysis of 13C-1H HSQC NMR spectra showed that MHL contained 42.6 % β-O-4 linkages whereas 35.9 % for the MWL, as well as 6.5 % β-5 and 3.2 % β-β, which was less than MWL. Moreover, 13C NMR spectroscopy confirmed that MHL was mainly isolated by the cleavage of the associated carbohydrates rather than the degradation of β-aryl ether bonds.
