A full factorial design (ascorbic acid/l-cysteine inhibitors, temperature, and time as factors) study was conducted to enhance inhibition of polyphenol oxidase (PPO) activity without decreasing cocoa polyphenol concentrations. The data obtained were modelled through a new equation, represented by Γ, which correlates both high polyphenol content with reduced specific PPO activity. At optimized values (70 mM inhibitory solution at 96 °C for 6.4 min, Γ = 11.6), 93.3% PPO inhibition and total polyphenol of 94.9 mg GAE/g were obtained. The second objective was to determine whether lateral compartment gapping affects clinical outcomes. Methods A prospective randomized clinical study was carried out that included 18 patients in Group 1 (FHO) and 18 in Group 2 (TFJD). Varus-stress radiographs of all the patients with both knees at full extension and at 30 ° of flexion were studied pre-operatively and 12 months post-operatively. Lateral compartment gapping was measured in millimeters. The Knee Society Score (KSS) was used to assess clinical stability. Results The difference between the pre- and post-operative measurements relative to gapping in the lateral knee compartment at 0 ° of knee flexion was 1.3 mm (SD 1.8) in Group 1 and 4.5 mm (SD 2.4) in Group 2 (p = 0.006). We demonstrate that multiple narrow-band LEDs have inferior color homogeneity compared to broadband light sources. Therefore, we consider the broadband option based on phosphor material excited by LEDs. A first prototype was realized and its details are discussed. It was tested within a remote-PPG monitoring scenario in darkness and the full system demonstrates robust pulse-rate measurement. Given its accuracy in pulse rate extraction, the proposed illumination principle is considered a valuable asset for large-scale NIR-PPG applications as it enables multi-wavelength processing, lightweight set-ups with relatively low-power infrared light sources.
