Clostridium ljungdahlii (C. ljungdahlii, CLJU) is natively endowed producing acetic acid, 2,3-butandiol, and ethanol consuming gas mixtures of CO2, CO, and H2 (syngas). Here, we present the syngas-based isobutanol formation using C. ljungdahlii harboring the recombinant amplification of the "Ehrlich" pathway that converts intracellular KIV to isobutanol. Autotrophic isobutanol production was studied analyzing two different strains in 3-L gassed and stirred bioreactors. Physiological characterization was thoroughly applied together with metabolic profiling and flux balance analysis. Thereof, KIV and pyruvate supply were identified as key "bottlenecking" precursors limiting preliminary isobutanol formation in CLJU[KAIA] to 0.02 g L-1. Additional blocking of valine synthesis in CLJU[KAIA]ilvE increased isobutanol production by factor 6.5 finally reaching 0.13 g L-1. Future metabolic engineering should focus on debottlenecking NADPH availability, whereas NADH supply is already equilibrated in the current generation of strains.Many single-use non-degradable plastics are a threat to life today, and several polyhydroxyalkanoates (PHAs) biopolymers have been developed in the bioplastic industry to place petrochemical-based plastics. One of such is the novel biomaterial poly(3-hydroxypropionate) [poly(3HP)] because of its biocompatibility, biodegradability, and high yield synthesis using engineered strains. To date, many bio-polymer-based functional composites have been developed to increase the value of raw microbial-biopolymers obtained from cheap sources. This review article broadly covers poly(3HP), a comprehensive summary of critical biosynthetic production pathways comparing the yields and titers achieved in different Microbial cell Factories. This article also provides extensive knowledge and highlights recent progress on biosensors' use to optimize poly(3HP) production, some bacteria host adopted for production, chemical and physical properties, life cycle assessment for poly(3HP) production using corn oil as carbon source, and some essential medical applications of poly(3HP).The results of visual prediction reflect the tendency and speed of visual development during a future period, based on which ophthalmologists and guardians can know the potential visual prognosis in advance, decide on an intervention plan, and contribute to visual development. In our study, we developed an intelligent system based on the features of optical coherence tomography images for long-term prediction of best corrected visual acuity (BCVA) 3 and 5 years in advance. Two hundred eyes of 132 patients were included. Six machine learning algorithms were applied. In the BCVA predictions, small errors within two lines of the visual chart were achieved. The mean absolute errors (MAEs) between the prediction results and ground truth were 0.1482-0.2117 logMAR for 3-year predictions and 0.1198-0.1845 logMAR for 5-year predictions; the root mean square errors (RMSEs) were 0.1916-0.2942 logMAR for 3-year predictions and 0.1692-0.2537 logMAR for 5-year predictions. This is the first study to predict post-therapeutic BCVAs in young children. This work establishes a reliable method to predict prognosis 5 years in advance. The application of our research contributes to the design of visual intervention plans and visual prognosis.Skin is a heterogeneous tissue that can undergo substantial structural and functional changes with age, disease, or following injury. Understanding how these changes impact the mechanical properties of skin requires three-dimensional (3D) quantification of the tissue microstructure and its kinematics. The goal of this study was to quantify these structure-function relationships via second harmonic generation (SHG) microscopy of mouse skin under tensile mechanical loading. Tissue deformation at the macro- and micro-scale was quantified, and a substantial decrease in tissue volume and a large Poisson's ratio was detected with stretch, indicating the skin differs substantially from the hyperelastic material models historically used to explain its behavior. selleck compound Additionally, the relative amount of measured strain did not significantly change between length scales, suggesting that the collagen fiber network is uniformly distributing applied strains. Analysis of undeformed collagen fiber organization and volume fraction revealed a length scale dependency for both metrics. 3D analysis of SHG volumes also showed that collagen fiber alignment increased in the direction of stretch, but fiber volume fraction did not change. Interestingly, 3D fiber kinematics was found to have a non-affine relationship with tissue deformation, and an affine transformation of the micro-scale fiber network overestimates the amount of fiber realignment. This observation has fundamental implications, as the formation of these assemblies is systematically associated with the insurgence of severe neurodegenerative diseases. Although the ability of proteins to form aggregates rich in cross-β structure has been highlighted by recent studies of structural biology, the determination of the underlying atomic models has required immense efforts and inventiveness. Interestingly, the progressive molecular and structural characterization of these assemblies has opened new perspectives in apparently unrelated fields. Indeed, the self-assembling through the cross-β structure has been exploited to generate innovative biomaterials endowed with promising mechanical and spectroscopic properties. Therefore, this structural motif has become the fil rouge connecting these diversified research areas. In the present review, we report a chronological recapitulation, also performing a survey of the structural content of the Protein Data Bank, of the milestones achieved over the years in the characterization of cross-β assemblies involved in the insurgence of neurodegenerative diseases. A particular emphasis is given to the very recent successful elucidation of amyloid-like aggregates characterized by remarkable molecular and structural complexities. We also review the state of the art of the structural characterization of cross-β based biomaterials by highlighting the benefits of the osmosis of information between these two research areas. Finally, we underline the new promising perspectives that recent successful characterizations of disease-related amyloid-like assemblies can open in the biomaterial field.
