RESULTS The obtained data demonstrate that the inhibition of PTP1B and LMPTP prevents apoptosis induced by palmitate and oleate in the HepG2 cell line. Moreover, mitochondrial dynamics were positively improved following inhibition of the enzyme, with concomitant oxidative stress reduction and ER stress abrogation. CONCLUSION In conclusion, PTP's inhibitory properties may be a promising therapeutic strategy for the treatment of FFA-induced lipotoxicity in the liver and ultimately in the management of the NAFLD condition. In this review, we discuss the growing literature demonstrating robust and pervasive associations between the microbiome and metabolome. We focus on the gut microbiome, which harbors the taxonomically most diverse and the largest collection of microorganisms in the human body. Methods for integrative analysis of these "omics" are under active investigation and we discuss the advances and challenges in the combined use of metabolomics and microbiome data. Findings from large consortia, including the Human Microbiome Project and Metagenomics of the Human Intestinal Tract (MetaHIT) and others demonstrate the impact of microbiome-metabolome interactions on human health. Mechanisms whereby the microbes residing in the human body interact with metabolites to impact disease risk are beginning to be elucidated, and discoveries in this area will likely be harnessed to develop preventive and treatment strategies for complex diseases. The properties of poly(vinyl alcohol) (PVA)-based composites recommend this material as a good candidate for the replacement of damaged cartilage, subchondral bone, meniscus, humeral joint and other orthopedic applications. The manufacturing process can be manipulated to generate the desired biomechanical properties. However, the main shortcomings of PVA hydrogels are related to poor strength and bioactivity. To overcome this situation, reinforcing elements are added to the PVA matrix. The aim of our work was to develop and characterize a novel composition based on PVA reinforced with Se-doped TiO2 nanoparticles and natural hydroxyapatite (HA), for possible orthopedic applications. The PVA/Se-doped TiO2 composites with and without HA were structurally investigated by FTIR and XRD, in order to confirm the incorporation of the inorganic phase in the polymeric structure, and by SEM and XRF, to evidence the ultrastructural details and dispersion of nanoparticles in the PVA matrix. Both the mechanical and structural properties of the composites demonstrated a synergic reinforcing effect of HA and Se-doped TiO2 nanoparticles. Moreover, the tailorable properties of the composites were proved by the viability and differentiation potential of the bone marrow mesenchymal stem cells (BMMSC) to osteogenic, chondrogenic and adipogenic lineages. The novel hybrid PVA composites show suitable structural, mechanical and biological features to be considered as a promising biomaterial for articular cartilage and subchondral bone repair. The delivery of drugs and bioactive molecules using pharmaceutical particulates and membranes are of great significance for various applications such as the treatment of secondary infections, cancer treatment, skin regeneration, orthopedic applications and others [...].Soil contamination by toxic heavy metals such as copper is a serious problem. In this study, the extracellular polymeric substance (EPS) extracted from Rahnella sp. LRP3 was found with the potential of immobilizing Cu-polluted in soil. The EPS could bond to Cu (II) through functional groups (polysaccharides, amide, proteins, and carboxyl groups), which further developed into the porous sphere with a diameter of 20 μm. Besides, EPS could induce the formation of Cu5(PO4)2(OH)4 crystal by the biomineralization process. Ertugliflozin manufacturer Finally, the EPS in the culture solution reduced 89.4 mg/kg of DTPA-Cu content by 78.99% in soil for 10 d under the condition of 25 °C via biomineralization. The results demonstrated that EPS produced by Rahnella sp. LRP3 will be a promising factor in the remediation of Cu contaminated soil. Microplastic pollution is a major global environmental problem in both aquatic and terrestrial environments. Pesticides are frequently applied to agricultural soil to reduce the effects of pests on crops, but may also affect the degradation of plastics. In this study, we generated microplastics from polyethylene (PE) film and biodegradable poly(butylene adipate-co-terephthalate) (PBAT) film and determined (1) the effect of prothioconazole on degradation of the microplastics, and (2) the adsorption and release characteristics of heavy metals (Cr, Cu, As, Pb, Ba, and Sn) by the microplastics during degradation process. Changes of surface functional groups and morphologies were measured by FTIR and SEM, while metal concentrations were determined by ICPMS. Prothioconazole was found to promote plastic degradation. PBAT degraded faster and adsorbed more heavy metals from the soil than PE. Whether the microplastics adsorb or release heavy metals depended on the metal and their concentrations. Prothioconazole inhibited the adsorption of Cr, As, Pb and Ba by microplastics, promoted the adsorption of Cu, and had no significant effect for Sn. These results can help to assess the ecological risk of microplastic pollution from plastic mulch when combined with heavy metals. Copper is known to interfere with fish olfaction. Although the chemosensory detection and olfactory toxicity of copper ions (Cu2+) has been heavily studied in fish, the olfactory-driven detection of copper nanoparticles (CuNPs)-a rapidly emerging contaminant to aquatic systems-remains largely unknown. This study aimed to investigate the olfactory response of rainbow trout to equitoxic concentrations of CuNPs or Cu2+ using electro-olfactography (EOG, a neurophysiological technique) and olfactory-mediated behavioural assay. In the first experiment, the concentration of contaminants known to impair olfaction by 20% over 24 h (EOG-based 24-h IC20s of 220 and 3.5 μg/L for CuNPs and Cu2+, respectively) were tested as olfactory stimuli using both neurophysiological and behavioural assays. In the second experiment, to determine whether the presence of CuNPs or Cu2+ can affect the ability of fish to perceive a social cue (taurocholic acid (TCA)), fish were acutely exposed to one form of Cu-contaminants (approximately 15 min).
