Electrons, however, are also efficient catalysts for conducting radical reactions, a property that is often overlooked by chemists. Special attention should be paid to electron catalysis when interpreting single-molecule conductance results for the simple reason that an unexpected reaction mediated or triggered by electrons might take place in the single-molecule junction. Here, we describe a counterintuitive structure-property relationship that molecules, both linear and cyclic, employing a saturated bipyridinium-ethane backbone, display a similar conductance signature when compared to junctions formed with molecules containing conjugated bipyridinium-ethene backbones. We describe an ethane-to-ethene transformation, which proceeds in the single-molecule junction by an electron-catalyzed dehydrogenation. Electrochemically based ensemble experiments and theoretical calculations have revealed that the electrons trigger the redox process, and the electric field promotes the dehydrogenation. This finding not only demonstrates the importance of electron catalysis when interpreting experimental results, but also charts a pathway to gaining more insight into the mechanism of electrocatalytic hydrogen production at the single-molecule level.Physisorption using gas or vapor probe molecules is the most common characterization technique for porous materials. The method provides textural information on the adsorbent as well as the affinity for a specific adsorbate, typically through equilibrium pressure measurements. Here, we demonstrate how low-field NMR can be used to measure full adsorption isotherms, and how by selectively measuring 1H spins of the adsorbed probe molecules, rather than those in the vapor phase, this "NMR-relaxorption" technique provides insights about local dynamics beyond what can be learned from physisorption alone. The potential of this double-barreled approach was illustrated for a set of microporous metal-organic frameworks (MOFs). For methanol adsorption in ZIF-8, the method identifies multiple guest molecules populations assigned to MeOH clusters in the pore center, MeOH bound at cage windows and to MeOH adsorption on defect sites. For UiO-66(Zr), the sequential pore filling is demonstrated and accurate pore topologies are directly obtained, and for MIL-53(Al), structural phase transitions are accurately detected and linked with two populations of dimeric chemical species localized to specific positions in the framework.Stem canker of Zanthoxylum bungeanum is a devastating disease that seriously affects the plantation and industrial development of Z. bungeanum due to a lack of effective control measures. The objective of this study was to screen out resistant Z. bungeanum varieties and further explore their resistance mechanisms against stem canker. Results showed that the most resistant and susceptible varieties were, respectively, Doujiao (DJ) and Fengxian Dahongpao (FD). Combining transcriptomic and metabolomic analyses, we found that the genes and metabolites associated with the phenylpropanoid metabolism, especially flavonoid biosynthesis, were highly significantly enriched in DJ following pathogen infection compared with that in FD, which indicated that the flavonoid metabolism may positively dominate the resistance of Z. bungeanum. This finding was further confirmed by quantitative real-time polymerase chain reaction analysis, through which higher expression levels of core genes involved in flavonoid metabolism in resistant variety were observed. Moreover, by analyzing the differences in the flavonoid content in the stems of resistant and susceptible varieties and the antifungal activities of flavonoids extracted from Z. bungeanum stems, the conclusion that flavonoid metabolism positively regulates the resistance of Z. bungeanum was further supported. Our results not only aid in better understanding the resistance mechanisms of Z. bungeanum against stem canker but also promote the breeding and utilization of resistant varieties.Different proteoform products of the same gene can exhibit differing associations with health and disease, and their patterns of modifications may offer more precise markers of phenotypic differences between individuals. However, currently employed protein-biomarker discovery and quantification tools, such as bottom-up proteomics and ELISAs, are mostly proteoform-unaware. Moreover, the current throughput for proteoform-level analyses by liquid chromatography mass spectrometry (LCMS) for quantitative top-down proteomics is incompatible with population-level biomarker surveys requiring robust, faster proteoform analysis. To this end, we developed immunoprecipitation coupled to SampleStream mass spectrometry (IP-SampleStream-MS) as a high-throughput, automated technique for the targeted quantification of proteoforms. We applied IP-SampleStream-MS to serum samples of 25 individuals to assess the proteoform abundances of apolipoproteins A-I (ApoA-I) and C-III (ApoC-III). The results for ApoA-I were compared to those of LCMS for these individuals, with IP-SampleStream-MS showing a >7-fold higher throughput with >50% better analytical variation. Proteoform abundances measured by IP-SampleStream-MS correlated strongly to LCMS-based values (R2 = 0.6-0.9) and produced convergent proteoform-to-phenotype associations, namely, the abundance of canonical ApoA-I was associated with lower HDL-C (R = 0.5) and glycated ApoA-I with higher fasting glucose (R = 0.6). selleck kinase inhibitor We also observed proteoform-to-phenotype associations for ApoC-III, 22 glycoproteoforms of which were characterized in this study.
