This analysis was conducted on safety data pooled from phase 3 studies using ertugliflozin 5 mg or 15 mg versus placebo or an active comparator. METHODS The placebo pool (n = 1544) comprised data from three similarly designed 26-week placebo-controlled studies. The broad pool (n = 4849) comprised these three placebo-controlled studies plus four placebo- or active-controlled studies with treatment durations of up to 104 weeks. RESULTS In the placebo pool, there were no notable differences across groups in the incidence of adverse events (AEs), serious AEs, or AEs resulting in discontinuation from study medication, while associations were observed with genital mycotic infection in both females (3.0%, 9.1%, and 12.2% in the placebo, ertugliflozin 5 mg, and ertugliflozin 15 mg groups, respectively) and males (0.4%, 3.7%, 4.2%), thirst (0.2%, 1.3%, 1.0%), andor other SGLT2 inhibitors. A synthetic microbial consortium can enable the simultaneous utilization of sugars such as glucose and xylose to produce biochemicals, where each consortium member converts one sugar into the target product. In this study, woody biomass was used to generate glucose and xylose after pretreatment with 20% (w/v) sulfuric acid and 60-min reaction time. We compared several strategies for detoxification with charcoal and sodium borohydride treatments to improve the fermentability of this hydrolysate in a defined medium for the production of the growth-associated product pyruvate. In shake flask culture, the highest pyruvate yield on xylose of 0.8 g/g was found using pH 6 charcoal-treated hydrolysate. In bioreactor studies, a consortium of two engineered E. coli strains converted the mixture of glucose and xylose in batch studies to 12.8 ± 2.7 g/L pyruvate in 13 h. These results demonstrate that lignocellulosic biomass as the sole carbon source can be used to produce growth-related products after employing suitable detoxification strategies.This study examined whether cognitive processes in preschool, conceptualized as a unitary construct of executive control (EC) as well as foundational cognitive abilities (FCA), predict both maladaptive and adaptive functioning in middle childhood and mediate associations between early childhood socio-familial stress and those functional outcomes. Performance-based, multidimensional, and age-appropriate measures of EC and FCA were collected in a laboratory setting from 313 preschool-age children at age 5, along with questionnaire data from children and their parents on three dimensions of early socio-familial stress and parent smoking. Parent, teacher, and child self-report data on 285 of these children were obtained when they were in grade 3 or 4. Middle childhood data were used to create indices of maladaptive and adaptive functioning. A bi-factor structural equation modeling analysis captured distinct dimensions of preschool EC and FCA and was used to test the hypothesized pathways. EC had a statistically significant negative association with later maladaptive functioning. FCA, but not EC, served as a mediator in links between each type of family stressor and both maladaptive and adaptive functioning in middle childhood. Results suggest that EC may play a role in predicting maladaptation, whereas early childhood FCA may operate as an intervening variable in pathways from early family stressors to subsequent maladaptation as well as adaptation. Findings point to the need to address FCA by reducing early family stressors. Early interventions that enhance cognitive abilities may help reduce maladaptive and promote adaptive functioning later in childhood, thereby potentially preventing, in turn, later behavioral problems.PURPOSE Although multiple studies have examined anastomotic leakage (AL) after low anterior resection (LAR), their definitions of AL varied, and few have studied late diagnosed AL after surgery. This study aimed to characterize late AL after anal sphincter saving surgery (SSS) for rectal cancer by examining clinical characteristics, risk factors, and management of patients with late AL compared with early AL. METHODS Data from January 2005 to December 2014 were collected from a total of 1903 consecutive patients who underwent anal sphincter saving surgery for rectal cancer and were retrospectively reviewed. Late AL was defined as AL diagnosed more than 30 days after surgery. Variables and risk factors associated with early and late diagnosed AL were analyzed by multivariate logistic regression. RESULTS Overall, early, and late rates of AL were 13.7%, 6.7%, and 7%, respectively. Receiving neoadjuvant chemoradiotherapy (nCRT) was a risk factor for developing late AL, but not early AL (OR, 3.032; 95% CI, 1.947-4.722; p  less then  0.001). The current study aims to determine if classes of youth psychopathology can be predicted by mTBI status. The current study analyzed a large sample of children and adolescents from the Project on Human Development in Chicago Neighborhoods (n = 2978) to examine trajectories of psychopathologies. Using data from the Achenbach Childhood Behavior Checklist, latent classes of internalizing and externalizing psychopathology were identified. Multinomial logistic regression models were used to examine if mTBI predicted class membership, while controlling for a number of variables associated with psychopathology. The results of the current study suggest that mTBI may be an important transdiagnostic risk factor associated with developmental patterns of internalizing and externalizing psychopathology.The parental bonding instrument (PBI) is a self-report questionnaire that was initially designed to retrospectively assess perceived parenting style during childhood in adult respondents. Recent studies have used the PBI to assess current perception of parenting in children. However, few studies have evaluated the psychometric properties of the PBI in children. This study examined the factor structure and reliability of the PBI in a sample of Canadian children aged 7-18 years (n = 257). Confirmatory factor analyses (CFA) were conducted separately for the mother and father form of the PBI and composite reliability was used to determine internal consistency of the PBI. A four-factor model (care, overprotection, autonomy, indifference) showed the best fit to the data. The PBI exhibited good internal consistency but poor convergent validity. Configural invariance was not found for the PBI between two age groups (7-12 and 13-18 years), however these findings should be interpreted with caution due to the small sample size in each age group. This study suggests the PBI may be a valid and reliable self-report measure of parental bonding in children but further research is needed.Children with internalizing and externalizing difficulties are at risk for long-term negative effects in adulthood and are impacted by several caregiver factors. Findings of the present study are consistent with previous studies that found direct associations between caregiver victimization history (e.g., physical and sexual abuse) and child behavior problems. Examination of potential mechanisms revealed that caregiver everyday stress related to relationships/responsibilities (RR) served as a mediator between caregiver victimization history and increased children's internalizing symptoms. Though there may be other pathways that contribute to this relation, there does seem to be clinical and policy utility of this knowledge, particularly for at-risk families that are faced with high levels of everyday RR stress. A retrospective cohort study included 273 patients. Genomic DNA was extracted from peripheral blood samples, and genotyping of selected SNP was performed by PCR-RFLP assays. Statistical analysis was then carried out to assess genotypic frequencies and genetic association in relation to breast cancer subtypes. SHEsis software was applied to IL-6 haplotypic structure analysis. The distribution of genotype frequencies of rs2517956, rs1800795 and rs2069837 showed no statistically difference between HER2-positive and HER2-negative breast cancer.
