Currently, the detailed mechanism is still unclear. Proteomics can offer a powerful set of tools for the direct high-throughput study and a key contribution to the understanding of protein functions. This approach can also allow us to compare the protein profiling of the cells in healthy and diseased states that can be used to identify proteins associated with disease development and progression. In the present study, we first established an autoimmune uveitis (EAU) rat model. On day 12 after immunization, we isolated the rat retinas from both normal and EAU animals to collect total proteins. Using tandem mass tag (TMT) peptide labeling coupled with LC-MS/MS quantitative proteomics technique, we identified the differentially expressed proteins in EAU rat retinas, performed bioinformatics analyses, validated the expression of the COX1, NADH1, C3, and C9 proteins, and determined the adenosine triphosphate (ATP) levels. The results indicated that there were 190 upregulated and 103 downregulated proteins in EAU rat retinas. Bioinformatics analysis revealed the differentially expressed proteins were mainly involved in acute inflammatory response, visual perception and eye photoreceptor cell differentiation that were mainly related to complement and coagulation cascades, phagosome, PI3K-Akt signaling, and metabolic pathways. In conclusion, based on the TMT-based quantitative proteomics technique, the differentially expressed proteins in EAU rat retinas were mainly associated with complement and coagulation cascades and metabolic pathways. Our findings will facilitate the understanding of the pathogenesis of uveitis and will be useful for subsequent studies.An accurate prognosis on the outcome of brain-injured patients with disorders of consciousness (DOC) remains a significant challenge, especially in the acute stage. In this study, we applied a multiple-technique approach to provide accurate predictions on functional outcome after 6 months in 15 acute DOC patients. Electrophysiological correlates of implicit cognitive processing of verbal stimuli and data-driven voxel-wise resting-state fMRI signals, such as the fractional amplitude of low-frequency fluctuations (fALFF), were employed. Event-related electrodermal activity, an index of autonomic activation, was recorded in response to emotional words and pseudo-words at baseline (T0). On the same day, patients also underwent a resting-state fMRI scan. Six months later (T1), patients were classified as outcome-negative and outcome-positive using a standard functional outcome scale. We found that only outcome-positive patients had an earlier, higher autonomic response for words compared to pseudo-words, a pattern similar to that of healthy awake controls. We propose that a residual autonomic response to cognitively salient stimuli, together with a preserved resting-state activity in the PCC, can provide a useful prognostic index in acute DOC.Thirty-four seco-3'R,4'R-disubstituted-2',2'-dimethyldihydropyrano[2,3-f]chromone (seco-DSP) derivatives were designed, synthesized and evaluated for chemo-reversal activity when combined with paclitaxel or vincristine in P-gp overexpressing A2780/T and KB-VIN drug-resistant cancer cell lines. Most of the compounds displayed moderate to significant MDR reversal activities. Compound 7e showed the most potent chemo-sensitization activity with more than 1471 reversal ratio at a concentration of 10 μM, which was higher than verapamil (VRP) (212-fold). Unexpectedly the newly synthesized compounds did not show chemosensitization activities in a non-P-gp overexpressing cisplatin resistant human ovarian cancer cell line (A2780/CDDP), implying that the MDR reversal effects might be associated with P-gp overexpression. Moreover, the compounds did not exhibit significant anti-proliferative activities against non-tumorigenic cell lines (HUVEC, HOSEC and T29) compared to VRP at the tested concentration and might be safer than VRP. In preliminary pharmacological mechanism studies, the compounds increased accumulation of DOX and promoted P-gp ATPase activity in A2780/T cell lines. Western blot analysis indicated they did not affect the expression level of P-gp in the tested MDR cell lines. Thus, further studies on these seco-DSP derivatives are merited with the goal of developing a desirable chemosensitizer drug candidate.

A constant voltage (DC voltage) is usually used in radiofrequency ablation (RFA) computer models to mimic the radiofrequency voltage. Our objective was to assess the thermal impact of replacing DC voltage by low-frequency AC voltage in RFA computer modeling.
