Cold-adapted alginate lyases have unique advantages for alginate oligosaccharide (AOS) preparation and brown seaweed processing. Robust and cold-adapted alginate lyases are urgently needed for industrial applications. VX-803 In this study, a cold-adapted alginate lyase-producing strain Vibrio sp. W2 was screened. Then, the gene ALYW201 was cloned from Vibrio sp. W2 and expressed in a food-grade host, Yarrowia lipolytica. The secreted Alyw201 showed the activity of 64.2 U/mL, with a molecular weight of approximate 38.0 kDa, and a specific activity of 876.4 U/mg. Alyw201 performed the highest activity at 30 °C, and more than 80% activity at 25-40 °C. Furthermore, more than 70% of the activity was obtained in a broad pH range of 5.0-10.0. Alyw201 was also NaCl-independent and salt-tolerant. The degraded product was that of the oligosaccharides of DP (Degree of polymerization) 2-6. Due to its robustness and its unique pH-stable property, Alyw201 can be an efficient tool for industrial production.This paper reports on high-accuracy simulation of a grating structure based fiber optic plasmonic sensor for salivary cortisol sensing. Gratings of SiO2 and SiC (one at a time) in combination with a thin Ag layer are considered to be in direct contact with analyte medium (solutions containing different concentrations of cortisol) considering that the groove regions are also filled with analyte. The optimization of Ag layer thickness is carried out to achieve maximum power loss (PL) corresponding to cortisol concentration variation. The variation of PL (in dB) spectra with the angle of incidence (α) is the sensing mechanism of the proposed scheme. Sensing performance is extensively analyzed in terms of sensitivity, limit-of-detection (LOD) and figure-of-merit (FOM) that incorporates both the sensitivity and the width of the corresponding PL curves. While the sensitivity and FOM values are significantly large, the results also reveal that in angular interrogation mode (AIM), an average LOD of 9.9 pg/mL and 9.8 pg/mL is obtained for SiO2 and SiC-based sensor designs, respectively. When the intensity interrogation method (IIM) in place of AIM is considered, an average LOD of 22.6 fg/mL and 68.17 fg/mL is obtained for SiO2 and SiC-based sensor designs, respectively. LOD (with IIM, in particular) is considerably better than the present-state-of-art related to cortisol monitoring. Pragmatic model for possible practical implementation of sensor scheme is also discussed. The involvement of optical fiber in the proposed sensor design makes it possible to implement it as a flexible sensor or for wearable solution for cortisol detection via sweat monitoring as well as for measuring cortisol level in aquaculture tanks where concentration levels are much lower than 10 ng/mL.Background and objectives This study was to clarify how the frequency of participation in a health class affected the reduction in sedentary behavior after two years, and whether decreases in sedentary behavior in elderly people who participated in a health class persisted two years after the end of an intervention. Materials and Methods This study was longitudinal, and the results of a previous study conducted in 2017 were added to the findings of a different year. The participants were elderly health class members at a community dwelling in Japan who participated between 2016 and 2018. Of the 86 participants that were enrolled, the data of 80 were collected in 2016. A year later, in a 2017 follow-up, the number of participants was 80, and two years later, in a 2018 follow-up, there were 71 participants. Results There was a significant difference with regards to the reduction of the sedentary behavior rate between two different groups (a health class participation rate of more than 75% and the other with less than 75%) two years later. However, no difference in sedentary behavior rate was found between the two groups (intervention group and control group) at a two-year follow-up, despite observing differences after one year. Conclusions Continuous participation in a health class may help reduce sedentary behavior. After two years, "Active Guide" brochures and documents may not help in reducing sedentary behavior, despite seeing improvements after one year. Persistent social participation may have a more lasting effect than one-off interventions in reducing the sedentary behavior ratio.Chronic wounds often occur in patients with diabetes mellitus due to the impairment of wound healing. This has negative consequences for both the patient and the medical system and considering the growing prevalence of diabetes, it will be a significant medical, social, and economic burden in the near future. Hence, the need for therapeutic alternatives to the current available treatments that, although various, do not guarantee a rapid and definite reparative process, appears necessary. We here analyzed current treatments for wound healing, but mainly focused the attention on few classes of drugs that are already in the market with different indications, but that have shown in preclinical and few clinical trials the potentiality to be used in the treatment of impaired wound healing. In particular, repurposing of the antiglycemic agents dipeptidylpeptidase 4 (DPP4) inhibitors and metformin, but also, statins and phenyotin have been analyzed. All show encouraging results in the treatment of chronic wounds, but additional, well designed studies are needed to allow these drugs access to the clinics in the therapy of impaired wound healing.Helicobacter pylori is considered the most prevalent bacterial pathogen in humans. The increasing antibiotic resistance evolved by this microorganism has raised alarm bells worldwide due to the significant reduction in the eradication rates of traditional standard therapies. A major challenge in this antibiotic resistance crisis is the identification of novel microbial targets whose inhibitors can overcome the currently circulating resistome. In the present study, we have validated the use of the essential response regulator ArsR as a novel and promising therapeutic target against H. pylori infections. A high-throughput screening of a repurposing chemical library using a fluorescence-based thermal shift assay identified several ArsR binders. At least four of these low-molecular weight compounds noticeably inhibited the DNA binding activity of ArsR and showed bactericidal effects against antibiotic-resistant strains of H. pylori.
