This study thus provides an enzyme loaded hydrogel platform with a pH-sensitive photothermal effect for mild-temperature-mediated combinational cancer therapy.Herein, we document a highly optimized synthesis of the Sec-to-Cys homologue of the human selenoprotein F (SelF) through a three-segment two-ligation semisynthesis strategy. Highlighted in this synthetic route are two one-pot manipulations, i.e. the first ligation followed by a desulfurization and the second ligation followed by the protein refolding. This way multi-milligrams of the folded synthetic protein was obtained, which set the stage for the synthesis of the natural selenoprotein. Moreover, the disulfide pairing mode of the SelF was elucidated through a combination of site-directed mutagenesis and LC-MS study. It provides not only a criterion to judge the viability of the synthetic protein, and more importantly, useful structural insights into the previously unresolved UGGT-binding domain of SelF.Photothermal therapy is a very promising treatment method in the field of cancer therapy. The photothermal nanomaterials in near-infrared region (NIR-I, 750-900 nm) attracts extensive attention in recent years because of the good biological penetration of NIR light. However, the penetration depth is still not enough for solid tumors due to high tissue scattering. The light in the second near-infrared region (NIR-II, 1000-1700 nm) allows deeper tissue penetration, higher upper limit of radiation and greater tissue tolerance than that in the NIR-I, and it shows greater application potential in photothermal conversion. This review summarizes the photothermal properties of Au nanomaterials, two-dimensional materials, metal oxide sulfides and polymers in the NIR-II and their application prospects in photothermal therapy. It will arouse the interest of scientists in the field of cancer treatment as well as nanomedicine.Viruses are natural supramolecular nanostructures that form spontaneously by molecular self-assembly of complex biomolecules. Peptide self-assembly is a versatile tool that allows mimicking viruses by creating their simplified versions through the design of functional, supramolecular materials with modularity, tunability, and responsiveness to chemical and physical stimuli. The main challenge in the design and fabrication of peptide materials is related to the precise control between the peptide sequence and its resulting supramolecular morphology. We provide an overview of existing sequence patterns employed for the development of spherical and fibrillar peptide assemblies that can act as viral mimetics, offering the opportunity to tackle the challenges of viral infections.Rapid glucose testing is very important in the care of diabetes. Monitoring of blood glucose is the most critical indicator of disease control in diabetic patients. The invention and popularity of electrochemical sensors have made glucose detection fast and inexpensive. The first generation of glucose sensors had limitations in terms of sensitivity and selectivity. In order to overcome these problems, scientists have used a range of new materials to produce new glucose electrochemical sensors with higher sensitivity, selectivity and lower cost. A variety of different electrochemical sensors including enzymatic electrochemical sensors and enzyme-free electrochemical sensors have been extensively investigated. We discussed the development process of electrochemical glucose sensors in this review. We focused on describing the benefits of carbon materials in nanomaterials, specially graphene for sensors. In addition, we discussed the limitations of the sensors and challenges in future research.Nanocarriers are rapidly growing in popularity in the field of drug delivery. The ability of nanocarriers to encapsulate and distribute poorly soluble drugs while minimising their undesired effects is significantly advantageous over traditional drug delivery. Nanocarriers can also be decorated with imaging moieties and targeting agents, further incrementing their functionality. Of recent interest as potential nanocarriers are spiropyrans; a family of photochromic molecular switches. Due to their multi-responsiveness to endo- and exogenous stimuli, and their intrinsic biocompatibility, they have been utilised in various drug delivery systems (DDSs) to date. In this review, we provide an overview of the developments in spiropyran-based DDSs. The benefits and drawbacks of utilising spiropyrans in drug delivery are assessed and an outline of spiropyran-based drug delivery systems is presented.The development of near-infrared-II (NIR-II) fluorescence imaging has implemented real-time detection of biological cells, tissues and body, monitoring the disease processes and even enabling the direct conduct of surgical procedures. NIR-II fluorescence imaging provides better imaging contrast and penetration depth, benefiting from the reducing photon scattering, light absorption and autofluorescence. The majority of current NIR-II fluorophores suffer from uncontrollable emission wavelength and low quantum yields issues, impeding the clinical translation of NIR-II bioimaging. By lengthening the polymethine chain, tailoring heterocyclic modification and conjugating electron-donating groups, cyanine dyes have been proved to be ideal NIR-II fluorophores with both tunable emission and brightness. However, a simpler and faster method for synthesizing NIR-II dyes with longer wavelengths and better stability still needs to be explored. This minireview will outline the recent progress of cyanine dyes with NIR-II emission, particularly emphasizing their pharmacokinetic enhancement and potential clinical translation.In the zinc hydrometallurgical purification process, the concentration ratio of zinc ion to trace nickel ion is as high as 105, so that the nickel spectral signal is completely covered by high concentration zinc signal, resulting in low sensitivity and nonlinear characteristics of nickel spectral signal. Aiming at the problem that it is difficult to detect nickel in zinc sulfate solution, this paper proposes a nonlinear integrated modeling method of extended Kalman filter based on Adaboost algorithm. First, a non-linear nickel model is established based on nickel standard solution. Second, an extended Kalman filter wavelength optimization method based on correlation coefficient is proposed to select wavelength variables with high signal sensitivity, large amount of information and strong nonlinear correlation. Finally, a nonlinear integrated modeling method based on Adaboost algorithm is proposed, which uses extended Kalman filter as a basic submodel, and realizes the stable detection of trace nickel through the weighted combination of multiple basic models. The results show that the average relative error of this method for detecting nickel is 4.56%, which achieves accurate detection of trace nickel in zinc sulfate solution.In this work, a facile one-pot hydrothermal route was employed to synthesize a series of fluorescent carbon dots (CDs) by using 20 natural amino acids, respectively, as the starting materials. It was found that the CDs synthesized using phenylalanine could possess the intrinsic peroxidase-like activity that could effectively catalyze a traditional peroxidase substrate like 3, 3', 5, 5'- tetramethylbenzidine (TMB) in the presence of H2O2 to produce a blue solution; thereby, a catalytic sensing system for H2O2 has been developed. On the basis of this catalytic reaction, together with the fact that glucose oxidase (GOx) can catalyze the hydrolysis of glucose to generate H2O2, a sensitive catalytic sensing system for glucose could be further established. Furthermore, based on this catalytic reaction, taken together with the two enzymatic catalytic systems of acetylcholinesterase (AChE) and choline oxidase (CHO), a highly sensitive multi-catalytic sensing system could be successfully developed for organophosphorus (OPs) pesticides such as dimethoate, DDVP, and parathion-methyl. Limit of detections (LODs) of H2O2 and glucose were estimated to be 6.5 and 0.84 μM, respectively. Due to their flexibility, proteins tend to respond to their environmental conditions and can undergo structural and conformational changes. For instance, alterations in physiological factors such as temperature, ions concentration, or pH as well as the adsorption to an interface can initiate protein aggregation. Therefore, at different surfaces and interfaces the characterization of the structural and colloidal stability of proteins, which is mainly influenced by their electrostatic and hydrophobic interactions, is of fundamental importance. link2 In this study, we utilized sum frequency generation (SFG) spectroscopy to assess the role of solution pH on the polarity and magnitude of the electric field within the hydration shell of selected model proteins adsorbed to a hydrophobic surface. We used polystyrene (PS) as a model hydrophobic surface and determined the isoelectric point (IEP) of four structurally different model proteins. Comparing the measured IEP of proteins at the PS/solution or air/solution interface with that determined in the bulk solution via zeta potential measurement, we found significant similarities between the IEP of surface adsorbed proteins and those in the bulk aqueous phase. The pH dependence behavior of proteins was correlated to their amino acid composition and degree of hydrophobicity.MALDI-MS-based glycan isotope labeling methods have been effectively and widely used for quantitative glycomics. However, interpretation of the data produced by MALDI-MS is inaccurate and tedious because the bioinformatic tools are inadequate. In this work, we present gQuant, an automated tool for MALDI-MS-based glycan isotope labeling data processing. gQuant was designed with a set of dedicated algorithms to improve the efficiency, accuracy and convenience of quantitation data processing. When tested on the reference data set, gQuant showed a fast processing speed, as it was able to search the glycan data of model glycoproteins in a few minutes and reported more results than the manual analysis did. The reported quantitation ratios matched well with the experimental glycan mixture ratios ranging from 110 to 101. In addition, gQuant is fully open-source and is coded in Python, which is supported by most operating systems, and it has a user-friendly interface. gQuant can be easily adapted by users for specific experimental designs, such as specific glycan databases, different derivatization types and relative quantitation designs and can thus facilitate fast glycomic quantitation for clinical sample analysis using MALDI-MS-based stable isotope labeling.Aspergillus niger, one of the most common and important fungal species, is ubiquitous in various environments. A. link3 niger isolates possess a large number of cryptic biosynthetic gene clusters (BGCs) and produce various biomolecules as secondary metabolites with a broad spectrum of application fields covering agriculture, food, and pharmaceutical industry. By extensive literature search, this review with a comprehensive summary on biological and chemical aspects of A. niger strains including their sources, BGCs, and secondary metabolites as well as biological properties and biosynthetic pathways is presented. Future perspectives on the discovery of more A. niger-derived functional biomolecules are also provided in this review.
