Physiological data were obtained to further confirm the role CIPK23 in alleviating NH4+ toxicity. Taken all together, CIPK23 might function in different tissues to reduce stress-induced NH4+ toxicity associated with high NH4+/K+ ratios by regulating the NH4+-K+ balance in Arabidopsis.Oligodendrocytes are the myelinating cells of the central nervous system (CNS). These cells arise during the embryonic development by the specification of the neural stem cells to oligodendroglial progenitor cells (OPC); newly formed OPC proliferate, migrate, differentiate, and mature to myelinating oligodendrocytes in the perinatal period. It is known that progesterone promotes the proliferation and differentiation of OPC in early postnatal life through the activation of the intracellular progesterone receptor (PR). Progesterone supports nerve myelination after spinal cord injury in adults. However, the role of progesterone in embryonic OPC differentiation as well as the specific PR isoform involved in progesterone actions in these cells is unknown. By using primary cultures obtained from the embryonic mouse spinal cord, we showed that embryonic OPC expresses both PR-A and PR-B isoforms. We found that progesterone increases the proliferation, differentiation, and myelination potential of embryonic OPC through its PR by upregulating the expression of oligodendroglial genes such as neuron/glia antigen 2 (NG2), sex determining region Y-box9 (SOX9), myelin basic protein (MBP), 2',3'-cyclic-nucleotide 3'-phosphodiesterase (CNP1), and NK6 homeobox 1 (NKX 6.1). These effects are likely mediated by PR-B, as they are blocked by the silencing of this isoform. AUZ454 The results suggest that progesterone contributes to the process of oligodendrogenesis during prenatal life through specific activation of PR-B.The purpose of the present study was to investigate the association between different levels of health-related physical fitness measurements and obesity status in Taiwanese adults. A cross-sectional study was conducted by reviewing the National Physical Fitness Survey in Taiwan (HPFSIT) database. Responses from 60,056 participants, aged 23-64 years from the database were collected in the present study. Data from a standardized structured questionnaire and health-related physical fitness tests were analyzed. The quartiles of each physical fitness measurement were used for unconditional logistic regression analyses. Our results indicated clear trends in the association between cardiorespiratory fitness and overweight/obesity. Overweight and obesity were associated with a 10% to 60% increased risk of low levels of cardiorespiratory fitness in men and a 10% to almost 30% increased risk in women. However, the association between muscle strength/endurance and obesity status as well as flexibility and obesity status needs further investigation.Action on the Sustainable Development Goals (SDGs) needs to become real and impactful, taking a "whole systems" perspective on levers for systems change. This article reviews what we have learned over the past century about the large-scale outcome of health inequality, and what we know about the behaviour of complex social systems. This combined knowledge provides lessons on the nature of inequality and what effective action on our big goals, like the SDGs, might look like. It argues that economic theories and positivist social theories which have dominated the last 150 years have largely excluded the nature of human connections to each other, and the environment. This exclusion of intimacy has legitimatised arguments that only value-free economic processes matter for macro human systems, and only abstract measurement constitutes valuable social science. Theories of complex systems provide an alternative perspective. One where health inequality is viewed as emergent, and causes are systemic and compounding. Action therefore needs to be intensely local, with power relationships key to transformation. This requires conscious and difficult intervention on the intolerable accumulation of resources; improved reciprocity between social groups; and reversal of system flows, which at present ebb away from the local and those already disadvantaged.Developments in mid-infrared Differential Absorption Lidar (DIAL), for gas remote sensing, have received a significant amount of research in recent years. In this paper, a high repetition rate tunable mid-infrared DIAL, mounted on a mobile platform, has been built for long range remote detection of gas plumes. The lidar uses a solid-state tunable optical parametric oscillator laser, which can emit laser pulse with repetition rate of 500 Hz and between the band from 2.5 μm to 4 μm. A monitoring channel has been used to record the laser energy in real-time and correct signals. Convolution correction technology has also been incorporated to choose the laser wavelengths. Taking NO2 and SO2 as examples, lidar system calibration experiment and open field observation experiment have been carried out. The observation results show that the minimum detection sensitivity of NO2 and SO2 can reach 0.07 mg/m3, and 0.31 mg/m3, respectively. The effective temporal resolution can reach second level for the high repetition rate of the laser, which demonstrates that the system can be used for the real-time remote sensing of atmospheric pollution gas.BACKGROUND Periodontal disease (PD) is a highly prevalent inflammatory disease in dogs. This disease is initiated by a polymicrobial biofilm on the teeth surface, whose control includes its prevention and removal. Recently, it was shown that nisin displays antimicrobial activity against canine PD-related bacteria. Moreover, guar gum biogel has shown to be a promising topical delivery system for nisin. METHODS In this study we aimed to evaluate the antimicrobial activity of the nisin-biogel in the presence of canine saliva and after a 24-month storage, at different conditions, using a canine oral enterococci collection. We also studied the nisin-biogel cytotoxicity using a Vero cell line and canine primary intestinal fibroblasts. RESULTS The presence of saliva hampers nisin-biogel antimicrobial activity, and higher nisin concentrations were required for an effective activity. A significant reduction (p ≤ 0.05) in inhibitory activity was observed for nisin-biogel solutions stored at 37 °C, over a 24-month period, which was not observed with the other conditions. The nisin-biogel showed no cytotoxicity against the cells tested at concentrations up to 200 µg/mL. CONCLUSIONS Our results confirmed the potential of the nisin-biogel for canine PD control, supporting the development of an in vivo clinical trial.Electronic medical records are an integral part of medical texts. Entity recognition of electronic medical records has triggered many studies that propose many entity extraction methods. In this paper, an entity extraction model is proposed to extract entities from Chinese Electronic Medical Records (CEMR). In the input layer of the model, we use word embedding and dictionary features embedding as input vectors, where word embedding consists of a character representation and a word representation. Then, the input vectors are fed to the bidirectional long short-term memory to capture contextual features. Finally, a conditional random field is employed to capture dependencies between neighboring tags. We performed experiments on body classification task, and the F1 values reached 90.65%. We also performed experiments on anatomic region recognition task, and the F1 values reached 93.89%. On both tasks, our model had higher performance than state-of-the-art models, such as Bi-LSTM-CRF, Bi-LSTM-Attention, and Vote. Through experiments, our model has a good effect when dealing with small frequency entities and unknown entities; with a small training dataset, our method showed 2-4% improvement on F1 value compared to the basic Bi-LSTM-CRF models. Additionally, on anatomic region recognition task, besides using our proposed entity extraction model, 12 rules we designed and domain dictionary were adopted. Then, in this task, the weighted F1 value of the three specific entities extraction reached 84.36%.The incidence of cacao swollen shoot disease (CSSD) in cacao (Theobroma cacao L.) has increased in West Africa since ~2000. To investigate the genomic and species diversity of the CSSD-badnaviruses infecting cacao in Côte d'Ivoire and Ghana, symptomatic leaves were subjected to high-throughput sequencing. Among the 30 newly determined genomes, three badnaviruses were identified, Cacao swollen shoot Togo B virus (CSSTBV), Cacao swollen shoot CD virus, and Cacao swollen shoot CE virus (CSSCEV). The phylogenetic trees reconstructed for the reverse transcriptase (RT) and ribonuclease H (RNase H) sequences were incongruent with the complete viral genomes, which had the most robust statistical support. Recombination seems to be involved in the CSSD-badnavirus diversification. The genomic diversity varied among different CSSD-badnaviruses, with CSSTBV showing the lowest nucleotide diversity (π = 0.06236), and CSSCEV exhibiting the greatest variability (π = 0.21911). Evidence of strong purifying selection was found in the coding regions of the CSSTBV isolates.The effect of neutral salts on protein conformation was first analyzed by Hofmeister in 1888, however, even today this phenomenon is not completely understood. To clarify this effect, we studied changes in the secondary structure of two proteins human serum albumin with predominantly α-helical structure and porcine pancreas β-trypsin with the typical β-structural arrangement in aqueous solutions of neutral salts (KSCN, KCl, (NH4)2SO4). The changes in the secondary structure were studied at 23 °C and 80 °C by using the second derivative deconvolution method of the IR spectra. Our results demonstrated that the ability of the salts to stabilize/destabilize these two proteins correlates with the Hofmeister series of ions. At the same time, some exceptions were also observed. The destabilization of the native structures of both α-helical albumin and β-structural trypsin upon interaction with neutral salts leads to the formation of intermolecular β-sheets typical for amyloid fibrils or amorphous aggregates. Thus, our quantitative FTIR-spectroscopy analysis allowed us to further clarify the mechanisms and complexity of the neutral salt actions on protein structures which may lead to strategies preventing unwelcome misfolding of proteins.Familial hypercholesterolemia (FH) is a genetic disorder of cholesterol metabolism that affects an estimated 1/250 persons in the United States and abroad. FH is hallmarked by high low-density lipoprotein (LDL) cholesterol and an increased risk of premature atherosclerotic cardiovascular disease. This review summarizes recent global evidence showing the utility of FH genetic testing across diverse populations. Clinical and other qualitative outcomes following FH genetic testing were improved FH diagnosis, treatment initiation or continued treatment, treatment modification, improved total or LDL cholesterol levels, education on lifestyle management, and genetic counseling. This summary of evidence should be considered by those seeking overall evidence and knowledge gaps on the utility of FH genetic testing from a global perspective and for certain ethnic and age populations. These findings can be used to inform insurance policies and coverage decisions for FH genetic testing, policy recommendations to reduce the clinical and public health burden of FH, clinical practice and guidelines to improve the management of FH populations, and ongoing research involving FH genetic testing.
