CNP-stimulated cGMP efflux from C6 glioma cells was significantly reduced under conditions of hyperammonaemia, potentially via a mechanism involving changed in phosphodiesterase expression. Hyperammonaemia-stimulated ROS production was unaffected by CNP but enhanced by a nitric oxide donor in C6 cells. Extracellular vesicle production from C6 cells was enhanced by hyperammonaemia, and these vesicles caused impaired CNP-stimulated cGMP signalling in GPNT cells. Collectively, these data demonstrate functional interaction between CNP signalling and hyperammonaemia in C6 glioma and GPNT cells, but the exact mechanisms remain to be established.P53, P63, and P73 proteins belong to the P53 family of transcription factors, sharing a common gene organization that, from the P1 and P2 promoters, produces two groups of mRNAs encoding proteins with different N-terminal regions; moreover, alternative splicing events at C-terminus further contribute to the generation of multiple isoforms. P53 family proteins can influence a plethora of cellular pathways mainly through the direct binding to specific DNA sequences known as response elements (REs), and the transactivation of the corresponding target genes. However, the transcriptional activation by P53 family members can be regulated at multiple levels, including the DNA topology at responsive promoters. Here, by using a yeast-based functional assay, we evaluated the influence that a G-quadruplex (G4) prone sequence adjacent to the p53 RE derived from the apoptotic PUMA target gene can exert on the transactivation potential of full-length and N-terminal truncated P53 family α isoforms (wild-type and mutant). Our results show that the presence of a G4 prone sequence upstream or downstream of the P53 RE leads to significant changes in the relative activity of P53 family proteins, emphasizing the potential role of structural DNA features as modifiers of P53 family functions at target promoter sites.Epoxy vitrimers with reprocessability, recyclability, and a self-healing performance have attracted increasingly attention, but are usually fabricated through static curing procedures with a low production efficiency. Herein, we report a new approach to fabricate an epoxy vitrimer by dynamic crosslinking in a torque rheometer, using diglycidyl ether of bisphenol A and sebacic acid as the epoxy resin and curing agent, respectively, in the presence of zinc acetylacetonate as the transesterification catalyst. The optimal condition for fabricating the epoxy vitrimer (EVD) was dynamic crosslinking at 180 °C for ~11 min. A control epoxy vitrimer (EVS) was prepared by static curing at 180 °C for ~11 min. The structure, properties, and stress relaxation of the EVD and EVS were comparatively investigated in detail. The EVS did not cure completely during static curing, as evidenced by the continuously increasing gel fraction when subjected to compression molding. The gel fraction of the EVD did not change with compression molding at the same condition. The physical, mechanical, and stress relaxation properties of the EVD prepared by dynamic crosslinking were comparable to those of the EVS fabricated by static curing, despite small differences in the specific property parameters. This study demonstrated that dynamic crosslinking provides a new technique to efficiently fabricate an epoxy vitrimer.Candida auris is an emerging MDR pathogen raising major concerns worldwide. In Italy, it was first and only identified in July 2019 in our hospital (San Martino Hospital, Genoa), where infection or colonization cases have been increasingly recognized during the following months. To gain insights into the introduction, transmission dynamics, and resistance traits of this fungal pathogen, consecutive C. auris isolates collected from July 2019 to May 2020 (n = 10) were subjected to whole-genome sequencing (WGS) and antifungal susceptibility testing (AST); patients' clinical and trace data were also collected. WGS resolved all isolates within the genetic clade I (South Asian) and showed that all but one were part of a cluster likely stemming from the index case. Phylogenetic molecular clock analyses predicted a recent introduction (May 2019) in the hospital setting and suggested that most transmissions were associated with a ward converted to a COVID-19-dedicated ICU during the pandemic. All isolates were resistant to amphotericin B, voriconazole, and fluconazole at high-level, owing to mutations in ERG11(K143R) and TACB1(A640V). Continued efforts to implement stringent infection prevention and control strategies are warranted to limit the spread of this emerging pathogen within the healthcare system.The underwater wireless sensor networks (UWSNs) have been applied in lots of fields such as environment monitoring, military surveillance, data collection, etc. Deployment of sensor nodes in 3D UWSNs is a crucial issue, however, it is a challenging problem due to the complex underwater environment. This paper proposes a growth ring style uneven node depth-adjustment self-deployment optimization algorithm (GRSUNDSOA) to improve the coverage and reliability of UWSNs, meanwhile, and to solve the problem of energy holes. In detail, a growth ring style-based scheme is proposed for constructing the connective tree structure of sensor nodes and a global optimal depth-adjustment algorithm with the goal of comprehensive optimization of both maximizing coverage utilization and energy balance is proposed. Initially, the nodes are scattered to the water surface to form a connected network on this 2D plane. Then, starting from sink node, a growth ring style increment strategy is presented to organize the common nodes as tree structures and each root of subtree is determined. Meanwhile, with the goal of global maximizing coverage utilization and energy balance, all nodes depths are computed iteratively. Finally, all the nodes dive to the computed position once and a 3D underwater connected network with non-uniform distribution and balanced energy is constructed. A series of simulation experiments are performed. The simulation results show that the coverage and reliability of UWSN are improved greatly under the condition of full connectivity and energy balance, and the issue of energy hole can be avoided effectively. Therefore, GRSUNDSOA can prolong the lifetime of UWSN significantly.
