The extensive use of antibiotics in aquaculture has resulted in the prevalence of antibiotic-resistant bacteria and, consequently, new antibacterial strategies or drugs with clear modes of action are urgently needed. Antimicrobial peptides (AMPs) are currently widely considered as alternatives to antibiotics in the treatment of infections in aquatic animals. In this study, we aimed to evaluate the effects of NKL-24, a truncated peptide derived from zebrafish NK-lysin, against Yesso scallop (Patinopecten yessoensis) pathogen, Vibrio parahaemolyticus. The results showed that NKL-24 had a potent antibacterial effect against V. parahaemolyticus via a membrane active cell-killing mechanism. The in vitro study showed that sub-lethal levels of NKL-24 obviously reduced bacterial swimming ability and downregulated the transcription of the selected genes associated with V. parahaemolyticus virulence. Studies on NKL-24 biosafety in hemocytes and in Yesso scallop have shown no adverse effects from this peptide. Bacteria challenge test results demonstrated that NKL-24 significantly decreased the mortality and inhibited bacterial growth in the scallop infected with V. parahaemolyticus, while further in vivo examination revealed that NKL-24 could enhance non-specific immune parameters. Moreover, NKL-24 was capable of modulating a series of V. parahaemolyticus-responsive genes in the scallop. These results suggest the protective action of NKL-24 against V. parahaemolyticus and the potential of this peptide as a promising candidate for aquaculture applications.Effects of dietary hydroalcoholic extract of Zataria multiflora (ZE) on growth performance, plasma and hepatic antioxidant capacities, and humoral and skin mucus immune parameters were evaluated in rainbow trout (Oncorhynchus mykiss) juveniles. in vitro tests showed that ZE had antioxidant property comparable to butylated hydroxytoluene (BHT) at 100-200 μg/mL concentrations, although its antioxidant property was lower than BHT at concentration below 100 μg/mL. Moreover, ZE had anti-bacterial activity against Aeromonas hydrophila, which was 30-50% lower than that of tetracycline. After feeding the fish with diets supplemented with 0 (CT, 1 (ZE1), 2 (ZE2), and 3 (ZE3) g/kg ZE for eight weeks, there were no significant differences in growth performance and feed efficiency among the treatments; however, the fish in ZE2 and ZE3 treatments showed significantly higher survival than the fish in CT treatment. Blood leukocyte counts, plasma globulin, total immunoglobulin, lysozyme and bactericidal activity against A. hevel of 2 g/kg is suggested for rainbow trout feed supplementation to augment fish survival, antioxidant and immune strength.Gut microbiota plays a central part in the regulation of multiple host metabolic pathways, such as homeostasis, immunostasis, mucosa permeability, and even brain development. In this study, germ-free (GF) and conventionally reared (CV) zebrafish models utilized for studying the role of gut bacteria Vibrio sp. and Aeromonas sp. After the analysis of gut microbial profile in zebrafish male and female at three-month age, Proteobacteria and Fusobacteria dominated the main composition of zebrafish intestinal microflora. However, the relative richness of them was different base on gender variance. Aeromonas sp. and Vibrio sp. belonging to Proteobacteria phylum of bacteria were isolated from zebrafish gut, and their potential capacities to trigger innate immunity were investigated. In gut microbiota absence, the expression levels of the innate immunity genes in the GF group were not significantly changed compared to the CV group. After exposure to Aeromonas sp. and Vibrio sp., the expression levels of myd88, TLRs-, and inflammation-related genes were increased in both GF and CV groups, except tlr2 and NLRs-related genes. However, the expression level of NF-κB and JNK/AP-1 pathway genes were all decreased after exposure to Aeromonas sp. and Vibrio sp. in both GF and CV groups. Interestingly, inflammation-related genes (tnfa, tnfb, and il1β) were activated in the CV group, and there were not significantly changed in the GF group, indicating that other bacteria were indispensable for Aeromonas sp. or Vibrio sp. to activate the inflammation response. Taken together, this is the first study of gut bacteria Vibrio sp. and Aeromonas sp. prompting the innate immune response using the GF and CV zebrafish model.The integration of multimodal functions into one nanoplatform holds great promise for enhancing anticancer drug action and mitigating adverse effects. Herein, we prepared hyaluronic acid-functionalized regenerated silk fibroin-based nanoparticles (NPs) loading with photosensitizer (NIR770) and doxorubicin (DOX). The resultant HNDNPs had a desirable diameter of 161.0 nm and a negative zeta-potential of -30.5 mV. Interestingly, they showed excellent responses when triggered with various stimuli (acidity, reactive oxygen species, glutathione, hyaluronidase, or hyperthermia). Cell experiments revealed that HNDNPs could be specifically internalized by A549 cells, and efficiently released the payloads into the cytoplasm. Moreover, NIR770 was preferentially retained in mitochondria due to its lipophilic and cationic properties, which exhibited highly efficient photothermal therapy and photodynamic therapy upon near infrared (NIR) irradiation. Meanwhile, DOX molecules were mainly accumulated in the nucleus. Intravenous injection of HNDNPs into mice followed by NIR irradiation provided excellent multimodal imaging (NIR, photothermal, and photoacoustic imaging), almost eliminated the entire tumor, and greatly prolonged mice survival time with no side effects. However, before these procedures, it is necessary to follow sample pre-conditioning steps, such as several precipitation stages and/or ultrafiltration/diafiltration processes. In this work, an efficient affinity chromatographic matrix for eCG purification directly from plasma was developed. The matrix consisted of chitosan mini-spheres with immobilized wheat germ agglutinin (WGA). The matrix allowed 98% adsorption of eCG directly from plasma without any pre-treatment with an overall yield of around 60%. The matrix chosen was able to maintain the efficient performance of the purification process for three consecutive cycles.
