Although widely used in the detection and characterization of protein-protein interactions, Y2H screening has been under-used for the engineering of new optogenetic tools or the improvement of existing tools. Here we explore the feasibility of using Y2H selection and screening to evaluate libraries of photoswitchable protein-protein interactions. We targeted the interaction between circularly permuted photoactive yellow protein (cPYP) and its binding partner binder of PYP dark-state (BoPD) by mutating a set of four surface residues of cPYP that contribute to the binding interface. A library of ~10,000 variants was expressed in yeast together with BoPD in a Y2H format. An initial selection for the cPYP/BoPD interaction was performed using a range of concentrations of the cPYP chromophore. As expected, the majority (>90% of cPYP variants) no longer bound to BoPD. Replica plating was then used to evaluate the photoswitchability of the surviving clones. Photoswitchable cPYP variants with BoPD affinities equal to, or higher than, native cPYP were recovered in addition to variants with altered photocycles and binders that interacted with BoPD as apo-proteins. Y2H results reflected protein-protein interaction affinity, expression, photoswitchability, and chromophore uptake, and correlated well with results obtained both in vitro and in mammalian cells. Thus, by systematic variation of selection parameters, Y2H screens can be effectively used to generate new optogenetic tools for controlling protein-protein interactions for use in diverse settings. Therefore, this review presented recent (2012-2018) advances in alkaloids, focusing on new compounds, extraction methods and biological activities. As such, approximately 70 articles were identified, which showed 261 new compounds produced by plants of the families Apocynaceae, Rubiaceae, Annonaceae and Loganiaceae. In addition, different extraction methods were identified, and the structures of the new compounds were analysed. In addition to indole molecules, there were mono-indole-, di-indole-, vinblastine-, vimblastine-, gelsedine-, geissospermidine-, koumine-, geissospermidine-, iboga-, perakine-, corynanthe-, vincamine-, ajmaline-, aspidorpema-, strychnos-type, β-carboline alkaloids and indole alkaloid glucosides. The reported biological activities are mainly anticancer, antibacterial, antimalarial, antifungal, antiparasitic, and antiviral, as well as anti-acetylcholinesterase and anti-butyrylcolinesterase properties. This review serves as a guide for those wishing to find the most recently identified alkaloid structures and their associated activities. AIM Neurodegeneration is one of the serious adverse effects of stimulant agents such as nicotine. Minocycline possess established neuroprotective properties. The role of CREB-BDNF signaling pathway in mediating the neuroprotective effects of minocycline against nicotine-induced neurodegeneration in rats was evaluated in current study. METHODS Seventy adult male rats were divided randomly into seven groups. Group 1 and 2, received 0.7 ml/rat of normal saline (i.p) and nicotine (10 mg/kg, s.c) respectively. Groups 3, 4, 5 and 6, treated concurrently with nicotine (10 mg/kg) and minocycline (10, 20, 30 and 40 mg/kg, i.p, respectively) for 21 days. Group 7 received minocycline alone (40 mg/kg, i.p) for 21 days.
