Glutamine inhibition has been demonstrated an antifibrotic effect in iatrogenic laryngotracheal stenosis (iLTS) scar fibroblasts in vitro. We hypothesize that broadly active glutamine antagonist, DON will reduce collagen formation and fibrosis-associated gene expression in iLTS mice.

Prospective controlled animal study.

iLTS in mice were induced by chemomechanical injury of the trachea using a bleomycin-coated wire brush. PBS or DON (1.3 mg/kg) were administered by intraperitoneal injection (i.p.) every other day. Laryngotracheal complexes were harvested at days 7 and 14 after the initiation of DON treatment for the measurement of lamina propria thickness, trichrome stain, immunofluorescence staining of collagen 1, and fibrosis-associated gene expression.

The study demonstrated that DON treatment reduced lamina propria thickness (P=.025) and collagen formation in trichrome stain and immunofluorescence staining of collagen 1. In addition, DON decreased fibrosis-associated gene expression in iLTS mice. At day 7, DON inhibited Col1a1 (P < .0001), Col3a1 (P=.0046), Col5a1 (P < .0001), and Tgfβ (P=.023) expression. At day 14, DON reduced Co1a1 (P =.0076) and Tgfβ (P =.023) expression.

Broadly active glutamine antagonist, DON, significantly reduces fibrosis in iLTS mice. These results suggest that the concept of glutamine inhibition may be a therapeutic option to reduce fibrosis in the laryngotracheal stenosis.

N/A Laryngoscope, 131E2125-E2130, 2021.A computational method to investigate the global conformational change of a protein is proposed by combining the linear response path following (LRPF) method and three-dimensional reference interaction site model (3D-RISM) theory, which is referred to as the LRPF/3D-RISM method. The proposed method makes it possible to efficiently simulate protein conformational changes caused by either solutions of varying concentrations or the presence of cosolvent species by taking advantage of the LRPF and 3D-RISM. The proposed method is applied to the urea-induced denaturation of ubiquitin. The LRPF/3D-RISM trajectories successfully simulate the early stage of the denaturation process within the simulation time of 300 ns, whereas no significant structural change is observed even in the 1 μs standard MD simulation. The obtained LRPF/3D-RISM trajectories reproduce the mechanism of the urea denaturation of ubiquitin reported in previous studies, and demonstrate the high efficiency of the method.Protective efficacy of rutin over liver, kidney, and brain dysfunctions was evaluated in this investigation. Rutin (5, 10, and 20 mg/kg) was administered continuously for 6 days followed by single dose of D-galactosamine (300 mg/kg I.P.) and lipopolysaccharide (50 µg/kg I.P.) on the 6th day. Hematological, serological, biochemical, and histological aspects were considered for this study. One-way ANOVA (p ≤ .05) followed by Tukey's HSD post hoc test determined the statistical significance. Serum AST, ALT, ALP, urea, uric acid, and creatinine were increased significantly, whereas albumin and glucose were significantly decreased after combined exposure to LPS and D-GalN. Glutathione level and activity of SOD and catalase were decreased, whereas lipid peroxidation, triglycerides, and cholesterol were increased in tissue samples due to LPS- and D-GalN-induced toxicity. Prophylactic treatment of rutin maintained studied variables toward control claiming the protective role of rutin. PRACTICAL APPLICATION Rutin is plenteous in a variety of commonly ingested foods such as onion, wine, grape, citrus fruits, tea, and buckwheat. Rutin supplement is recommended for the treatment of various diseases such as varicose veins, internal bleeding, or hemorrhoids. Rutin is better than well-known antithrombic agent, Juniferdin, or Bacitracin. In the present study, rutin showed protective effects against LPS- and D-GalN-induced multiorgan dysfunctions due to its anti-inflammatory and antioxidant properties. Therefore, rutin may be developed and practiced as a food supplement to cope with acute organ dysfunctions caused by inflammatory and oxidative damage.Long bones comprise articular ends (epiphyses) joined by transitional metaphyses and a diaphysis (shaft). The structure of the latter is often viewed as regularly tubular across tetrapods (limbed vertebrates). However, assessments of the bone structure along the whole diaphysis are rare. Here, I assess whole-diaphysis profiles of global compactness (bone fraction) of 164 species of extant and extinct therian mammals (marsupials + placentals) in a phylogenetically informed context. Generally terrestrial, mammals have acquired multiple times the highly specialized aerial, fully aquatic, and subterranean lifestyles, allowing to potentially associate specific traits with these lifestyles. I show that there is a consistent increase in global compactness along the diaphysis in most mammals. This pattern is modified in a limited number of specialized species all aerial clades (gliders and bats) have rather uniform and low values, while cetaceans' humeral diaphysis is marked by a slightly more compact mid-diaphyseal region. Among subterranean clades, structure alterations are most obvious in fossorial talpids (true moles) and their highly modified humerus. These results call for the investigation of bone structure in whole skeletal elements of key fossils in order to reconstruct the patterns of evolutionary modifications associated with lifestyle transitions.To investigate the dynamic changes of Krebs von den Lungen-6 (KL-6) among patients with coronavirus disease 2019 (COVID-19) and the role of KL-6 as a noninvasive biomarker for predicting long-term lung injury, the clinical information and laboratory tests of 166 COVID-19 patients were collected, and a correlation analysis between KL-6 and other parameters was conducted. There were 17 (10.2%, 17/166) severe/critical and 149 (89.8%, 149/166) mild COVID-19 patients in our cohort. Serum KL-6 was significantly higher in severe/critical COVID-19 patients than in mild patients (median 898.0 vs. 451.2 U/ml, p 100 days postonset) was well correlated to those concentrations within 10 days postonset (Pearson r = .867, p  less then  .001), indicating the prognostic value of KL-6 levels in predicting lung injury after discharge. Finally, elevated KL-6 was found to be significantly correlated to coagulation disorders, and T cells subsets dysfunctions. As mutations in RdRp may affect formation of the RdRp-nsp7-nsp8 supercomplex, we analyzed viral genomes to identify mutations in nsp7 and nsp8 protein sequences. Based on in silico analysis of predicted structures of the supercomplex comprising of native and mutated proteins, we demonstrate that specific mutations in nsp7 and nsp8 proteins may have a role in stabilization of the replication/transcription complex.

Rating scales are designed to complement traditional performance-based measures, and both can provide useful information about the functioning of youth with histories of prenatal alcohol exposure. Few studies, however, have compared ratings from multiple informants or the relationship between these subjective rating scale scores and the objective results from laboratory performance-based scales.

The current study addressed both of these questions in 3 study groups children with histories of prenatal alcohol exposure (n = 47), attention-deficit/hyperactivity disorder (ADHD; n=41), and typically developing controls (CON; n=73). All subjects completed a standardized neuropsychological test battery, including laboratory measures of executive functioning and a self-report measure of executive function behaviors. Parents and teachers completed corresponding rating scales of executive function behaviors for each subject. This study assessed the relationship between these behavior rating scales and corresponding udy indicate that behavior ratings can be used to supplement laboratory measures but may not be measuring cognitive abilities regardless of whether a clinical diagnosis is present. A multimethod approach should be used when measuring skills in this domain. This was one of the first studies to examine cross-informant agreement in a sample of children with prenatal alcohol exposure. For specified values of the particle volume fraction and particle-to-fluid density ratio in a suspension, the relaxation times in the growth of the particle mobility in transient electrophoresis and transient sedimentation are equivalent.In a sample of 559 children (ages 9-18), researchers investigated whether (a) fear of abandonment mediated the association between postdivorce interparental conflict (IPC) and mental health problems, and (b) parent-child relationship quality moderated the association between IPC and fear of abandonment. Mediation analyses indicated that pretest IPC predicted fear of abandonment 3 months later, which then predicted child- and teacher-reported mental health problems 10 months later. The hypothesized protective effect of a high-quality parent-child relationship was not observed. IPC predicted fear of abandonment for all children, except for those with low- and moderate-quality father-child relationships, for whom IPC was not significantly related to fear of abandonment. Findings highlight the need to optimize child coping programs and improve parenting-after-divorce programs to reduce IPC.
