The aim of this study was to assess sperm motility, morphology and DNA fragmentation before and after swim-up of a second ejaculate obtained after a short abstinence period in normozoospermic men and oligo-astheno-teratozoospermic (OAT) patients. MATERIAL AND METHODS Semen analyses and swim-up preparations of two consecutive semen samples (collected within 1 h) were carried out in 30 normozoospermic and 35 OAT patients enrolled in an assisted reproductive technique (ART) program. RESULTS Compared to the first ejaculate, the second sample showed a higher percentage of spermatozoa with normal form (p less then 0.01) and lower percentage of spermatozoa with DNA fragmentation (p less then 0.01) in normozoospermic men, whereas a higher percentage of spermatozoa with progressive motility (p less then 0.001) and normal morphology (p less then 0.0001) was found in OAT patients. Swim-up separation showed a lower DNA fragmentation rate (p less then 0.05) in the second ejaculate in normozoospermic men, whereas the second ejaculate of OAT patents showed an increase in normally-shaped spermatozoa (p less then 0.01) and lower percentage of spermatozoa with fragmented DNA (p less then 0.001) compared to the first one. CONCLUSIONS Swim-up separation of a second ejaculate collected within 1 h might be suggested for ART procedures, especially in OAT patients.Scrub typhus, a disease caused by Orientia tsutsugamushi, affects more than one billion people globally with an average fatality rate of 6%. Humans are accidentally infected through the bite of trombiculid mite larvae (chiggers). Chiggers feed on hosts' extracellular fluid for survival and development. O. tsutsugamushi is maintained throughout the chigger's lifespan and over several generations. Although disease-related knowledge is essential in designing effective control strategies, many personnel in related sectors are unfamiliar with this disease and its vector. To tackle this issue, we developed a distance learning tool using educational videos on scrub typhus- and vector-related topics. The learning method is facilitated online, and students and tutors are not required to be physically present at the same place and time, thus allowing flexibility and accessibility. Knowledge improvement of 34 participants from related sectors was evaluated by pre- and post-test questionnaires. Although 54% of participants had prior knowledge of scrub typhus, 76.5% still lack basic knowledge of vector identification. After the distance learning, the average score increased significantly from the baseline (p less then 0.05). Most participants showed interest in the topic and learning method. These results suggest that the distance learning method was promising in distributing health-related information and might be applied to other diseases and communities.BACKGROUND Inflammation is often considered relating to pediatric obstructive sleep apnea (OSA). We conducted a study investigating cytokines, including Il-17 and Il-23, in children with OSA before and after adenotonsillectomy (T&A), compared with controls. METHODS Children with OSA between age 4 and 12 receiving T&A were prospectively followed. Evaluation before and reevaluation six months after the treatment were done, including polysomnography (PSG), blood tests, and questionnaires. Blood samples were obtained to determine the values of high-sensitivity-C-reactive-protein (HS-CRP); tumor-necrosis-factor-alpha (TNF-α); and interleukin (IL)-1, 6, 10, 12, 17, and 23. We compared the results with an age-matched control group. RESULTS We included 55 OSA children and 32 controls. Children with OSA presented significant improvement after T&A in complaints, signs, apnea hypopnea index (AHI) (p less then 0.001), mean oxygen desaturation index (p less then 0.001), and mean oxygen saturation (p = 0.010). Upon entering this study, children with OSA had significantly higher cytokine levels than the controls and significant changes in HS-CRP (p = 0.013), TNF-α (p = 0.057), IL-1β (p = 0.022), IL-10 (p = 0.035), and IL-17 (p = 0.010) after T&A. Children with improved but persistently abnormal AHI did not have all cytokine levels normalized, particularly IL-23 and HS-CRP. CONCLUSION Sleep-disordered breathing can persist after T&A and can continue to have a negative inflammatory effect. HS-CRP and IL-23 may serve as blood markers for the persistence of sleep-disordered breathing after T&A.Caffeine is the most consumed psychostimulant worldwide. The aim of this study was to evaluate the ability to cross the BBB of triterpenoid saponins occurring in Astragalus mongholicus roots. The research was carried out using in silico methods combined with postmortem studies on the brain tissues of mice treated with isolated astragaloside IV (AIV). Firstly, to estimate the ability to cross the BBB by the tested saponins, new quantitative structure-activity relationship (QSAR) models were established. The reliability and predictability of the model based on the values of the blood-brain barrier penetration descriptor (logBB), the difference between the n-octanol/water and cyclohexane/water logP (ΔlogP), the logarithm of n-octanol/water partition coefficient (logPow), and the excess molar refraction (E) were both confirmed using the applicability domain (AD). The critical leverage value h* was found to be 0.128. The relationships between the standardized residuals and the leverages were investigated here. The application of an in vitro acetylcholinesterase-inhibition test showed that AIV can be recognized as the strongest inhibitor among the tested compounds. Therefore, it was isolated for the postmortem studies on brain tissues and blood using semi-preparative HPLC with the mobile phase composed of water, methanol, and ethyl acetate (1.72.116.2 v/v/v). Subjects with asymmetrical brachycephaly started CRO treatment between 3 and 18 months of age. These infants had a cranial vault asymmetry index (CVAI) ≥ 3.5 and a cranial index (CI) ≥ 90. Subjects were excluded if they had any comorbidities affecting growth, dropped out of treatment, were lost to follow-up, or were noncompliant. Factors which were found to statistically influence treatment outcomes were subject initial age, initial CVAI, and initial CI. Overall, younger subjects were more likely to achieve a corrected head shape. The presence of prematurity or torticollis had statistically nonsignificant effects on the success of treatment. Initial CI was found to be a stronger predictor than initial CVAI as to which subjects achieved correction. The less severe the starting CI, the more likely the subject was to achieve full correction. The clinical understanding is that it requires more cranial growth to "round out" a full posterior skull flattening than an asymmetry. Based on the study results, infants with asymmetrical brachycephaly should be treated as early as possible to increase chances of achieving full correction of the deformity.Despite numerous attempts to fabricate the core-shell nanoparticles, novel, simple, and low-cost approaches are still required to produce these efficient nanosystems. In this study, we propose the synthesis of bimetallic core-shell nanoparticles of gold (AuNP) and silver (AgNP) nanostructures via a bioinspired polydopamine (PDOP) layer and their employment as a surface-enhanced Raman spectroscopy (SERS) platform. Herein, the PDOP layer was used as an interface between nanostructures as well as stabilizing and reducing agents for the deposition of silver ions onto the AuNPs. UV-vis absorption spectra and electron microscope images confirmed the deposition of the silver ions and the formation of core-shell nanoparticles. SERS activity tests indicated that both the PDOP thickness and silver deposition time are the dominant parameters that determine the SERS performances of the proposed core-shell system. In comparison to bare AuNPs, more than three times higher SERS signal intensity was obtained with an enhancement factor of 3.5 × 105.Recently, carbon allotropes have received tremendous research interest and paved a new avenue for optical fiber sensing technology. Carbon allotropes exhibit unique sensing properties such as large surface to volume ratios, biocompatibility, and they can serve as molecule enrichers. Meanwhile, optical fibers possess a high degree of surface modification versatility that enables the incorporation of carbon allotropes as the functional coating for a wide range of detection tasks. Moreover, the combination of carbon allotropes and optical fibers also yields high sensitivity and specificity to monitor target molecules in the vicinity of the nanocoating surface. In this review, the development of carbon allotropes-based optical fiber sensors is studied. The first section provides an overview of four different types of carbon allotropes, including carbon nanotubes, carbon dots, graphene, and nanodiamonds. The second section discusses the synthesis approaches used to prepare these carbon allotropes, followed by some deposition techniques to functionalize the surface of the optical fiber, and the associated sensing mechanisms. Numerous applications that have benefitted from carbon allotrope-based optical fiber sensors such as temperature, strain, volatile organic compounds and biosensing applications are reviewed and summarized. Finally, a concluding section highlighting the technological deficiencies, challenges, and suggestions to overcome them is presented.
