A specific inhibitor of the β HSP90 isoform was ineffective suggesting the importance of the α isoform in inflammatory signalling. However, the results from these studies have been inconsistent. The current study investigated the correlation between Hcy and lipid profiles in Chinese community-based population. The participants were composed of 4012 Chinese people aged 30-92 years old, who were recruited from rural and urban communities in the Hunan Province. Non-parametric test and logistic regression were used to examine the distribution of Hcy and lipid profiles (triglyceride [TG], total cholesterol [TC], low-density lipoprotein cholesterol [LDL-C], high-density lipoprotein cholesterol [HDL-C]) and the relationship between them. The median age of subjects was 54.50 years old, and 40.98% were male. Median Hcy was 13.20 μmol/L, and 35.39% had hyperhomocysteinemia (HHcy). Median TG was 1.51 mmol/L, TC was 4.77 mmol/L, LDL-C was 2.62 mmol/L, and HDL-C was 1.27 mmol/L. In multivariable logistic regression analysis, HHcy was associated with high levels of TG (ORmale = 2.240, p less then 0.001; ORfemale = 2.539, p less then 0.001), TC (ORmale = 2.237, p less then 0.001; ORfemale = 2.202, p less then 0.001), and LDL-C (ORmale = 1.413, p = 0.010; ORfemale = 1.617, p less then 0.001) in the different sexes population and low level of HDL-C in females (OR = 1.326, p = 0.023) after adjusting for confounders. HHcy was independently associated with an increasing risk of low HDL-C among females. The regression analysis showed that HHcy was also associated with hypertriglyceridemia, hypercholesterolemia, and high level of LDL-C in males and females from Chinese community-based population, which provides a basis for the treatment and prevention of abnormal lipid metabolism.Chronic granulomatous disease (CGD) is a rare inherited disorder in which phagocytes lack nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity. The most common form is the X-linked CGD (X91-CGD), caused by mutations in the CYBB gene. Clinical, functional and genetic characterizations of 16 CGD cases of male patients and their relatives were performed. We classified them as suffering from different variants of CGD (X910 , X91- or X91+ ), according to NADPH oxidase 2 (NOX2) expression and NADPH oxidase activity in neutrophils. Eleven mutations were novel (nine X910 -CGD and two X91- -CGD). One X910 -CGD was due to a new and extremely rare double missense mutation Thr208Arg-Thr503Ile. We investigated the pathological impact of each single mutation using stable transfection of each mutated cDNA in the NOX2 knock-out PLB-985 cell line. Both mutations leading to X91- -CGD were also novel; one deletion, c.-67delT, was localized in the promoter region of CYBB; the second c.253-1879A>G mutation activates a splicing donor site, which unveils a cryptic acceptor site leading to the inclusion of a 124-nucleotide pseudo-exon between exons 3 and 4 and responsible for the partial loss of NOX2 expression. Both X91- -CGD mutations were characterized by a low cytochrome b558 expression and a faint NADPH oxidase activity. The functional impact of new missense mutations is discussed in the context of a new three-dimensional model of the dehydrogenase domain of NOX2. Our study demonstrates that low NADPH oxidase activity found in both X91- -CGD patients correlates with mild clinical forms of CGD, whereas X910 -CGD and X91+ -CGD cases remain the most clinically severe forms.
Early care and education settings (ECE) are potential venues for young children to develop healthy lifestyle habits. The study assesses training needs and associations with relevant practices of licensed ECE providers across Minnesota and Wisconsin.
A random sample of 823 providers completed a 97-item survey assessing nutrition and physical activity (PA) practices and training needs. Logistic regression, adjusted for program type (center- and family home-based), and location (urban/rural) examined associations between the top 3 selected training needs and provider practices.
Top training needs (1) ways to effectively engage parents about healthy eating and PA, (2) low-cost ways to serve healthy foods, and (3) fun and easy nutrition education curricula. Providers who reported being not happy/somewhat happy vs happy/very happy with parent communication were more likely to need training to engage parents. Among providers who prepared food on-site, shopping at Farmer's Market had lower odds of needing training for serving healthy meals on a budget. Not having completed nutrition training in the past year providers were more likely to need training for fun and easy nutrition education curricula.
