Melanoma is inherently heterogeneous, providing resistance to apoptosis. Anoikis resistance is a hallmark feature of metastatic melanoma to escape apoptosis when cells lose contact with adjacent cells or extracellular matrix. The yes-associated protein transcription co-activator is the effector of Hippo pathway. Herein, we investigated the function of yes-associated protein in anoikis resistance of melanoma cells. When melanoma cells were grown under anchorage-independent condition, anoikis-resistant cells displayed higher levels of yes-associated protein activation than the cells that were attached to the basement membrane, as evidenced by downregulated phosphorylated yes-associated protein at Ser127 and higher expression of downstream genes BCL2 and MCL-1. Yes-associated protein overexpression directly enhanced the anoikis resistance and metastatic potential of melanoma cells. Conversely, yes-associated protein inhibitor CA3 exhibited Dose-dependent induction of anoikis in resistant melanoma cells and exerted great inhibition on cell migration. Knockdown of yes-associated protein expression by shRNA also rendered melanoma cells susceptible to anoikis and interrupted cell invasiveness. Yes-associated protein inhibition in anoikis-resistant cells also reduced the number of metastatic nodules in the lung sections of SCID mice. Clinically, higher yes-associated protein level in the lung metastasis tissues correlated with higher BCL2 and MCL1 expressions compared with the non-metastasis tissues. Overall, our finding suggests that the aberrant activation of yes-associated protein exerts important role on anoikis resistance and metastatic capability of melanoma cells.

Rupture of the plantar fascia is a rare condition. It can also occur spontaneously and with no history of disease of the plantar fascia, above all in athletes. This review aims to systematically analyze all cases described in the literature regarding the rupture of the plantar fascia, evaluating incidence, risk factors, and treatments, considering which procedures show the best outcomes and the highest success rate.

A systematic review of PubMed, Google Scholar, and Cochrane review computerized databases was performed, focusing on articles about cases of rupture of the plantar fascia; 18 studies fulfilled all the criteria and were analyzed. There were no randomized controlled trials.

A total of 155 patients (157 foot) were included in this systematic review. Considering all the studies included, 12 patients had a spontaneous rupture, 138 patients had a diagnosis of plantar fasciitis, and 130 patients were treated with local injections of corticosteroid before the rupture. Only 2 cases of bilateral rupture were reported. In all, 15 studies reported conservative treatment, with a total of 154 patients (156 feet) included. Operative treatment was reported in 3 studies, with 3 patients (3 feet) treated.

Ruptures of the plantar fascia are very rare in asymptomatic patients and more common in patients treated with injection of steroids in the plantar fascia. Conservative treatment, although not standardized in the literature, led to good outcomes in most cases. Chronic ruptures of the fascia should be considered for operative treatment.

This study aimed to assess the effects of extruded diets containing enzymatically hydrolysed poultry byproduct meal (HPM) on cat serum ACE activity and aldosterone (ALD) concentration, nutrient digestibility, and faecal characteristics. On the basis of a preliminary invitro ACE inhibitory activity assay, a commercial HPM and a commercial conventional poultry byproduct meal (CPM) were selected for further investigation. Two isoenergetic and isonitrogenous diets were formulated CPM diet (25.7% CPM) and HPM diet (24.7% HPM). In trial 1, the effect of diet on serum ACE activity and ALD concentration was evaluated using 8 healthy cats (4 female and 4 male, 4.1 ± 0.38 kg BW) in a crossover design, with 5 d of adaptation and blood collection on d 6. In trial 2, apparent total tract digestibility and faecal characteristics were evaluated using 12 cats (6 female and 6 male, 4.0 ± 0.72 kg BW) in a completely randomised design. Serum ACE and ALD were analysed using a mixed model, with diet as the fixed effect and cat as the random effect. Data from trial 2 were subjected to analysis of variance, and means were compared by Tukey's test. In vitro ACE inhibitory activity of HPM (90.4%) was higher than that of CPM (52.0%). Cats fed the HPM diet tended to have lower serum ACE activity than those fed the CPM diet (126 versus 142 U/l, p = 0.09). Serum ALD was not influenced by diet. Diets had similar digestibility values, and faecal consistency scores tended to be higher (firmer faeces) in cats fed the CPM diet than in cats fed the HPM diet (4.6 versus 4.0, p = 0.09). Inclusion of HPM in extruded diets may reduce cat serum ACE activity and promote good faecal consistency without affecting digestibility. Further investigations are needed to explore the potential health benefits of HPM in hypertensive cats.
