Acteoside is an important bioactive natural product distributed in many plant species, composed of four moieties such as caffeic acid, glucose, rhamnose and phenylethyl alcohol, and possesses some bioactivities such as anti-inflammatory, anti-oxidant, neuro-protective, anti-tumor and so on. However, acteoside content in medicinal plants is low, and acteoside stability is bad, so acteoside biosynthesis is a problem. Recent years, acteoside biosynthesis pathway elucidation and bio-production have been widely investigated, so many achievements have been made up to now. In this study, we reviewed current advances in both the elucidation and bio-production such as the putative methods and enzymatic determination of acteoside biosynthesis pathway, functional analyses of the roles of some candidate genes for verbascoside biosynthesis by transgenic technology, acteoside production via metabolic engineering and synthetic biology approaches and plant tissue culture. Moreover, we first established a combined putative acteoside biosynthesis pathway based on its recent studies in animals, plants and microbes. Meanwhile, we pointed out both problems to shortcomings, and highlighted its future development trend. These results will provide references for the complete elucidation of acteoside biosynthesis pathway and the improvement of acteoside content in medicinal plants and acteoside production via microbial and plant metabolic engineering and synthetic biology approaches, and inform the readers critically of the latest developments of them. Agarwood is a resinous wood of Aquilaria species and has been used for various applications. Burning agarwood incense is a common practice in temples and homes in Asia. Kynam is widely regarded as high-quality agarwood in the market. Recently, cultivated grafting Kynam (CGK) has emerged as a new agarwood product in the market, which greatly affects the price of high grading Kynam agarwood. In this study, the morphology, ethanol extract content, and incense chemical profile of CGK was investigated and compared with those of wild Kynam (WK) and cultivated common agarwood (CCA). The incense smoke of CGK was analyzed by thermogravimetric Fourier transform infrared spectroscopy (TG-FTIR) and headspace gas chromatography-mass spectrometry (HS-GC-MS). The results showed that the heating of most incenses occurred below 200 °C, and the mass-loss rate value of CGK was between those of WK and CCA. The HS-GC-MS analysis showed the chemical compounds of incense smoke of CGK at 40, 100, and 180 °C, corresponding to the head, middle, and tail of the heating process, respectively. The results suggested that the sesquiterpenes compounds were the major contributors to the mysterious and elegant odoriferous character of agarwood incense. However, a peak area percentage analysis revealed a significant difference in the predominant compounds between CGK and WK, especially at lower temperatures. Therefore, it is not straightforward to substitute WK with CGK. The results are helpful for the study and usage of the new cultivated grafting Kynam agarwood and the development of the agarwood incense industry. Alzheimer's disease (AD) is a common and severe brain disease with a high mortality among the elders, but no highly efficient medications are currently available. For example, timosaponin BII, an efficient anti-AD agent, has low oral bioavailability. Here, timosaponin BII was formulated in a temperature/ion-sensitive in situ hydrogel (ISG) that was well transformed into gels in the nasal environment. Timosaponin BII protected the PC12 cells injured by lipopolysaccharides (LPS) by decreasing TNF-α and IL-1β and stabilizing F-actin. Timosaponin BII ISGs were intranasally administered to the mice every day for 38 days. On Day 36, LPS was injected to the mice to establish an AD model. Morris water maze experiments showed that the number of the animals that were able to cross the platform returned to normal and the total distance over which the animals moved in the open field also increased, which demonstrated that the spatial memory and spontaneous behavior were improved after treatment compared to the model. Moreover, an AD improver, inducible nitric oxide synthase (iNOS) in the brain, was reduced after treatment. High brain targeting effect of timosaponin BII ISGs was confirmed by in vivo fluorescence imaging. The nasal timosaponin BII dually sensitive ISGs can serve as a promising medication for local prevention of AD. In order to improve the immunogenicity of the highly purified vaccine antigens, addition of an adjuvant to formulation, without affecting the safety of the vaccine, has been the key aim of the vaccine formulators. In recent years, adjuvants which are composed of a delivery system and immunopotentiators have been preferred to induce potent immune responses. In this study, we have combined Salmonella Typhi porins and chitosan to develop a new adjuvant system to enhance the immunogenicity of the highly purified antigens. Cationic gels, microparticle (1.69 ± 0.01 μm) and nanoparticles (337.7 ± 1.7 nm) based on chitosan were prepared with high loading efficiency of porins. Cellular uptake was examined by confocal laser scanning microscopy, and the macrophage activation was investigated by measuring the surface marker as well as the cytokine release in vitro in J774A.1 macrophage murine cells. Porins alone were not taken up by the macrophage cells whereas in combination with chitosan a significant uptake was obtained. Porins-chitosan combination systems were found to induce CD80, CD86 and MHC-II expressions at different levels by different formulations depending on the particle size. Similarly, TNF-α and IL-6 levels were found to increase with porins-chitosan combination. Our results demonstrated that combination of porins with chitosan as a particulate system exerts enhanced adjuvant effect, suggesting a promising adjuvant system for subunit vaccines with combined immunostimulating activity. PURPOSE The aim of the research was to assess the quality of obtained information from YouTube videos related to dental implant. MATERIALS AND METHODS "Dental implant" word was used as a key word in YouTube videos in this study. Videos were evaluated by three oral and maxillofacial surgeons. The first 150 videos were screened. After exclusion criterias, seventy-four videos were investigated regard to content's usefulness. The usefulness scores of the videos were poor (0), moderate (1) and excellent (2). Upload sources were recorded that were categorized as the individual users, health care professionals, and health companies. The type of videos was classified 3 sections educational videos, patient's experience, and scientifically erroneous. We examined videos for the content presence in eight non-mutually exclusive domains of dental implant. RESULTS Seventy-four videos were evaluated. The usefulness score of the videos ranged from 0-2 and the average was 0.324. The most discussed topics were regarding with procedures of dental implant (42.57%), however, the least one was related to contraindications (1%). No significant correlation was found between the source of upload and video usefulness score (P=0.086). There was a significant difference between number of comments and the video types. (P=0.002). Many issues regarding dental implant were ignored in these videos. PURPOSE The purpose of this study was to evaluate long-term three-dimensional graft resorption following reconstruction of the severely atrophic maxilla with anterior iliac crest bone grafting. METHODS Twenty-two patients (13 males), who underwent autogenous bone grafting and implant placement to their severely atrophic maxillary alveolar ridges were identified and included in the study. Pre- and postoperative cone-beam computed tomography (CBCT) scans of 40 recipient grafting sites were evaluated to calculate volumetric changes over time. CBCT scans were performed preoperatively (V0) and one week (V1), three months (V2), one year (V3), and three years (V4) following the augmentation operation. RESULTS The average graft resorption from V1 to V2, V1 to V3, and V1 to V4 was 31.42%, 33.96%, and 37.96%, respectively. Initial graft volume reduction within the first three months was statistically higher compared to other postoperative periods (P0.05). CONCLUSION The overall success rate of the iliac bone block grafts was found to be high. The volumetric resorption rates associated with the graft were favourable for the reconstruction of the maxilla and for permitting the placement of dental implants three months after augmentation. The highest graft resorption was found at the third postoperative month. Placement and loading of the implants reduced the resorption rate slightly over time. The present study unravels origin of nitric oxide (NO) and the interaction between 24-Epibrassinolide (EBL) and nitrate reductase (NR) for NO production in Indian mustard (Brassica juncea L.) under salinity stress. Two independent experiments were performed to check whether (i) Nitrate reductase or Nitric oxide synthase takes part in the biosynthesis of endogenous NO and (ii) EBL has any regulatory effect on NR-dependent NO biosynthesis in the alleviation of salinity stress. Results revealed that NR-inhibitor tungstate significantly (P ≤ 0.05) decreased the NR activity and endogenous NO content, while NOS inhibitor l-NAME did not influence NO biosynthesis and plant growth. Under salinity stress, inhibition in NR activity decreased the activities of antioxidant enzymes, increased H2O2, MDA, protein carbonyl content and caused DNA damage, implying that antioxidant defense might be related to NO signal. EBL supplementation enhanced the NR activity but did not influence NOS activity, suggesting that NR was involved in endogenous NO production. EBL supplementation alleviated the inhibitory effects of salinity stress and improved the plant growth by enhancing nutrients, photosynthetic pigments, compatible osmolytes, and performance of AsA-GSH cycle. It also decreased the superoxide ion accumulation, leaf epidermal damages, cell death, DNA damage, and ABA content. Comet assay revealed significant (P ≤ 0.05) enhancement in tail length and olive tail moment, while flow cytometry did not showed any significant (P ≤ 0.05) changes in genome size and ploidy level under salinity stress. Moreover, EBL supplementation increased the G6PDH activity and S-nitrosothiol content which further boosted the antioxidant responses under salinity stress. Taken together, these results suggested that NO production in mustard occurred in NR-dependent manner and EBL in association with endogenous NO activates the antioxidant system to counter salinity stress. ETHNOPHARMACOLOGICAL RELEVANCE Orostachys japonicus A. Berger (O. japonicus), so-called Wa-song in Korea, a traditional food and medicine that grows on mountain rocks and roof tiles. Wa-song containing various phenolic compounds have been reported as a medicinal plant for prevention of fibrosis, cancer, inflammation, and oxidative damage. AIM OF THE STUDY The present study was designed to examine the anti-angiogenic effects of cultivated Orostachys japonicus 70% ethanol extract (CE) in vascular endothelial growth factor (VEGF)-stimulated human umbilical vein endothelial cells (HUVECs). MATERIALS AND METHODS CE was prepared with 70% ethanol. HUVECs, rat aortic rings, and matrigel plug in mice were treated with CE (10-20 μg/mL) and VEGF (20-50 ng/mL), and the anti-angiogenic activities of CE were analyzed by SRB, wound healing, trans-well invasion, capillary-like tubule formation, rat aortas, Western blot, and matrigel plug assay. Phenolic compounds in CE were analyzed using a high-performance liquid chromatography (HPLC)-PDA system.
