In shake flask cultures, the highest PHBV yield from sucrose (0.16 g g-1) and 3-hydroxyvalerate (3HV) fraction in the PHA chain (27.4 mol%) were obtained with valerate (1.0 g L-1). Epigenetics inhibitor In the bioreactor, the cultures were grown under oxygen-limited conditions, and the maximum OTR (OTRmax) was varied by adjusting the agitation rate. In the cultures grown at low OTRmax (4.3 mmol L-1 h-1), the intracellular content of PHBV (73% w w-1) was improved, whereas a maximum 3HV fraction (35 mol %) was obtained when a higher OTRmax (17.2 mmol L-1 h-1, to 600 rpm) was employed. The findings obtained suggest that the PHBV production and the content of 3HV incorporated into the polymer were affected by the OTR. In this study, we compiled a dataset of soil animal communities for which the abundance and body mass of multiple soil animal groups were recorded. The mass-abundance scaling relationships were then used to investigate multiple environmental controls on soil animal community composition. The data reveal latitudinal shifts from high abundances of small soil animals at high latitudes to greater relative abundances of large soil animals at low latitudes. A hierarchical linear mixed effects model was applied to reveal the environmental variables shaping these latitudinal trends. The final hierarchical model identified mean annual temperature, soil pH and soil organic carbon content as key environmental controls explaining global mass-abundance scaling relationships in soil animal communities (R2c = 0.828, Ngroup = 117). Such relationships between soil biota with climate and edaphic conditions have been previously identified for soil microbial, but not soil animal, communities at a global scale. More comprehensive global soil community datasets are needed to better understand the generality of these relationships over a broader range of global ecosystems and soil animal groups.Extensive branching creates the complex dendritic arbor of mammalian CNS neurons but capturing the complete process of branch formation with time-lapse recordings has been challenging. Here, we report that application of BMP7 to cultured hippocampal neurons accelerated dendritic growth sufficiently to document branches forming in less than 20 h via frequent time-lapse imaging (10-min intervals). In these recordings, most branches emerged as collateral sprouts from the shaft of a parent branch. Analysis of the recordings showed that filopodia were abundant and formed transiently throughout the length of dendrites but among these, only a small subset occurred at sites where branches later emerged. Conversely, formation of lamellipodia was rare and coincided with sites where collateral branches emerged. This pattern suggests that lamellipodial structures act as an important intermediate form of cytoskeletal remodeling related to a cellular commitment to branch, whereas filopodia appear to be related to events prior to such commitment.
