We found that splenic γδ+ CD3+ cells were rapidly expanded (10-14 days post infection), which was accompanied by an early γδ T cell infiltration into the heart. In the following days, intracardiac parasitism was reduced, the protective immunity being accompanied by decreased γδ T cells tissue infiltration. As predicted, there was a drastic reduction of γδ T cells in Myd88- and Il18r1-deficient mice, both transgenic strains displaying a susceptible phenotype with increased intracardiac parasitism. In vivo and in vitro assays confirmed that IL-18R deficiency hampered γδ T cell proliferation. Further characterization revealed that T. cruzi infection up-regulates IL-18R expression in WT γδ+ T cell population whereas Il18r1-/- mice showed impaired generation of cytotoxic GzB+ and IFN-γ-producing γδ T cells. We aimed to assess sleep quality and its relationship to mood, general cognition, and sociodemographic factors in a sample of cognitively active older adults. Methods We assessed 105 non-clinical older adults (mean age ± SD 69.64 ± 0.66 years) based on a sociodemographic profile questionnaire, the Beck Anxiety Inventory, the Beck Depression Inventory II, the Montreal Cognitive Assessment, the Pittsburgh Sleep Quality Index, and the Epworth Sleepiness Scale. Separate analyses were conducted, controlled by sleep quality and daytime sleepiness, to understand how variables were associated. Results About 46.7% of individuals had significantly poor sleep quality. Univariate analysis showed that non-workers had a lower risk of impaired sleep quality (prevalence ratio (PR) = 0.67; P = 0.044). However, there was an increased risk of poor sleep quality in those experiencing depressive symptoms (PR = 1.78; P less then 0.001) and anxiety symptoms (PR = 1.98; P less then 0.001). To investigate whether S. aureus viability influences EV biogenesis, EVs were isolated from human neutrophils incubated with viable bacteria (bEVs) or heat-killed bacteria (heat-killed EVs). Protein analysis, nanoparticle tracking and transmission electron microscopy showed comparable EV production between subsets, and both viable and nonviable bacteria were also detected in respective EV subsets. As anticipated, S. aureus, as well as bEVs with viable bacteria, were proinflammatory, and killing bacteria with gentamicin reduced cytokine production to baseline levels. Although heat-killed bacteria induced macrophage IL-6 production, heat-killed EVs did not. Additionally, we found that human and bacterial DNA associated with bEVs, but not heat-killed EVs, and that the DNA association could be partially decreased by disrupting electrostatic interactions.
