Norovirus is the leading cause of acute gastroenteritis worldwide. The pathogenesis of norovirus and the induced immune response remain poorly understood due to the lack of a robust virus culture system. Selleck Inavolisib The monolayers of two secretor-positive Chinese human intestinal enteroid (HIE) lines were challenged with two norovirus pandemic GII.4 Sydney strains. Norovirus RNA replication in supernatants and cell lysates were quantified by RT-qPCR. RNA expression levels of immune-related genes were profiled using PCR arrays. The secreted protein levels of shortlisted upregulated genes were measured in supernatants using analyte-specific enzyme-linked immunosorbent assay (ELISA). Productive norovirus replications were achieved in three (75%) out of four inoculations. The two most upregulated immune-related genes were CXCL10 (93-folds) and IFI44L (580-folds). Selleck Inavolisib Gene expressions of CXCL10 and IFI44L were positively correlated with the level of norovirus RNA replication (CXCL10 Spearman's r = 0.779, p less then 0.05; IFI44L r = 0.881, p less then 0.01). The higher level of secreted CXCL10 and IFI44L proteins confirmed their elevated gene expression. The two genes have been reported to be upregulated in norovirus volunteer challenges and natural human infections by other viruses. Our data suggested that HIE could mimic the innate immune response elicited in natural norovirus infection and, therefore, could serve as an experimental model for future virus-host interaction and antiviral studies.Chikungunya virus is an Alphavirus belonging to the family Togaviridae that is transmitted to humans by an infected Aedes mosquito. Patients develop fever, inflammatory arthritis, and rash during the acute stage of infection. Although the illness is self-limiting, atypical and severe cases are not uncommon, and 60% may develop chronic symptoms that persist for months or even for longer durations. link2 Having a distinct periodical epidemiologic outbreak pattern, chikungunya virus reappeared in Thailand in December 2018. Here, we describe a cohort of acute chikungunya patients who had presented to the Bangkok Hospital for Tropical Diseases during October 2019. Infection was detected by a novel antigen kit and subsequently confirmed by real-time RT-PCR using serum collected at presentation to the Fever Clinic. Other possible acute febrile illnesses such as influenza, dengue, and malaria were excluded. We explored the sequence of clinical manifestations at presentation during the acute phase and associated the viral load with the clinical findings. Most of the patients were healthy individuals in their forties. Fever and arthralgia were the predominant clinical manifestations found in this patient cohort, with a small proportion of patients with systemic symptoms. Higher viral loads were associated with arthralgia, and arthralgia with the involvement of the large joints was more common in female patients.Recently, the application of endophytes in the alleviation of different types of stresses has received considerable attention, but their role in drought stress alleviation and growth promotion in soybean is not well-stated. In this study, twenty bacterial endophytes were isolated from soybean root tissues and screened for plant growth-promoting (PGP) traits, biocontrol potential, and drought stress alleviation. Out of them, 80% showed PGP traits, and 20% showed antagonistic activity against Fusarium oxysporum (ITCC 2389), Macrophomina phaseolina (ITCC 1800), and Alternaria alternata (ITCC 3467), and only three of them showed drought tolerance up to 15% (-0.3 MPa). link2 Results indicated that drought-tolerant PGP endophytic bacteria enhanced soybean seedling growth under drought stress conditions. Morphological, biochemical, and molecular characterization (16S rRNA) revealed that these three bacterial isolates, AKAD A1-1, AKAD A1-2, and AKAD A1-16, closely resemble Bacillus cereus (GenBank accession No. MN079048), ation of bacterial root endophytes as a potential tool to mitigate the effect of drought as well as of fungal diseases on the early seedling growth of soybean. The expanding scope of chemical reactions applied to nucleic acids has diversified the design of nucleic acid-based technologies that are essential to medicinal chemistry and chemical biology. Among chemical reactions, visible light photochemical reaction is considered a promising tool that can be used for the manipulations of nucleic acids owing to its advantages, such as mild reaction conditions and ease of the reaction process. Of late, inspired by the development of visible light-absorbing molecules and photocatalysts, visible light-driven photochemical reactions have been used to conduct various molecular manipulations, such as the cleavage or ligation of nucleic acids and other molecules as well as the synthesis of functional molecules. Selleck Inavolisib link3 In this review, we describe the recent developments (from 2010) in visible light photochemical reactions involving nucleic acids and their applications in the design of nucleic acid-based technologies including DNA photocleaving, DNA photoligation, nucleic acid sensors, the release of functional molecules, and DNA-encoded libraries.Underwater acoustic sensor networks (UWASNs) can revolutionize the subsea domain by enabling low-cost monitoring of subsea assets and the marine environment. Accurate localization of the UWASNs is essential for these applications. link2 In general, range-based localization techniques are preferred for their high accuracy in estimated locations. However, they can be severely affected by variable sound speed, multipath spreading, and other effects of the acoustic channel. In addition, an inefficient localization scheme can consume a significant amount of energy, reducing the effective life of the battery-powered sensor nodes. In this paper, we propose robust, efficient, and practically implementable localization schemes for static UWASNs. The proposed schemes are based on the Time-Difference-of-Arrival (TDoA) measurements and the nodes are localized passively, i.e., by just listening to beacon signals from multiple anchors, thus saving both the channel bandwidth and energy. The robustness in location estimates is achieved by considering an appropriate statistical noise model based on a plausible acoustic channel model and certain practical assumptions. To overcome the practical challenges of deploying and maintaining multiple permanent anchors for TDoA measurements, we propose practical schemes of using a single or multiple surface vehicles as virtual anchors. The robustness of localization is evaluated by simulations under realistic settings. By combining a mobile anchor(s) scheme with a robust estimator, this paper presents a complete package of efficient, robust, and practically usable localization schemes for low-cost UWASNs.Considering the huge economic loss caused by postharvest diseases, the identification and prevention of citrus postharvest diseases is vital to the citrus industry. In 2018, 16 decayed citrus fruit from four citrus varieties-Satsuma mandarin (Citrus unshiu), Ponkan (Citrus reticulata Blanco cv. Ponkan), Nanfeng mandarin (Citrus reticulata cv. nanfengmiju), and Sugar orange (Citrus reticulata Blanco)-showing soft rot and sogginess on their surfaces and covered with white mycelia were collected from storage rooms in seven provinces. The pathogens were isolated and the pathogenicity of the isolates was tested. The fungal strains were identified as Lasiodiplodia pseudotheobromae based on their morphological characteristics and phylogenetic analyses using the internal transcribed spacer regions (ITS), translation elongation factor 1-α gene (TEF), and beta-tubulin (TUB) gene sequences. The strains could infect wounded citrus fruit and cause decay within two days post inoculation, but could not infect unwounded fruit. To our knowledge, this is the first report of citrus fruit decay caused by L. pseudotheobromae in China.Previously, studies reported that falls analysis is possible in the elderly, when using wearable sensors. However, these devices cannot be worn daily, as they need to be removed and recharged from time-to-time due to their energy consumption, data transfer, attachment to the body, etc. This study proposes to introduce a radar sensor, an unobtrusive technology, for risk of falling analysis and combine its performance with an instrumented insole. We evaluated our methods on datasets acquired during a Timed Up and Go (TUG) test where a stride length (SL) was computed by the insole using three approaches. Only the SL from the third approach was not statistically significant (p = 0.2083 > 0.05) compared to the one provided by the radar, revealing the importance of a sensor location on human body. While reducing the number of force sensors (FSR), the risk scores using an insole containing three FSRs and y-axis of acceleration were not significantly different (p > 0.05) compared to the combination of a single radar and two FSRs. We concluded that contactless TUG testing is feasible, and by supplementing the instrumented insole to the radar, more precise information could be available for the professionals to make accurate decision.Musculoskeletal injuries (MSI) continue to affect Thoroughbred racehorses internationally. There is a strong interest in developing training and management strategies to reduce their impact, however, studies of risk factors report inconsistent findings. link3 Furthermore, many injuries and fatalities occur during training rather than during racing, yet most studies report racing data only. By combining racing and training data a larger exposure to risk factors and a larger number of musculoskeletal injuries are captured and the true effect of risk factors may be more accurately represented. Furthermore, modifications to reduce the impact of MSI are more readily implemented at the training level. Our study aimed to (1) determine the risk factors for musculoskeletal injuries and whether these are different for two-year-old and older horses and (2) determine whether risk factors vary with type of injury. link3 This was performed by repeating analyses by age category and injury type. Data from 202 cases and 202 matched contrare perceived at a higher risk, than resting the horse altogether. Early identification of horses at increased risk and appropriate intervention could substantially reduce the impact of musculoskeletal injuries in Thoroughbred racehorses. In this work, an omnidirectional sensor that enables identification of the direction of the celestial sphere with maximum solar irradiance is presented. The sensor, based on instantaneous measurements, functions as a position server for dual-axis solar trackers in photovoltaic plants. The proposed device has been developed with free software and hardware, which makes it a pioneering solution because it is open and accessible as well as capable of being improved by the scientific community, thereby contributing to the rapid advancement of technology. In addition, the device includes an algorithm developed ex professo that makes it possible to predetermine the regions of the celestial sphere for which, according to the geometric characteristics of the PV plant, there would be shading between the panels. In this way, solar trackers do not have to locate the Sun's position at all times according to astronomical models, while taking into account factors such as shadows or cloudiness that also affect levels of incident irradiance on solar collectors.
