Tetracyclines can inhibit microglial reactivity and neuroinflammation by inhibiting nuclear factor kappa B (NF-kB) signaling, cyclooxygenase 2, and matrix metalloproteinases (MMPs). This drug class also has a broad profile of activity against bacteria associated with community-based pneumonia, including atypical agents. Despite the achievements of these delivery approaches, the development of a product with the ability of delivering drug molecules at a specific site and according to patients' needs remains a challenging endeavour. The complexity of the physicochemical properties of colloidal systems, their stability in different regions of the gastrointestinal tract, and interaction with the restrictive biological barriers hampered their success for oral precise medicine. To overcome these issues, nanoparticles have been combined with polymers to create hybrid nanosystems, namely nanocomposites. They offer enormous possibilities of structural and mechanical modifications to both nanoparticles and polymeric matrixes to generate systems with new properties, functions, and applications for oral delivery. In this review, nanocomposites' physicochemical and functional properties intended to target specific regions of the GI tract-oral cavity, stomach, small bowel, and colon-are analysed. In parallel, it is provided an insight in the nanocomposite solutions for oral delivery intended for systemic and local absorption, together with a focus on inflammatory bowel diseases (IBDs). Additional difficulties in managing IBD related to the alteration in the physiology of the intestine are described. Finally, future perspectives and opportunities for advancement in this field are discussed.Cutaneous leishmaniasis (CL), the most common clinical form of human leishmaniasis, is a non-fatal chronic and disabling disease characterized by erythema and nodular or ulcerative skin lesions that may cause permanent scars and disfigurement. Topical drug delivery represents a simple and efficacious approach to treat CL skin lesions. The association of drugs with nanocarrier systems enhances their permeation properties and increases the drug amount available in the dermis. Here, a deformable lipid vesicle (DLV) was optimized for the topical administration of Amphotericin B (AmB), with the aim of studying and understanding the advantages of this type of delivery system in the transport of a drug through the skin layers. AmB-DVL were characterized in terms of incorporation parameters, stability, and elasticity, and evaluated in vitro for their permeation properties, cytotoxicity, and anti-leishmanial activity. The AmB-DVL exhibited a translucent fluid gel-like aspect and a yellow color, a mean size of 132 nm (PdI ≤ 0.1), zeta potential values around zero (mV), and an AmB incorporation efficiency of 95%. Permeation and penetration assays suggest that AmB-DLV are suitable for topical administration since AmB was detected in the epidermal and dermal skin layers. AmB-DVL was able to reduce promastigote viability in a dose-dependent manner, as well as the number of intracellular amastigotes in THP-1 macrophages. Selectivity index (SI) value for AmB-DLV was considerably higher than that observed for free AmB. Results suggest that DLV may represent an attractive vehicle for dermal delivery of AmB and a new low-cost and safe therapeutic option in CL treatment.The question of pain prevention is becoming increasingly important, both in society and in science. According to the International Society for the Study of Pain, general areas for which pain prevention measures can be recommended have been defined. These approaches are mostly limited to unspecific recommendations with the aim of improving general health behaviour. Common to all of them is that they essentially address psychosocial and psychobehavioral aspects. In contrast to genetic factors or other non-modifiable environmental factors, psychosocial and psychobehavioral aspects are potentially modifiable variables, making them possible starting points for prevention programs. Furthermore, recent studies provide important knowledge about psychological and social risk factors of pain chronification and thus offer new approaches for future pain prevention strategies. At the same time, the efficacy and successful implementation of such programs is so complex that valid statements on effectiveness and benefit can only be made through care-related evaluation. This review addresses psychological and social factors in the prevention of pain. A selective literature search was carried out to this end. Based on selected studies, psychological and social predictors of pain development are presented and their potential for future pain prevention programs discussed. The article concludes with a discussion of possible implications.Accumulation of heavy metals in zooplanktons is used as an essential tool for the evaluation of health of an ecosystem. Such data are used to set further monitoring strategies especially in (coastal) water bodies. In the present study, seasonal bioaccumulation of heavy metals such as Cu, Zn, Pb, Cd and Hg, organic loads such as nitrite and phosphate, contents loads and physicochemical parameters in Haripur creek along the Bay of Bengal in east coast of India is measured in zooplanktons. Coastal organisms are considered an integral part of marine ecosystems and their frequent massive die-off events along the coast of the Bay of Bengal are correlated with the obtained pollutant data. The levels of heavy metals were also correlated with several extrinsic environmental factors such as water transparency, dissolved oxygen, salinity, pH and temperature. Discriminant function analyses and standardised coefficients for canonical variables for the obtained data indicate that the studied environmental factors and organic loads varied as a function of season. Bioaccumulation of the studied heavy metals in zooplanktons was seasonal and infrequently noticed above the standard limit. The computed bioaccumulation factor of the five metals showed that accumulation of Zn was higher in zooplanktons as compared to that of the other studied metals. So, the entry of heavy metals through the food chains or via direct exposure (to organic loads also) to the other inhabiting organisms including fish may be assumed. This could be one of the possible reasons for the observed frequent (fish) die-off events at Gopalpur Coast along the Bay of Bengal and may be considered a clue for future investigations.Vetiver has a broad history of traditional medicinal uses, but only a handful of research article has reported its utility in treating diseases. But unfortunately, no work has been reported on the anti-inflammatory activity of its plant extract and inflammatory-linked diseases. Hence, the present review focuses on investigating the several presumptions which can be put forward to explain its anti-inflammatory property. Amalgamating all the factors together, the plant can be utilized as anti-inflammatory can be and also can be proved as a beneficial perspective in the treatment of inflammation-linked disorders.The study describes the loading of the quartz SiO2 nanoparticles (NPs) with (3-aminopropyl)triethoxysilane (APTES) linker with simultaneous lengthening of the linker through the terminal amine group by glutaraldehyde (GA). The reactive polyethylenimine (PEI) was introduced to the surface to increase the ability to capture Cu(II) ions. The composite got the abbreviation SiO2/PEI-Cu(II). The Cu(II) ions were the active center with a peroxo-complex activation state. The composite characterization included scanning electron microscopy (SEM), transmission electron microscopy (TEM), electron-dispersive X-ray analysis (EDX), Fourier transform infrared spectroscopy (FT-IR), X-ray powder diffraction (XRD), thermogravimetric analysis (TGA), and Brunauer-Emmett-Teller (BET) surface analyzer. The kinetics of the oxidative degradation of Rhodamine B (RhB) dye obeyed the pseudo-first order under flooding conditions. The reaction parameters including the catalyst dose, solution pH, initial concentration of reactants, and temperature got some attention. The obtained results showed that more than 91.7 ± 1% of RhB dye was degraded to CO2, NH4+, NO3-, H2O, and some inorganic acids after 30 min as confirmed by gas chromatography mass spectrometry and total organic carbon (TOC) measurements. Also, GC-MS spectra for water samples drawn from the reaction in successive periods had suggested a conceivable degradation pathway for RhB by hydroxyl radicals. Degradation starts with de-alkylation then carboxyphenyl removal followed by two successive ring-opening stages. Both the effects of the catalyst recycling and treated water reusability on the reaction rate were studied. The catalyst provided noticeable stability over three consecutive cycles.
