Metallothionein concentrations in exposed earthworms were not statistically different from levels in untreated earthworms. Finally, the short-term toxicokinetic models predicted the bioaccumulation in earthworms exposed to Ag-NP, AgNO3 after nine months on the whole. Although the bioaccumulation for Ag2S-NPs was somewhat under-predicted, the rate of accumulation of Ag2S-NPs is much lower than that of Ag-NPs or AgNO3 and thus potentially of lower concern. Nevertheless, better understanding about the exposure kinetics of Ag2S-NP would help to address potential nano-specific toxicokinetic and toxicodynamics, also of other sulfidized metal NPs.Zearalenone, which is ubiquitous in grains and animal feed, is a mycotoxin that can cause serious damage to animals and humans. Sertoli cells (SCs) can be used to study ZEA male reproductive toxicity in vitro. SCs provide energy for germ cells, where AMPK regulates intracellular energy. In order to explore the regulatory effect of AMPK on ZEA-induced lactate decline, we activated AMPK by AICAR and then inhibited AMPK by Compound C with ZEA-treated SCs for 24 h to detect intracellular lactate production-related indicators. Cell viability in the presence of 20 μmol/L ZEA and either 50 μmol/L AICAR or 5 μmol/L Compound C, respectively, did not damage SCs, and could effectively either activate or inhibit AMPK. Inhibition of AMPK promoted the production of pyruvate and lactate via increased expression of the glycolysis-related genes Pgam1 and the lactate production-related proteins GLUT1, LDHA, and MCT4. Activating AMPK inhibited the production of lactate and pyruvate by suppressing the expression of glycolysis-related genes HK1, Pgam1, and Gpi1 and that of lactate production-related proteins LDHA and MCT4. Zearalenone destroys the energy balance in SCs, activates P-AMPK, which inhibit the production of lactate and pyruvate in SCs. This also leads to the decrease of energy supply of SCs to spermatogenic cells, damages to reproductive system.Exposure to ethephon (ETH), a plant growth regulator commonly used for several purposes, can potentially decrease sperm numbers and viability. Occasional findings regarding ETH effects on female reproduction during early pregnancy have also been reported. During early pregnancy, endometrial decidualization is a critical event for embryo implantation and pregnancy maintenance. Thus, we aimed to explore the effect and mechanism of ETH on endometrial decidualization both in vivo and in vitro. Mice were gavaged with 0 and 285 mg/kg b.w. ETH from gestational days (GD)1 until sacrifice, whereas pseudopregnant mice from pseudopregnant day 1 (PPD-1) until PPD-8. Primary mouse endometrial stromal cells (mESCs) received 640 ug/ml ETH and added E2 and P4 to induce decidualization. Results indicated female albino CD1 mice exposed to high dose of ETH (285 mg/kg b.w.) by oral gavage, the number of embryo implantation sites on GD6 and GD8 were significantly decreased, the levels of serum E2 and P4 on GD8 were significantly R120 and ETH are crucial for impaired decidualization mediated via ETH.Heavy metals, like many other chemical elements, are naturally present in the environment; however, the concentrations of these metals in various environmental matrices have increased through their intensive use in many human activities (such as industry, mining and agriculture). Among the heavy metals, cadmium (Cd) and mercury (Hg) induce a wide variety of defects in animals. While the effects of these heavy metals have been widely documented, a single exposure paradigm is typically used. Few studies have focused on evaluating combined exposure to these metals. However, in the environment, animals are confronted with a plethora of substances simultaneously; thus, the presence and origin of such substances must be determined to reduce the sources of contamination. Using the model of the fruit fly Drosophila melanogaster, for which many tools are readily available, we investigated how different concentrations of Cd and Hg in single and combined exposures impact post-embryonic development. In parallel, we evaluated the extended expression pattern of 38 molecular targets used as potential biomarkers of exposure through qPCR. Our results showed that both metals caused developmental delays and mortality in dose-dependent responses. Both metals were able to deregulate genes involved in hormonal control, general stress, and oxidative stress. Importantly, we confirmed synergistic interactions between Cd and Hg. Our results indicate the importance of assessing several biomarkers and their kinetics in mixtures. Drosophila represents a useful model for monitoring the toxicity of substances in polluted environments.Triclosan (TCS), a ubiquitous antimicrobial agent, has been frequently detected in wild fish, leading to concerns regarding TCS safety in the aquatic environment. The present work aims to investigate the TCS-mediated effects on various tissues (the liver, gills, brain, and testes) of wild-sourced adult mosquitofish based on histological analysis and transcriptome. Severe morphological injuries were only found in the liver and gills. VO-Ohpic research buy The histopathological alterations in the liver were characterized by cytoplasmic vacuolation and degeneration, eosinophilic cytoplasmic inclusions, and nuclear polymorphism. The gill lesions contained epithelial lifting, intraepithelial edema, fusion and shortening of the secondary lamellae. Consistently, the numbers of differently expressed genes (DEGs) identified by transcriptome were in the order of liver (1627) > gills (182) > brain (9) > testes (4). Trend-aligned histopathological and transcriptomic changes in the 4 tissues, suggesting the tissue-specific response manner of mosquitofish to TCS, and the liver and gills were the target organs. TCS interrupted many biological pathways associated with lipogenesis and lipid metabolism, transmembrane transporters, protein synthesis, and carbohydrate metabolism in the liver, and it induced nonspecific immune response in the gills. TCS-triggered hepatotoxicity and gills damnification may lead to inflammation, apoptosis, diseases, and even death in mosquitofish. TCS showed moderate acute toxicity and bioaccumulative property on mosquitofish, suggesting that prolonged or massive use of TCS may pose an ecological risk.
