aureus and other pathogenic microorganisms can provide possibilities for the development of new, more effective treatments for bacterial infections.Polycyclic aromatic hydrocarbons (PAHs) are contaminants introduced by different pathways in the marine ecosystem, affecting both aquatic and sediment bodies. Identification of their sources is of vital importance for protecting the marine ecosystem. The attribution of the pollution sources is usually made by using diagnostic molecular ratios of PAHs isomers. The reliability of this approach diminishes when PAHs contents are measured far from their original source, for example in water bodies or in bottom sediments. Conventionally the source attribution is based on time consuming univariate methods. In the present work coupling of molecular ratios with advanced supervised statistical techniques was used to increase the accuracy of the PAH source attribution in bottom sediments. Data on PAHs distribution within 5 port areas, with known pattern port activity, were collected. Evaluation of multiple PAHs ratios at once by means of supervised OPLS-DA technique was performed. A robust descriptive and predictive model was set up and successfully validated. The proposed methodology helps identify PAH transport pathways, highlighting interactions between pollution patterns, port activities and coastal land-use supporting decision makers in defining monitoring and mitigation procedures.Delivery vehicle drivers are experiencing increasing challenges in finding available curb space to park in urban areas, which increases instances of cruising for parking and parking in unauthorized spaces. Policies traditionally used to reduce cruising for parking for passenger vehicles, such as parking fees and congestion pricing, are not effective at changing delivery drivers' travel and parking behaviors. Intelligent parking systems that use real-time curb availability information to better route and park vehicles can reduce cruising for parking, but they have never been tested for delivery vehicle drivers. The current study tested whether providing real-time curb availability information to delivery drivers reduces the travel time and distance spent cruising for parking. A curb parking information system deployed in a study area in Seattle, Wash., displayed real-time curb availabilities on a mobile app called OpenPark. A controlled experiment assigned drivers' deliveries in the study area with and without access to OpenPark. The data collected showed that when curb availability information was provided to drivers, their cruising for parking time significantly decreased by 27.9 percent, and their cruising distance decreased by 12.4 percent. These results demonstrate the potential for implementing intelligent parking systems to improve the efficiency of urban logistics systems.The common final pathway to blindness in many forms of retinal degeneration is the death of the light-sensitive primary retinal neurons. However, the normally light-insensitive second- and third-order neurons persist optogenetic gene therapy aims to restore sight by rendering such neurons light-sensitive. Here, we investigate whether bReaChES, a newly described high sensitivity Type I opsin with peak sensitivity to long-wavelength visible light, can restore vision in a murine model of severe early-onset retinal degeneration. Intravitreal injection of an adeno-associated viral vector carrying the sequence for bReaChES downstream of the calcium calmodulin kinase IIα promoter resulted in sustained retinal expression of bReaChES. Retinal ganglion cells (RGCs) expressing bReaChES generated action potentials at light levels consistent with bright indoor lighting (from 13.6 log photons cm-2 s-1). They could also detect flicker at up to 50 Hz, which approaches the upper temporal limit of human photopic vision. Topological response maps of bReaChES-expressing RGCs suggest that optogenetically activated RGCs may demonstrate similar topographical responses to RGCs stimulated by photoreceptor activation. Furthermore, treated dystrophic mice displayed restored cortical neuronal activity in response to light and rescued behavioral responses to a looming stimulus that simulated an aerial predator. Finally, human surgical retinal explants exposed to the bReaChES treatment vector demonstrated transduction. Together, these findings suggest that intravitreal gene therapy to deliver bReaChES to the retina may restore vision in human retinal degeneration in vivo at ecologically relevant light levels with spectral and temporal response characteristics approaching those of normal human photopic vision.Dust storms have many negative consequences, and affect all kinds of ecosystems, as well as climate and weather conditions. Therefore, classification of dust storm sources into different susceptibility categories can help us mitigate its negative effects. This study aimed to classify the susceptibility of dust sources in the Middle East (ME) by developing two novel deep learning (DL) hybrid models based on the convolutional neural network-gated recurrent unit (CNN-GRU) model, and the dense layer deep learning-random forest (DLDL-RF) model. The Dragonfly algorithm (DA) was used to identify the critical features controlling dust sources. Game theory was used for the interpretability of the DL model's output. Predictive DL models were constructed by dividing datasets randomly into train (70%) and test (30%) groups, six statistical indicators being then applied to assess the DL hybrid model performance for both datasets (train and test). Among 13 potential features (or variables) controlling dust sources, seven variables were selected as important and six as non-important by DA, respectively. Based on the DLDL-RF hybrid model - a model with higher accuracy in comparison with CNN-GRU-23.1, 22.8, and 22.2% of the study area were classified as being of very low, low and moderate susceptibility, whereas 20.2 and 11.7% of the area were classified as representing high and very high susceptibility classes, respectively. Among seven important features selected by DA, clay content, silt content, and precipitation were identified as the three most important by game theory through permutation values. Overall, DL hybrid models were found to be efficient methods for prediction purposes on large spatial scales with no or incomplete datasets from ground-based measurements.Firn (compressed snow) covers approximately 90[Formula see text] of the Greenland ice sheet (GrIS) and currently retains about half of rain and meltwater through refreezing, reducing runoff and subsequent mass loss. The loss of firn could mark a tipping point for sustained GrIS mass loss, since decades to centuries of cold summers would be required to rebuild the firn buffer. Here we estimate the warming required for GrIS firn to reach peak refreezing, using 51 climate simulations statistically downscaled to 1 km resolution, that project the long-term firn layer evolution under multiple emission scenarios (1850-2300). We predict that refreezing stabilises under low warming scenarios, whereas under extreme warming, refreezing could peak and permanently decline starting in southwest Greenland by 2100, and further expanding GrIS-wide in the early 22[Formula see text] century. After passing this peak, the GrIS contribution to global sea level rise would increase over twenty-fold compared to the last three decades.Treadmill training (TT) has been extensively used as an intervention to improve gait and mobility in patients with Parkinson's disease (PD). Regional and global effects on brain activity could be induced through TT. Training effects can lead to a beneficial shift of interregional connectivity towards a physiological range. The current work investigates the effects of TT on brain activity and connectivity during walking and at rest by using both functional near-infrared spectroscopy and functional magnetic resonance imaging. Nineteen PD patients (74.0 ± 6.59 years, 13 males, disease duration 10.45 ± 6.83 years) before and after 6 weeks of TT, along with 19 age-matched healthy controls were assessed. Interregional effective connectivity (EC) between cortical and subcortical regions were assessed and its interrelation to prefrontal cortex (PFC) activity. Support vector regression (SVR) on the resting-state ECs was used to predict prefrontal connectivity. In response to TT, EC analysis indicated modifications in the patients with PD towards the level of healthy controls during walking and at rest. SVR revealed cerebellum related connectivity patterns that were associated with the training effect on PFC. click here These findings suggest that the potential therapeutic effect of training on brain activity may be facilitated via changes in compensatory modulation of the cerebellar interregional connectivity.This systematic review and meta-analysis investigated the impact of anti-vascular endothelial growth factor (VEGF) treatment in management of eyes with non-proliferative diabetic retinopathy (NPDR) without centre involving diabetic macular oedema (CI-DMO). We searched multiple databases for all randomised clinical trials (RCTs) that evaluated anti-VEGF treatment versus observation in eyes with NPDR without CI-DMO. Data was collected for six outcomes (best corrected visual acuity (BCVA) improvement, diabetic retinopathy severity score (DRSS), central subfield thickness, progression to vision threatening complications (VTCs), ocular adverse events and quality of life measures). Risk of bias was assessed using Cochrane risk-of-bias tool for randomised trials (RoB 2) and certainty of evidence was assessed using Grade of Recommendations, Assessment, Development and Evaluation (GRADE). We identified a total of 2 unique RCTs that compared aflibercept and sham to treat a total of 811 eyes. For BCVA change, there was a small, clinically insignificant benefit for aflibercept treatment at year 2 (MD 0.70, 95% CI 0.02-1.38, GRADE rating MODERATE). DRSS demonstrated a statistically significant improvement with aflibercept use at year 2 (RR 3.76, 95% CI 2.75-5.13, GRADE rating MODERATE). VTCs were significantly less in aflibercept arm at year 2 (RR 0.30, 95% CI 0.23-0.40, GRADE rating MODERATE). In conclusion, aflibercept treatment versus observation in eyes with NPDR without CI-DMO can result in reduced risk of development of VTCs and regression of DRSS score over 2 years. Future trials are needed to increase the precision of the treatment effect and to provide data on quality-of-life metrics.PROSPERO Registration CRD42021288608.A common issue when analyzing real-world complex systems is that the interactions between their elements often change over time. Here we propose a new modeling approach for time-varying interactions generalising the well-known Kinetic Ising Model, a minimalistic pairwise constant interactions model which has found applications in several scientific disciplines. Keeping arbitrary choices of dynamics to a minimum and seeking information theoretical optimality, the Score-Driven methodology allows to extract from data and interpret the presence of temporal patterns describing time-varying interactions. We identify a parameter whose value at a given time can be directly associated with the local predictability of the dynamics and we introduce a method to dynamically learn its value from the data, without specifying parametrically the system's dynamics. We extend our framework to disentangle different sources (e.g. endogenous vs exogenous) of predictability in real time, and show how our methodology applies to a variety of complex systems such as financial markets, temporal (social) networks, and neuronal populations.
