No treatment has been proved to be efficient in case of early or advanced SSc to prevent or reduce fibrosis. There are strong arguments for a key role of topo-I in the pathogenesis of diffuse SSc. Irinotecan, a semisynthetic derivative of Camptothecin, specifically target topo-I. This study was undertaken to evaluate the effects of noncytotoxic doses of irinotecan or its active metabolite SN38 on collagen production in SSc fibroblasts. Dermal fibroblasts from 4 patients with SSc and 2 healthy donors were cultured in the presence or absence of irinotecan or SN38. Procollagen I release was determined by ELISA and expression of a panel of genes involved in fibrosis was evaluated by qRT-PCR. Subcytotoxic doses of irinotecan and SN38 caused a significant and dose-dependent decrease of the procollagen I production in dermal fibroblasts from SSc patients, respectively - 48 ± 3%, p  less then  0.0001 and - 37 ± 6.2%, p = 0.0097. Both irinotecan and SN38 led to a global downregulation of genes involved in fibrosis such as COL1A1, COL1A2, MMP1 and ACTA2 in dermal fibroblasts from SSc patients (respectively - 27; - 20.5; - 30.2 and - 30% for irinotecan and - 61; - 55; - 50 and - 54% for SN38). This retrospective study was conducted on 135 Thai hemodialysis (HD) patients with secondary hyperparathyroidism (SHPT). CASR A990G genotypes were determined. The patients were identified as either G carriers (heterozygous or homozygous CASR 990G allele carriers) or noncarriers (homozygous CASR 990A carriers). Tested covariates were baseline PTH level (bPTH), baseline serum phosphate (bPhos), baseline serum calcium (bCa), baseline calcitriol equivalent dose (bCtriol), baseline ergocalciferol dose (bErgo), and age. The ANCOVA showed that intact PTH levels after 12 weeks of cinacalcet treatment (PTHw12) was significantly lower among G carriers compared with noncarriers after controlling for bPTH, bPhos, bCtriol, and bErgo (F(1, 127) = 15.472, p  less then  0.001), with the adjusted mean difference of 253.7 pg/mL. We analyzed age, gender, systolic (SBP), diastolic blood pressure (DBP) and heart rate (HR) by merging three large databases of patients considered likely to be of vasovagal reflex etiology, comparing patients who had tilt-induced reflex response with those who did not. Tilt-induced reflex response was defined as spontaneous symptom reproduction with characteristic hypotension and bradycardia. Relationship of demographics and baseline supine BP to tilt-test were assessed using logistic regression models. Individual records of 5236 patients (45% males; mean age 60 ± 22 years; 32% prescribed antihypertensive therapy) were analyzed. However, quantitative assessments of the transformation and distribution of exogenous carbon (C) in soil aggregates under various field fertilization practices have been lacking. In this study, we collected topsoil samples (0-20 cm) from three fertilization treatments (no fertilization control, CK; inorganic fertilizer, IF; inorganic fertilizer plus manure, IFM) at a 29-year long-term Mollisol experiment in Northeast China. We then mixed the soil samples with 13C-labeled maize straw (δ13C = 246.9‰), referred as CKS, IFS, and IFMS, and incubated them in-situ for 360 days. Initial and incubated soil samples were separated into four aggregate fractions (> 2, 1-2, 0.25-1, and  2 mm and 1-2 mm aggregates, but an increase in the less then  0.25 mm aggregate. At the end of incubation, the relative distribution of straw-derived C showed a decrease in the 0.25-1 mm aggregate but an increase in the less then  0.25 mm aggregate under the IFMS treatment. This study indicates that more straw-derived C would be accumulated in smaller aggregates over longer period in Mollisols, and combined inorganic and organic fertilization is an effective measure for C sequestration in Northeast China.Sepsis is a leading cause of morbidity and mortality worldwide. Early identification of sepsis is important as it allows timely administration of potentially life-saving resuscitation and antimicrobial therapy. We present COMPOSER (COnformal Multidimensional Prediction Of SEpsis Risk), a deep learning model for the early prediction of sepsis, specifically designed to reduce false alarms by detecting unfamiliar patients/situations arising from erroneous data, missingness, distributional shift and data drifts. COMPOSER flags these unfamiliar cases as indeterminate rather than making spurious predictions. Six patient cohorts (515,720 patients) curated from two healthcare systems in the United States across intensive care units (ICU) and emergency departments (ED) were used to train and externally and temporally validate this model. In a sequential prediction setting, COMPOSER achieved a consistently high area under the curve (AUC) (ICU 0.925-0.953; ED 0.938-0.945). Out of over 6 million prediction windows roughly 20% and 8% were identified as indeterminate amongst non-septic and septic patients, respectively. COMPOSER provided early warning within a clinically actionable timeframe (ICU 12.2 [3.2 22.8] and ED 2.1 [0.8 4.5] hours prior to first antibiotics order) across all six cohorts, thus allowing for identification and prioritization of patients at high risk for sepsis.Temporary water bodies, especially vernal pools, are the most sensitive aquatic environments to climate change yet the least studied. Their functioning largely depends on their phytoplankton community structure. This study aimed to determine how temperature and photoperiod length (by simulating inundation in different parts of the year under five climate scenarios) affect the succession and structure of phytoplankton communities soon after inundation. Photoperiod was the most important factor affecting phytoplankton species richness, total abundance and the abundance of taxonomic groups in the course of succession. A long photoperiod (16 h) and a moderate temperature (16 °C) in vernal pool microcosms (late spring inundation after a warm snowless winter) were the most favourable conditions for phytoplankton growth (especially for the main taxonomic groups chlorophytes and cryptophytes) and species richness. With short photoperiods (inundation in winter) and low temperatures, the communities transformed towards diatoms, euglenoids and cyanobacteria. In line with our predictions, a high temperature (25 °C) favoured a decline in phytoplankton species diversity. Our study shows that climate change will result in seasonal shifts in species abundance or even in their disappearance and, finally, in potential strong changes in the biodiversity and food webs of aquatic ecosystems in the future.Cuprizone is an amide compound that has been wildly used in various animal studies, such as in the investigation of remyelination in mouse model. It is important to control the amount of cuprizone dosed in animals to be consistent as different amounts may lead to different clinical observations. Cuprizone is usually administrated as a minor component (i.e., 0.3%) of a mixture with powdered or pelleted rodent chow. Its low content, combined with the complex nature of chow, represents a significant challenge for the quantification of cuprizone in the mixture.
