At last, in vivo experiments confirmed the role of MSI1 in tumor growth and chemoresistance in HCC. LEF1-AS1 might potentially affect the growth and chemoresistance of HCC cells by regulating miR-10a-5p and MSI1. LEF1-AS1 and MSI1 expression patterns were elevated, while miR-10a-5p was repressed in HCC tissues and cell lines. LEF1-AS1 combined to miR-10a-5p and regulated MSI1, thereby activating the protein kinase B (AKT) signaling pathway. Knockdown of LEF1-AS1 and MSI1 or elevation of miR-10a-5p compromised the proliferation of Huh7 cell line resistant to DDP and promoted its chemosensitivity and apoptosis. At last, these in vitro findings were also confirmed in vivo. Our results unraveled LEF1-AS1 acts as a miR-10a-5p modulator to promote chemoresistance of HCC cells by stimulating MSI1 and activating the AKT signaling pathway, which might provide a novel therapeutic target for HCC.Myxomatosis is an infectious disease caused by the myxoma virus (MYXV), which has very high mortality rates in European wild rabbits (Oryctolagus cuniculus). While sporadic cases of myxomatosis have also been reported in some hare species, these lagomorphs are considered to have a low susceptibility to MYXV infection. In the present study, we describe the spatiotemporal evolution and main epidemiological findings of novel hare MYXV (ha-MYXV or MYXV-Tol) epidemics in Iberian hares (Lepus granatensis) in Spain. In the period 2018-2020, a total of 487 hares from 372 affected areas were confirmed to be MYXV-infected by PCR. ha-MYXV outbreaks were detected in most of the Spanish regions where the Iberian hare is present. The spatial distribution was not homogeneous, with most outbreaks concentrated in the southern and central parts of Spain. Consecutive outbreaks reported in the last two years suggest endemic circulation in Spain of this emerging virus. A retrospective study carried out just after the first epidemic period (2018-2019) revealed that the virus could have been circulating since June 2018. The number of outbreaks started to rise in July, peaked during the first half of August and October and then decreased sharply until January 2019. The apparent mean mortality rate was 55.4% (median 70%). The results indicated high susceptibility of the Iberian hare to ha-MYXV infection, but apparent resistance in the sympatric hare species present in Spain and less infectivity in European rabbits. The novel ha-MYXV has had significant consequences on the health status of Iberian hare populations in Spain, which is of animal health and conservation concern. The present study contributes to a better understanding of ha-MYXV emergence and will provide valuable information for the development of control strategies. Further research is warranted to assess the impact of this emerging virus on wild lagomorph populations and to elucidate its ecological implications for Iberian Mediterranean ecosystems.
Fundamental movement skills proficiency and perceived physical competence have been shown to influence physical activity of children; the mechanisms and patterns of their relationships need to be investigated in early childhood. This study examined the direct and indirect effects of fundamental movement skills, and the mediating role of perceived physical competence on physical activity of young children.
Participants consist of 4- to 6-year-old children (N = 230; 121 girls, 109 boys). Fundamental movement skill components (i.e., locomotor, object control) were measured using the Test of Gross Motor Development-2; perceived physical competence was measured using the physical skills domain of the Pictorial Scale of Perceived Competence for young children; physical activity was monitored using pedometers over a 5-day period (n = 115). Conditional process analysis was performed to examine mediation.
Object control skills proficiency had a significant positive effect on perceived physical competence (B = 0.28, p = 0.02), and a significant positive effect on daily step counts (B = 0.18, p = 0.04). Perceived physical competence had a significant negative effect on daily step counts (B = -0.31, p = 0.003) and mediated the indirect effect of object control skills on daily step counts (B = -0.09, 95% CI = -0.17 to -0.002).
Object control skills proficiency had a significant direct effect on physical activity, and perceived physical competence mediates an indirect effect. The findings contribute to the evidence base supporting object control skills development as a factor that enables adequate physical activity of young children.

Anesthesia machines have evolved over the years to excel in delivering low-flow anesthesia (<1L fresh gas flow) in a closed-circuit system, with the obvious benefits being decreased costs and reduced emissions of greenhouse gases. At a pediatric hospital that provides over 25000 anesthetics a year, a quality improvement project was initiated with the aim of decreasing the amount of sevoflurane used per anesthetic by 20% over the course of a year.
Three Plan-Do-Study-Act cycles involving gathering comparative data, departmental education, improvement updates on our huddle board, and intraoperative confirmation rounds were completed. The bottles of sevoflurane used and the total number of anesthetics performed were collected each month. To account for the fluctuation of anesthetic cases per month, a metric of "Anesthetics Performed per Bottle of Sevoflurane Used" was created.
Compared to a prior twelve-month period, the Anesthetics Performed per Bottle of Sevoflurane Used were higher with a mean increase of 25%. The bottles of sevoflurane used per month was lower with a mean decrease of 20%. The carbon footprint of our sevoflurane use was also decreased and extrapolated over a year, and the decrease was equivalent to 70000 miles driven, over 3,200 gallons of gasoline consumed, or over 31000 pounds of coal burned.

Currently, there are no reliable clinical tools available for predicting asthma in pre-school-aged children with recurrent wheezing. The aim of this study was to evaluate the usefulness of serum periostin, YKL-40, and osteopontin biomarkers in wheezy pre-school-aged children for predicting the development of asthma in school ages.
The study was prospectively conducted between 2011 and 2017. The clinical features of the pre-school-aged children with recurrent wheezing and the levels of serum periostin, YKL-40, and osteopontin were measured. The same participants were reevaluated in school-age period, and participants with asthma were identified. Relative risk (RR) for the development of asthma was analyzed.
Of the 197 pre-school-aged children with recurrent wheezing who were reevaluated in school-age years, 32% of them had asthma. Serum periostin, YKL-40, and osteopontin levels at admission could not predict participants who would have asthma symptoms in school-age years. Consequently, we probe here the overall thermodynamic stability of cold shock protein B from Bacillus subtilis (BsCspB) in cell lysate. We found that an increase in cell lysate concentration causes a monotonic increase in the thermodynamic stability of BsCspB. This result strongly underlines the importance of considering the biological environment when inherent protein parameters are quantitatively determined. Moreover, we demonstrate that targeted application of 19 F NMR spectroscopy operates as an ideal tool for protein studies performed in complex cellular surroundings.A bifunctional catalyst comprising CuCl2 /Al2 O3 and nitrogen-doped carbon was developed for an efficient one-pot ethylene oxychlorination process to produce vinyl chloride monomer (VCM) up to 76 % yield at 250 °C and under ambient pressure, which is higher than the conventional industrial two-step process (≈50 %) in a single pass. In the second bed, active sites containing N-functional groups on the metal-free N-doped carbon catalyzed both ethylene oxychlorination and ethylene dichloride (EDC) dehydrochlorination under the mild conditions. Benefitting from the bifunctionality of the N-doped carbon, VCM formation was intensified by the surface Cl*-looping of EDC dehydrochlorination and ethylene oxychlorination. Both reactions were enhanced by in situ consumption of surface Cl* by oxychlorination, in which Cl* was generated by EDC dehydrochlorination. This work offers a promising alternative pathway to VCM production via ethylene oxychlorination at mild conditions through a single pass reactor.The study of epigenetics has greatly benefited from the development and application of various chemical biology approaches. However, there has been no resource in public databases examining the transcriptome of A. polytricha. In this study, high-throughput sequencing platform BGISEQ-500 was used to generate large amount of transcript sequences from A. polytricha for gene discovery and molecular marker development. A total of 28,102 unigenes were discovered from the assembly of clean reads. In addition, functional categorization of the gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) metabolic pathways revealed several important biological processes. GO annotation analysis presented 47 categories, with the major subcategories being catalytic activity, binding, cellular process, metabolic process, and cell. Among the five functional categories and 21 subcategories of processes discovered from KEGG, global and overview maps, carbohydrate metabolism, transport, and catabolism are the main subcategories.
