The causal link between chronic inflammation and PDAC suggests that molecular regulators of the immune system promote disease pathogenesis and/or therapeutic resistance, yet their identity is unclear. Here, we couple endoscopic ultrasound-guided fine-needle aspiration, which captures tumour biopsies from all stages, with whole transcriptome profiling of PDAC patient primary tumours to reveal enrichment of the innate immune Toll-like receptor 2 (TLR2) molecular pathway. Augmented TLR2 expression associated with a 4-gene "TLR2 activation" signature, and was prognostic for survival and predictive for gemcitabine-based chemoresistance. Furthermore, antibody-mediated anti-TLR2 therapy suppressed the growth of human PDAC tumour xenografts, independent of a functional immune system. Our results support TLR2-based therapeutic targeting for precision medicine in PDAC, with further clinical utility that TLR2 activation is prognostic and predictive for chemoresponsiveness.It is very challenging to fabricate spherical refractory material powders for additive manufacturing (AM) because of their high melting points and complex compositions. In this study, a novel technique, freeze-dry pulsated orifice ejection method (FD-POEM), was developed to fabricate spherical MoSiBTiC particles without a melting process. Elemental nanopowders were dispersed in water to prepare a high-concentration slurry, which was subsequently extruded from an orifice by diaphragm vibration and frozen instantly in liquid nitrogen. After a freeze-drying process, spherical composite particles with arbitrary composition ratios were obtained. The FD-POEM particles had a narrow size range and uniform elemental distribution. Mesh structures were formed within the FD-POEM particles, which was attributed to the sublimation of ice crystals. Furthermore, owing to their spherical morphology, the FD-POEM particles had a low avalanche angle of 42.6°, exhibiting good flowability. Consequently, the combination of FD-POEM and additive manufacturing has great potential for developing complex refractory components used in industrial applications.Epstein-Barr virus nuclear antigen 2 (EBNA2) is a transactivator of viral and cellular gene expression, which plays a critical role in the Epstein-Barr virus-associated diseases. It was reported that EBNA2 regulates gene expression by reorganizing chromatin and manipulating epigenetics. Recent studies showed that liquid-liquid phase separation plays an essential role in epigenetic and transcriptional regulation. Here we show that EBNA2 reorganized chromatin topology to form accessible chromatin domains (ACDs) of the host genome by phase separation. The N-terminal region of EBNA2, which is necessary for phase separation, is sufficient to induce ACDs. The C-terminal domain of EBNA2 promotes the acetylation of accessible chromatin regions by recruiting histone acetylase p300 to ACDs. According to these observations, we proposed a model of EBNA2 reorganizing chromatin topology for its acetylation through phase separation to explain the mechanism of EBNA2 hijacking the host genome by controlling its epigenetics.The processes underlying formation and growth of unfolded protein inclusions are relevant to neurodegenerative diseases but poorly characterized in living cells. In S. cerevisiae, inclusions formed by mutant huntingtin (mHtt) have some characteristics of biomolecular condensates but the physical nature and growth mechanisms of inclusion bodies remain unclear. We have probed the relationship between concentration and inclusion growth in vivo and find that growth of mHtt inclusions in living cells is triggered at a cytoplasmic threshold concentration, while reduction in cytoplasmic mHtt causes inclusions to shrink. The growth rate is consistent with incorporation of new material through collision and coalescence. A small remnant of the inclusion is relatively long-lasting, suggesting that it contains a core that is structurally distinct, and which may serve to nucleate it. These observations support a model in which aggregative particles are incorporated by random collision into a phase-separated condensate composed of a particle-rich mixture.An efficient composite was constructed based on aminated chitosan (NH2Cs), attapulgite (ATP) clay and magnetic Fe3O4 for adsorptive removal of Cr(VI) ions. The as-fabricated ATP@Fe3O4-NH2Cs composite was characterized by Fourier Transform Infrared Spectroscopy (FTIR), Thermal Gravimetric Analyzer (TGA), Scanning Electron Microscope (SEM), Zeta potential (ZP), Vibrating Sample Magnetometer (VSM), Brunauer-Emmett-Teller method (BET) and X-ray photoelectron spectroscope (XPS). A significant improve in the adsorption profile was established at pH 2 in the order of ATP@Fe3O4-NH2Cs(13) > ATP@Fe3O4-NH2Cs(11) > ATP@Fe3O4-NH2Cs(31) > Fe3O4-NH2Cs > ATP. The maximum removal (%) of Cr(VI) exceeded 94% within a short equilibrium time of 60 min. The adsorption process obeyed the pseudo 2nd order and followed the Langmuir isotherm model with a maximum monolayer adsorption capacity of 294.12 mg/g. In addition, thermodynamics studies elucidated that the adsorption process was spontaneous, randomness and endothermic process. Interestingly, the developed adsorbent retained respectable adsorption properties with acceptable removal efficiency exceeded 58% after ten sequential cycles of reuse. Besides, the results hypothesize that the adsorption process occurs via electrostatic interactions, reduction of Cr(VI) to Cr(III) and ion-exchanging. These findings substantiate that the ATP@Fe3O4-NH2Cs composite could be effectively applied as a reusable adsorbent for removing of Cr(VI) ions from aqueous solutions.The rapid industrial development, high population growth, and rapid urbanization of Saudi Arabia have led to increased pollution and waste levels. Every day, solid waste disposal for governments and local authorities becomes a significant challenge. Saudi Arabia produces over 15 million tonnes of solid waste annually, with a population of around 29 million. The waste production per person is estimated at between 1.5 and 1.8 kg per day per person. About 75% of the population in urban areas is concentrated, making it imperative that government steps are taken to boost the country's waste recycling and management scenario. The production of solid waste in Riyadh, Jeddah, and Dammam, three of the largest cities, exceeds seven million tonnes annually, which shows the enormity of the civic body problem. During this study, the design Expert software was involved in the optimization of process parameters during the collection of municipal solid waste (MSW) from Jeddah city. The use of design experiments and numerical optimization is quite effective in optimizing the different process parameters on the overall cost. Saudi Arabia has a critical need for a resilient waste system and agile waste management system to control its municipal solid waste quickly and environmentally friendly for achieve Saudi Vision 2030. For this study design of experiment, software was employed to optimize the cost per trip, thereby considering process parameters. It is therefore essential to examine the existing practices and future opportunities for solid waste collection, storage, and disposal. This study considered that MSW generated in Saudi Arabia which is having great potential to be converted into wealth. Hence, considering the current environment situation, energy prospective and future management strategies for MSW have also been reviewed.Anthropogenically-driven climate warming is a hypothesized driver of animal body size reductions. Less understood are effects of other human-caused disturbances on body size, such as urbanization. We compiled 140,499 body size records of over 100 North American mammals to test how climate and human population density, a proxy for urbanization, and their interactions with species traits, impact body size. We tested three hypotheses of body size variation across urbanization gradients urban heat island effects, habitat fragmentation, and resource availability. Our results demonstrate that both urbanization and temperature influence mammalian body size variation, most often leading to larger individuals, thus supporting the resource availability hypothesis. In addition, life history and other ecological factors play a critical role in mediating the effects of climate and urbanization on body size. Larger mammals and species that utilize thermal buffering are more sensitive to warmer temperatures, while flexibility in activity time appears to be advantageous in urbanized areas. This work highlights the value of using digitized, natural history data to track how human disturbance drives morphological variation.Autism spectrum disorders (ASD) are associated with disruption of large-scale brain network. Recently, we found that directed functional connectivity alterations of social brain networks are a core component of atypical brain development at early developmental stages in ASD. Here, we investigated the spatio-temporal dynamics of whole-brain neuronal networks at a subsecond scale in 113 toddlers and preschoolers (66 with ASD) using an EEG microstate approach. We first determined the predominant microstates using established clustering methods. We identified five predominant microstate (labeled as microstate classes A-E) with significant differences in the temporal dynamics of microstate class B between the groups in terms of increased appearance and prolonged duration. Our results revealed that the brain follows a universal pattern of high modular variability in metacognitive brain regions the association cortices. The brain in ageing moves towards a segregated modular structure despite presenting with increased modular heterogeneity-modules in older adults are not only segregated, but their shape and size are more variable than in young adults. In the presence of neurodegeneration, the brain maintains its segregated connectivity globally but not locally, and this is particularly visible in dementia with Lewy bodies and Parkinson's disease dementia; overall, the modular brain shows patterns of differentiated pathology.
