A major obstacle in diabetes is the metabolic or hyperglycemic memory, which lacks specific therapies. Here we show that glucose-mediated changes in gene expression largely persist in diabetic kidney disease (DKD) despite reversing hyperglycemia. The senescence-associated cyclin-dependent kinase inhibitor p21 (Cdkn1a) was the top hit among genes persistently induced by hyperglycemia and was associated with induction of the p53-p21 pathway. Persistent p21 induction was confirmed in various animal models, human samples and in vitro models. Tubular and urinary p21-levels were associated with DKD severity and remained elevated despite improved blood glucose levels in humans. Mechanistically, sustained tubular p21 expression in DKD is linked to demethylation of its promoter and reduced DNMT1 expression. Two disease resolving agents, protease activated protein C (3K3A-aPC) and parmodulin-2, reversed sustained tubular p21 expression, tubular senescence, and DKD. Thus, p21-dependent tubular senescence is a pathway contributing to the hyperglycemic memory, which can be therapeutically targeted.Water quality monitoring can inform policies that address pollution; however, inconsistent measurement and reporting practices render many observations incomparable across bodies of water, thereby impeding efforts to characterize spatial patterns and long-term trends in pollution. Here, we harmonized 9.2 million publicly available monitor readings from 226 distinct water monitoring authorities spanning the entirety of the Mississippi/Atchafalaya River Basin (MARB) in the United States. We created the Standardized Nitrogen and Phosphorus Dataset (SNAPD), a novel dataset of 4.8 million standardized observations for nitrogen- and phosphorus-containing compounds from 107 thousand sites during 1980-2018. To the best of our knowledge, this dataset represents the largest record of these pollutants in a single river network where measurements can be compared across time and space. The database was obtained through a literature review and field survey reports in the epicenter area, combined with high-resolution remote sensing image and extensive data collection and processing. The first dataset covers an area of 892 km2, including debris flows from 2008 to 2020 (an updated version). 186 debris flows affecting 79 watersheds were identified. 89 rainfall stations were collected to determine the rainfall events for the post-earthquake debris flow outbreak. The second database is a list of mitigation measures for post-earthquake debris flows, including catchment name, check dam number, coordinates, construction time, and successful mitigation date. The datasets can aid different applications, including the early warning and engineering prevention of post-earthquake debris flow, as well as provide valuable data support for research in related disciplines.

Measuring health-related diet and exercise self-efficacy is an important first step in improving healthy behaviors and health outcomes. However, we did not find a self-efficacy measurement in Chinese that is specifically targeted at diet and exercise self-efficacy among healthy adults.

The present study aimed to translate the Health-Related Diet and Exercise Self-Efficacy Scale -simplified version into Mandarin Chinese (HRDESES) and evaluate its reliability and validity in Chinese healthy adults.

The HRDESES was translated and adapted to the Chinese context, with a good content validity of 0.86 among seven experts. The survey was then carried out in 216 adults in Hunan, China. Testing of the reliability included internal consistency reliability and test-retest reliability, while validity included content validity, construct validity, and criterion validity.

The Cronbach's α of the HRDESES was 0.87 for the total scale, 0.86 for the diet subscale and 0.91 for the exercise subscale; the McDonald's ω of t HRDESES had good reliability and validity and could be used as a simple and effective tool for assessing the health-related diet and exercise self-efficacy in Chinese healthy adults.Disability prevention and preservation of independence is crucial for successful aging of older adults. To date, relatively little is known regarding disparities in independent aging in a disadvantaged older adult population despite widely recognized health disparities reported in other populations and disciplines. In the U.S., the Southeastern region also known as "the Deep South", is an economically and culturally unique region ravaged by pervasive health disparities - thus it is critical to evaluate barriers to independent aging in this region along with strategies to overcome these barriers. The objective of this narrative review is to highlight unique barriers to independent aging in the Deep South and to acknowledge gaps and potential strategies and opportunities to fill these gaps. We have synthesized findings of literature retrieved from searches of computerized databases and authoritative texts. Ultimately, this review aims to facilitate discussion and future research that will help to address the unique challenges to the preservation of independence among older adults in the Deep South region.Inflammasomes are multiprotein platforms responsible for the release of pro-inflammatory cytokines interleukin (IL)-1β and IL-18. Mouse studies have identified inflammasome activation within dendritic cells (DC) as pivotal for driving tubulointerstitial fibrosis and inflammation, the hallmarks of chronic kidney disease (CKD). However, translation of this work to human CKD remains limited. Here, we examined the complex tubular cell death pathways mediating inflammasome activation in human kidney DC and, thus, CKD progression. Ex vivo patient-derived proximal tubular epithelial cells (PTEC) cultured under hypoxic (1% O2) conditions modelling the CKD microenvironment showed characteristics of ferroptotic cell death, including mitochondrial dysfunction, reductions in the lipid repair enzyme glutathione peroxidase 4 (GPX4) and increases in lipid peroxidation by-product 4-hydroxynonenal (4-HNE) compared with normoxic PTEC. The addition of ferroptosis inhibitor, ferrostatin-1, significantly reduced hypoxic PTEC death. Human CD1c+ DC activated in the presence of hypoxic PTEC displayed significantly increased production of inflammasome-dependent cytokines IL-1β and IL-18. Treatment of co-cultures with VX-765 (caspase-1/4 inhibitor) and MCC950 (NLRP3 inflammasome inhibitor) significantly attenuated IL-1β/IL-18 levels, supporting an NLRP3 inflammasome-dependent DC response. In line with these in vitro findings, in situ immunolabelling of human fibrotic kidney tissue revealed a significant accumulation of tubulointerstitial CD1c+ DC containing active inflammasome (ASC) specks adjacent to ferroptotic PTEC. These data establish ferroptosis as the primary pattern of PTEC necrosis under the hypoxic conditions of CKD. Moreover, this study identifies NLRP3 inflammasome signalling driven by complex tubulointerstitial PTEC-DC interactions as a key checkpoint for therapeutic targeting in human CKD.
