Apoptotic pathways in platelets are important for their survival and function. Platelet apoptosis may be involved in the pathogenesis of immune thrombocytopenia (ITP), an autoimmune-mediated disease. In contrast to the intrinsic apoptosis pathway, not much is known about the extrinsic pathway mechanisms in platelets.
To investigate the expression of proteins involved in the extrinsic apoptosis pathway, including the death receptors, adaptor and regulator proteins in human platelets. To determine a possible trigger of the extrinsic apoptosis pathway in platelets.
To investigate the expression of key markers of the extrinsic pathway we used targeted immunofluorescence and flow cytometry assays. To study their expression and interaction we performed Western blotting and co-immunoprecipitation. Treated platelets with different apoptosis triggers were subjected to flow cytometry.
We could identify the protein expression of the pro-apoptotic proteins TRADD (Tumor Necrosis Factor Receptor type 1- Associated r have been yet identified.Human papillomavirus (HPV) is responsible for almost all of the 570,000 new cases of cervical cancer and approximately 311,000 deaths per year. HPV vaccination is an integral component of the World Health Organization's (WHO) global strategy to fight the disease. However, high vaccine prices enforced through patent protection are limiting vaccine expansion, particularly in low- and middle-income countries. By limiting market power, patent buyouts could reduce vaccine prices and raise HPV vaccination rates while keeping innovation incentives. We estimate the global patent buyout price as the present discounted value (PDV) of the future profit stream over the remaining patent length for Merck's HPV vaccines (Gardasil-4 and 9), which hold 87% of the global HPV vaccine market, in the range of US$ 15.6-27.7 billion (in 2018 US$). The estimated PDV of the profit stream since market introduction amounts to US$ 17.8-42.8 billion and the estimated R&D cost to US$ 1.05-1.21 billion. Thus, we arrive at a ratio of R&D costs to the patent value of the order of 2.5-6.8%. We relate this figure to typical estimates of the probability of success (POS) for clinical trials of vaccines to discuss if patent protection provides Merck with extraordinarily strong price setting power.Borrelia burgdorferi must acquire all of its amino acids (AAs) from its arthropod vector and vertebrate host. Previously, we determined that peptide uptake via the oligopeptide (Opp) ABC transporter is essential for spirochete viability in vitro and during infection. Our prior study also suggested that B. burgdorferi employs temporal regulation in concert with structural variation of oligopeptide-binding proteins (OppAs) to meet its AA requirements in each biological niche. Selleck Telacebec Herein, we evaluated the contributions to the B. burgdorferi enzootic cycle of three of the spirochete's five OppAs (OppA1, OppA2, and OppA5). An oppA1 transposon (tn) mutant lysed in the hyperosmolar environment of the feeding tick, suggesting that OppA1 imports amino acids required for osmoprotection. The oppA2tn mutant displayed a profound defect in hematogenous dissemination in mice, yet persisted within skin while inducing only a minimal antibody response. These results, along with slightly decreased growth of the oppA2tn mutant within DMCs, suggest that OppA2 serves a minor nutritive role, while its dissemination defect points to an as yet uncharacterized signaling function. Previously, we identified a role for OppA5 in spirochete persistence within the mammalian host. We now show that the oppA5tn mutant displayed no defect during the tick phase of the cycle and could be tick-transmitted to naïve mice. Instead of working in tandem, however, OppA2 and OppA5 appear to function in a hierarchical manner; the ability of OppA5 to promote persistence relies upon the ability of OppA2 to facilitate dissemination. Structural homology models demonstrated variations within the binding pockets of OppA1, 2, and 5 indicative of different peptide repertoires. Rather than being redundant, B. burgdorferi's multiplicity of Opp binding proteins enables host-specific functional compartmentalization during the spirochete lifecycle.Reaching with a visuomotor distortion in a virtual environment leads to reach adaptation in the trained hand, and in the untrained hand. In the current study we asked if reach adaptation in the untrained (right) hand is due to transfer of explicit adaptation (EA; strategic changes in reaches) and/or implicit adaptation (IA; unconscious changes in reaches) from the trained (left) hand, and if this transfer changes depending on instructions provided. We further asked if EA and IA are retained in both the trained and untrained hands. Participants (n = 60) were divided into 3 groups (Instructed (provided with instructions on how to counteract the visuomotor distortion), Non-Instructed (no instructions provided), and Control (EA not assessed)). EA and IA were assessed in both the trained and untrained hands immediately following rotated reach training with a 40° visuomotor distortion, and again 24 hours later by having participants reach in the absence of cursor feedback. Participants were to reach (1) so that the cursor landed on the target (EA + IA), and (2) so that their hand landed on the target (IA). Results revealed that, while initial EA observed in the trained hand was greater for the Instructed versus Non-Instructed group, the full extent of EA transferred between hands for both groups and was retained across days. IA observed in the trained hand was greatest in the Non-Instructed group. However, IA did not significantly transfer between hands for any of the three groups. Limited retention of IA was observed in the trained hand. Together, these results suggest that while initial EA and IA in the trained hand are dependent on instructions provided, transfer and retention of visuomotor adaptation to a large visuomotor distortion are driven almost exclusively by EA.Quantitative models of social differences have not only made major contributions to the fields of cross-cultural anthropology, psychology and sociology, but also have allowed for interdisciplinary studies that bring together engineering, life sciences, and social sciences. In this study, the authors use a data science approach to discover a set of quantitative social dimensions based on the World Values Survey, a nationally representative survey covering 60 countries and 90,000 individuals. Five national social dimensions, representing 198 questions and 56 countries are discovered using multidimensional item response theory (MIRT). They are (1) Religiosity, (2) Neutrality, (3) Fairness, (4) Skepticism, and (5) Societal Tranquility. This approach is unique from previous quantitative models because it groups responses by country and analyzes binary, nominal, and ordinal survey questions. It is possible today due to recent advancements in computing power and programming. Furthermore, this methodology tests the validity of previous quantitative dimensions and finds that some of the existing social and cultural dimensions are not clearly discernable.
