Dysregulation of adenosine 5'-monophosphate-activated protein kinase (AMPK) is closely related to antioxidant responses and autophagy of adipocytes in animal models of metabolic disorders, but its role in bovine adipose tissue during periods of stress is unknown. We hypothesized that AMPK may play important roles in the regulation of oxidative stress in adipose tissue of ketotic cows. Specific objectives were to evaluate autophagy status and AMPK activity in adipose tissue of ketotic cows, and their link with oxidative stress in isolated bovine adipocytes. Selection of 15 healthy and 15 clinically ketotic Holstein cows at 17 (±4) d postpartum was performed after a thorough veterinary evaluation for clinical symptoms an of ROS and malondialdehyde (MDA), whereas H2O2 stimulation inhibited activities of the antioxidant enzymes glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD). Addition of AMPK activator A769662 increased antioxidant response via activating NFE2L2 and its downstream targets heme oxygenase 1 (HMOX1), superoxide dismutase 1 (SOD1), catalase (CAT), and glutathione-S-transferase (GST) to improve H2O2-induced oxidative stress in adipocytes. Simultaneously, activation of AMPK increased abundance of Beclin1, SQSTM1, ATG7, ATG5, and ratio of LC3II to LC3I. In this study, we isolated 42 S. agalactiae strains in 379 milk samples from cows with subclinical mastitis on 15 dairy farms in 12 Chinese provinces. Analysis based on capsular typing and multilocus sequence typing, combined with patterns of virulence gene scanning and antimicrobial resistance, identified the lineages and populations of the isolates. We grouped the 42 isolates into 7 sequence types belonging to 6 clonal complexes, mainly CC103 (31/42 isolates; 73.8%). We identified an ST-23 strain named Sa 129 for the first time on Chinese dairy farms-this strain is usually associated with human isolates. Capsular types Ia and II were predominant in capsular typing. The prevalence of virulence profile 1 (bibA, cfb, cspA, cylE, fbsA, fbsB, hylB, and pavA) was 64.3%, and represented the main trend in China. With respect to antimicrobial resistance, most isolates were susceptible to β-lactams, rifamycin, glycopeptides, and oxazolidone; resistance to several antimicrobial agents, including lincomycin, clindamycin, and doxycycline, varied in 4 different regions. Our research provides a profile for the molecular epidemiology, multilocus sequence typing, antimicrobial resistance, and virulence gene clustering of S. agalactiae, and may be beneficial for the clinical monitoring, prevention, and control of mastitis in dairy cattle.The objective of this study was to identify the calf-level colostrum management practices associated with an adequate transfer of passive immunity (TPI; defined as serum Brix refractance ≥8.4% in the first week of life) in small-sized herds. A total of 818 calves from 61 commercial Holstein dairy farms were included in this observational cross-sectional study. For each calf, sex, colostrum delivery method, colostrum volume fed at first meal, and time to first feeding (delay between birth and first colostrum meal) were noted. Blood and colostrum samples were collected to estimate the serum and colostrum quality using Brix refractometry. To quantify the level of bacterial contamination in colostrum samples, total bacteria count and total coliform count (TCC) were measured using the Petrifilm (3M, St. Paul, MN) culture system. In this study, 68% of calves had an adequate TPI (≥8.4%). For data distribution, the 25th, 50th, and 75th percentiles were 1.3, 2.8, and 3.3 L for the colostrum volume fed at the first meal; 20.9, 23.5, and 26.5% Brix; and 1.1, 3.1, and 6.5 h for the time to first feeding of colostrum, respectively. The odds of adequate TPI were 2.6 times higher in calves receiving ≥2.5 L colostrum at their first meal, 2.9 times higher in calves receiving colostrum with ≥24.5% Brix, and 1.6 times higher in calves receiving colostrum within 3 h after birth, than in calves not meeting these criteria. In the present study, median bacterial contamination distribution (interquartile range) in the first colostrum meal was 14,000 cfu/mL (3,000-83,000 cfu/mL) for total bacteria count, and 0 cfu/mL (0-1,000 cfu/mL) for TCC. Total bacteria count and TCC were not associated with the odds of adequate TPI in the final model. Overall, these results suggest that specific calf-level colostrum management practices are associated with adequate TPI in small- to medium-sized dairy herds.Increased concentrations of some serum biomarkers are known to be associated with impaired health of dairy cows. Each herd had at least 60 cows per year, production data from 3 consecutive years, and at least 10 primiparous cows per year with their first milk recording 5 to 35 d after calving and their second milk recording 20 to 40 d after the first milk recording. In each herd, primiparous cows with a low (≤75,000 cells/mL) cow somatic cell count (CSCC) at both the first and second milk recording were categorized as low-low (LL); those with a high (>100,000 cells/mL) CSCC at both recordings were categorized as high-high (HH). Cows with high CSCC at the first recording and low at the second were categorized as high-low (HL). The annual proportions of LL, HL, and HH cows within each herd were calculated.
