In this work, it is shown how a spacecraft equipped with a thrust and subjected to a drag force can be bounded at specific altitudes as function of the parameters of the thrust. It is used nonlinear dynamics tools to find attractors, which bound the motion of the spacecraft. For a specific set of parameters of the thrust, the spacecraft is bounded to a given altitude. Several forms for the thrusts are proposed in order to bound the altitude of the spacecraft. The influence of several forms of perturbations in the altitude of the spacecraft is also investigated in this work, like the solar radiation pressure, gravity of the Moon and oblateness of the Earth. Finally, nonlinear dynamics tools are also used to investigate transfers among the bounded orbits in different altitudes.Amyloid aggregation of α-synuclein (α-syn) is closely associated with Parkinson's disease (PD) and other synucleinopathies. Several single amino-acid mutations (e.g. E46K) of α-syn have been identified causative to the early onset of familial PD. Here, we report the cryo-EM structure of an α-syn fibril formed by N-terminally acetylated E46K mutant α-syn (Ac-E46K). The fibril structure represents a distinct fold of α-syn, which demonstrates that the E46K mutation breaks the electrostatic interactions in the wild type (WT) α-syn fibril and thus triggers the rearrangement of the overall structure. Furthermore, we show that the Ac-E46K fibril is less resistant to harsh conditions and protease cleavage, and more prone to be fragmented with an enhanced seeding capability than that of the WT fibril. Our work provides a structural view to the severe pathology of the PD familial mutation E46K of α-syn and highlights the importance of electrostatic interactions in defining the fibril polymorphs.Tau is a hallmark pathology of Alzheimer's disease, and animal models have suggested that tau spreads from cell to cell through neuronal connections, facilitated by β-amyloid (Aβ). We test this hypothesis in humans using an epidemic spreading model (ESM) to simulate tau spread, and compare these simulations to observed patterns measured using tau-PET in 312 individuals along Alzheimer's disease continuum. Up to 70% of the variance in the overall spatial pattern of tau can be explained by our model. Altogether, our results provide evidence in humans that tau spreads through neuronal communication pathways even in normal aging, and that this process is accelerated by the presence of brain Aβ.The increased frequency of harmful algal blooms (HABs) is a major environmental concern worldwide, resulting not only in increased treatment costs for drinking water but also in impacts on tourism, commercial fishing and aquaculture and risks to human and animal health. Traditional strategies with ship-based approaches based on field sampling and laboratory analysis have been adopted to assess HABs. However, these methods are labour intensive and costly and do not provide synoptic views of the bloom conditions. Here, we show that the Sentinel-2 twin satellite mission of the Copernicus programme, in combination with in situ data, is a powerful tool that can offer valuable spatiotemporal information about a bloom of the dinoflagellate Lingulodinium polyedra that occurred on the SW Iberian Peninsula. Using the robust ACOLITE atmospheric correction processor combined with the normalized difference chlorophyll index (NDCI), the enhanced mapping of small blooms can be performed at a 10 m spatial resolution, revealing surface patches and a heterogeneous distribution. This research also demonstrates the improved capabilities of Sentinel-2 compared to those of Landsat-8 and Sentinel-3 for continuous monitoring. The Sentinel-3 and Sentinel-2 missions provide ecosystem observations that allow the environmental community and water managers to evaluate changes in water quality and bloom distribution and that facilitate field-based measurements. Field studies on whole colonies are limited in behavioural research, while results from lab studies allow only restricted conclusions on whole colony impacts. Methods for automated long-term investigations of behaviours within comb cells, such as brood care, were hitherto missing. In the present study, we demonstrate an innovative video method that enables within-cell analysis in honey bee (Apis mellifera) observation hives to detect chronic sublethal neonicotinoid effects of clothianidin (1 and 10 ppb) and thiacloprid (200 ppb) on worker behaviour and development. In May and June, colonies which were fed 10 ppb clothianidin and 200 ppb thiacloprid in syrup over three weeks showed reduced feeding visits and duration throughout various larval development days (LDDs). On LDD 6 (capping day) total feeding duration did not differ between treatments. Behavioural adaptation was exhibited by nurses in the treatment groups in response to retarded larval development by increasing the overall feeding timespan. Using our machine learning algorithm, we demonstrate a novel method for detecting behaviours in an intact hive that can be applied in a versatile manner to conduct impact analyses of chemicals, pests and other stressors.Blood vessels are constantly exposed to shear stress, a biomechanical force generated by blood flow. Normal shear stress sensing and barrier function are crucial for vascular homeostasis and are controlled by adherens junctions (AJs). Here we show that AJs are stabilized by the shear stress-induced long non-coding RNA LASSIE (linc00520). Silencing of LASSIE in endothelial cells impairs cell survival, cell-cell contacts and cell alignment in the direction of flow. LASSIE associates with junction proteins (e.g. PECAM-1) and the intermediate filament protein nestin, as identified by RNA affinity purification. The AJs component VE-cadherin showed decreased stabilization, due to reduced interaction with nestin and the microtubule cytoskeleton in the absence of LASSIE. This study identifies LASSIE as link between nestin and VE-cadherin, and describes nestin as crucial component in the endothelial response to shear stress. Furthermore, this study indicates that LASSIE regulates barrier function by connecting AJs to the cytoskeleton.
