Current treatment for invasive fungal diseases is limited to three classes of antifungal drugs azoles, polyenes, and echinocandins. The most recently introduced antifungal class, the echinocandins, was first approved nearly 30 years ago. The limited antifungal drug portfolio is rapidly losing its clinical utility due to the inexorable rise in the incidence of invasive fungal infections and the emergence of multidrug resistant (MDR) fungal pathogens. New antifungal therapeutic agents and novel approaches are desperately needed. Here, we detail attempts to exploit the antifungal and immunoregulatory properties of host defense peptides (HDPs) in the design and evaluation of new antifungal therapeutics and discuss historical limitations and recent advances in this quest.Human cytomegalovirus (HCMV) is a ubiquitous herpesviral pathogen that results in life-long infection. Archaea can presumably prosper in harsh and DNA-damaging environments in part due to robust DNA repair pathways but surprisingly, no DNA repair pathways unique to Archaea have been described. Here, we review the most recent advances in our understanding of archaeal DNA repair. We summarize DNA damage types and their consequences, their recognition by host enzymes, and how the collective activities of many DNA repair pathways maintain archaeal genomic integrity.General anaesthesia (GA) is implicated as a cause of postoperative sleep disruption and fatigue with part of the disturbance being attributed to a shift of the circadian clock. In this study, Drosophila melanogaster was used as a model to determine how Isoflurane affects the circadian clock at the behavioural and molecular levels. We measured the response of the clock at both of these levels caused by different durations and different concentrations of Isoflurane at circadian time 4 (CT4). Once characterized, we held the duration and concentration constants (at 2% in air for 6 h) and calculated the phase responses over the entire circadian cycle in both activity and period expression. Phase advances in behaviour were observed during the subjective day, whereas phase delays were associated with subjective night time GA interventions. The corresponding pattern of gene expression preceded the behavioural pattern by approximately four hours. We discuss the implications of this effect for clinical and research practice.Recently, with the continuous evolution of information technology, various products such as Building Information, Internet of Things (IoT), Big Data, Cloud Computing and Machine Learning have been developed and have created a lifestyle change. A smart Internet of Things (IoT) system is formed by combining the communication capabilities of the internet with control, monitoring and identification services to integrate people, things and objects. However, in some IoT environments that have a weak signal, such as remote areas, warehouses or basements, the network may become unstable, meaning that the IoT system is unable to provide efficient services. This paper therefore presents a framework that ensures the reliability of IoT system services so that even if the IoT system cannot connect to the network, the system can provide the services offline. To avoid increasing the installation cost or replacing existing traditional devices with modern smart devices, this framework can also be used to control traditional devices. The system operation is convenient because users can operate all their smart and traditional devices under the IoT system through voice commands and/or a handheld microcontroller, thus reducing the manual operation of the user. The framework proposed in this paper can be applied to various smart scenarios, including smart warehouses, smart restaurants, smart homes, smart farms and smart factories, to improve people's quality of life and convenience, and create a humane and comfortable smart living environment.The incidence of melanoma has been increasing worldwide during recent decades. The objective of the study was to analyse the trends in incidence for in situ and invasive melanoma in the Spanish region of Catalonia during the period of 2008-2017. We designed a cross-sectional study with an age-period-cohort analysis of melanoma patient data from the Network of Melanoma Centres in Catalonia. Our database covered a population of over seven million and included a total of 8626 patients with incident melanoma. The main outcome measures were crude and age-standardised incidence rates to the European 2013 standard population. Joinpoint regression models were used to evaluate the population trends. We observed an increase in the age-standardised incidence rate (per 100,000 population) of all melanoma subtypes from 11.56 in 2008 to 13.78 in 2017 with an average annual percent change (AAPC) of 3.5%. This incidence increase was seen exclusively in the older population. Moreover, the stratified analysis showed a statistically significant increase in the age-standardised incidence rate for invasive (AAPC 2.1%) and in situ melanoma (AAPC 6.5%). In conclusion, the incidence of melanoma has continued to increase in the elderly population over recent decades, with a rapidly increasing trend of in situ melanomas and the lentigo maligna subtype.
