Estrogen (ER), androgen (AR), and progesterone (PR) receptors bind directly to membrane-associated signaling molecules including mitogenic protein kinases (i.e. c-Src, AKT), G-proteins, and ion channels to mediate context-dependent actions via rapid activation of downstream signaling pathways. In addition to making direct contact with diverse signaling molecules, SRs are further fully integrated with signaling pathways by virtue of their N-terminal phosphorylation sites that act as regulatory hot-spots capable of sensing the signaling milieu. In particular, ER, AR, PR, and closely related glucocorticoid receptors (GR) share the property of accepting (i.e. sensing) ligand-independent phosphorylation events by proline-directed kinases in the MAPK and CDK families. These signaling inputs act as a "second ligand" that dramatically impacts cell fate. In the face of drugs that reliably target SR ligand-binding domains to block uncontrolled cancer growth, ligand-independent post-translational modifications guide changes in cell fate that confer increased survival, EMT, migration/invasion, stemness properties, and therapy resistance of non-proliferating SR+ cancer cell subpopulations. The focus of this review is on MAPK pathways in the regulation of SR+ cancer cell fate. MAPK-dependent phosphorylation of PR (Ser294) and GR (Ser134) will primarily be discussed in light of the need to target changes in breast cancer cell fate as part of modernized combination therapies.PURPOSE To investigate strength and structural adaptations after 12 weeks of resistance, endurance cycling, and concurrent training. METHODS Thirty-two healthy males undertook 12 weeks of resistance-only (RT; n = 10), endurance-only (END; n = 10), or concurrent resistance and endurance training (CONC; n = 12). Biceps femoris long head (BFlh) architecture, strength (3-lift 1-repetition maximum), and body composition were assessed. RESULTS Fascicle length of the BFlh reduced 15% (6%) (P less then .001) and 9% (6%) (P less then .001) in the END and CONC groups postintervention, with no change in the RT group (-4% [11%], P = .476). All groups increased BFlh pennation angle (CONC 18% [9%], RT 14% [8%], and END 18% [10%]). Thickness of the BFlh increased postintervention by 7% (6%) (P = .002) and 7% (7%) (P = .003) in the CONC and RT groups, respectively, but not in the END group (0% [3%], P = .994). Both the CONC and RT groups significantly increased by 27% (11%) (P less then .001) and 33% (12%) (P less then .001) in 3-lift totals following the intervention, with no changes in the END cohort (6% [6%], P = .166). No significant differences were found for total body (CONC 4% [2%], RT 4% [2%], and END 3% [2%]) and leg (CONC 5% [3%], RT 6% [3%], and END 5% [3%]) fat-free mass. CONCLUSIONS Twelve weeks of RT, END, or CONC significantly modified BFlh architecture. This study suggests that conventional resistance training may dampen BFlh fascicle shortening from cycling training while increasing strength simultaneously in concurrent training. Furthermore, the inclusion of a cycle endurance training stimulus may result in alterations to hamstring architecture that increase the risk of future injury. Therefore, the incorporation of endurance cycling training within concurrent training paradigms should be reevaluated when trying to modulate injury risk.BACKGROUND The interleukin-12/23p40-subunit-inhibitor ustekinumab significantly improved spondylitis-related symptoms through Week 24 in psoriatic arthritis (PsA) patients with peripheral arthritis and physician-reported spondylitis (PA-PRS) in PSUMMIT-1&2. We collected longitudinal fecal samples from eight unrelated male-female pairs of marmoset monkeys prior to pairing and for 8 weeks following pairing. We then sequenced 16S rRNA to characterize the changes in the gut microbiome that resulted from the pairing. Marmoset pairs had a higher similarity in gut microbiome communities after pairing than before pairing. We discovered sex differences in the degrees of change in gut microbiome communities following pairing. Specifically, the gut microbiome communities in males exhibited greater dissimilarity from the prepairing stage (baseline) than the gut microbiome communities in females. Conversely, females showed a gradual stabilization in the rate of the gut microbiome community turnover.
