However, cortical neurons in thalamocortical slices from knock-in mice had reduced miniature inhibitory post-synaptic current amplitude and prolonged spontaneous thalamocortical oscillations. Thus, the Gabrb3+/D120N knock-in mouse recapitulated human Lennox-Gastaut syndrome seizure types and behavioural abnormalities and was caused by impaired inhibitory GABAergic signalling in the thalamocortical loop. In addition, treatment with antiepileptic drugs and cannabinoids ameliorated atypical absence seizures in knock-in mice. This congenic knock-in mouse demonstrates that a single-point mutation in a single gene can cause development of multiple types of seizures and multiple behavioural abnormalities. The knock-in mouse will be useful for further investigation of the mechanisms of Lennox-Gastaut syndrome development and for the development of new antiepileptic drugs and treatments.The North European species of Elaphomyces section Elaphomyces (Eurotiales, Pezizomycotina) are studied. Three new species, E. citrinopapillatus, E. pusillus, and E. roseoviolaceus are introduced and verified by morphology and sequence data from ITS, nuclear LSU, mitochondrial SSU, and β-tubulin. A lectotype for Elaphomyces granulatus is selected. Elaphomyces granulatus and E. muricatus are epitypified with sequenced material from the Femsjö region in South Sweden. Elaphomyces striatosporus is epitypified with sequenced material from the vicinity of the type locality in Norway. A key to all species of Elaphomyces occurring in Denmark, Norway, and Sweden is provided.To clarify the diversity of plant-parasitic Alternaria species in Japan, diseased samples were collected, and fungal isolates established in culture. We examined 85 isolates representing 23 species distributed in 14 known sections based on conidial morphology and DNA phylogeny. Three species were found to be new, A. cylindrica, A. paragomphrenae and A. triangularis. Furthermore, a lectotype was designated for A. gomphrenae, and epitypes for A. cinerariae, A. gomphrenae, A. iridicola, and A. japonica. Species boundaries of isolates were also clarified by studying phenotypes and determining host ranges. Alternaria gomphrenae and related species in sect. Alternantherae were recognized as distinct species owing to their host specificity. Among the species infecting Apiaceae, the pathogenicity of A. cumini and a novel species, A. triangularis ex Bupleurum, were confirmed as host specific. Another novel species, A. cylindrica, proved to be host specific to Petunia. Alternaria iridicola was recognized as a large-spored species in sect. Alternaria, being host specific to Iris spp. On the other hand, the experimental host ranges of three morphologically and phylogenetically distinct species infecting Brassicaceae (A. brassicae, A. brassicicola, and A. japonica) showed almost no differences. Alternaria brassicicola and A. porri were even found on non-host plants. In general, host ranges of Alternaria species correlated with morphology and molecular phylogeny, and combining these datasets resulted in clearer species boundaries.Our understanding of the systematics of red yeasts has greatly improved with the availability of sequence data and it is now clear that the majority of these fungi belong to three different classes of Pucciniomycotina (Basidiomycota) Agaricostilbomycetes, Cystobasidiomycetes, and Microbotryomycetes. The small, black, cupulate Helvella specimens have mostly been assigned to Helvella corium, a broadly defined morpho-species. Recent phylogenetic analyses, however, have revealed an aggregate of species hidden under this name. We performed a multispecies coalescent analysis to re-assess species limits and evolutionary relationships of the Helvella corium species aggregate in the Nordic countries. To achieve this, we used morphology and phylogenetic evidence from five loci - heat shock protein 90 (hsp), translation elongation factor 1-alpha (tef), RNA polymerase II (rpb2), and the 5.8S and large subunit (LSU) of the nuclear ribosomal DNA. All specimens under the name Helvella corium in the larger university fungaria of Norway, Sweden and Denmark were examined and barcoded, using partial hsp and/or rpb2 as the preferential secondary barcodes in Helvella. Additional fresh specimens were collected in three years (2015-2018) to obtain in vivo morphological data to aid in species discrimination. The H. corium species aggregate consists of seven phylogenetically distinct species, nested in three divergent lineages, i.e. H. corium, H. alpina and H. pseudoalpina sp. nov. in the /alpina-corium lineage, H. alpestris, H. macrosperma and H. nannfeldtii in the /alpestris-nannfeldtii lineage, and H. alpicola as a weakly supported sister to the /alpestris-nannfeldtii lineage. Among the seven species, the ribosomal loci expressed substantial variation in evolutionary rates, suggesting care in the use of these regions alone in delimitation of Helvella species. Altogether, 469 out of 496 available fungarium specimens were successfully barcoded.
