The aim of this investigation was to determine whether substance P (SP) is able to protect the endothelium against inflammatory stress. To this end, aortic endothelial cells were pre-treated with SP, followed by tumour necrosis factor α (TNF-α), and cellular responses were evaluated using a combination of cell biology and quantification assays, as well as western blot analyses. Our results show that TNF-α enhanced ECM expression and reduced NO production within 4 hours, promoting immune cell adhesion to the endothelium and monocyte chemoattractant protein-1 (MCP-1) secretion from aortic smooth muscle cells. However, SP treatment ameliorated TNF-α-induced endothelial impairment by maintaining low ECM levels. Our data suggest that this protective effect is mediated by Akt activation and NO-enriched conditions. The inhibition of aortic endothelial cell injury by SP also reduced MCP-1 production in aortic smooth muscle cells. Together, our data indicate that SP can protect aortic endothelial and smooth muscle cells from inflammatory injury, which suggests that SP may prevent cardiovascular disease.Nanotechnology has revolutionized diverse fields, which include agriculture, the consumer market, medicine, and other fields. Here, we analysed, for the first time, NUMTs in the ovine genome. We obtained 760 alignment matches covering 513.8 kbp of the sheep nuclear genome. After a merging step, we identified 390 NUMT regions with a total length of ~720 kbp, representing 0.02% of the nuclear genome. We discovered copies of all mitochondrial regions and found that most NUMT regions are intergenic or intronic. Ovine NUMTs are mostly not transcribed. However, we identified within some of the NUMTs, potential new genes encoding nuclear humanin isoforms. To rule out the possibility that the identified NUMTs could be artifacts of the Oar Rambouillet v1.0 genome assembly, we validated experimentally nine NUMT regions by PCR amplification. As we found several NUMT regions showing high similarity to the mitochondrial genome that potentially could pose a risk to ovine DNA mitochondrial studies, special care must be taken for the selection of primers for PCR amplification of mitochondrial DNA sequences.

Grazed grass is an important component of the majority of beef production systems used in temperate climates. Compared to concentrate-fed beef, 'grass-fed' beef can command a premium in some markets based on perceived differences in appearance and sensory characteristics. The influence of grazed grass per se, as well as the duration of grazing, on selected sensory characteristics of beef within a heifer production system was examined.

In general, fat from grass-fed cattle was more yellow than fat from similar cattle fed concentrates, whereas muscle from grass-fed cattle was darker than muscle from cattle fed concentrates. At the same carcass weight, muscle from grass-fed cattle had a lower fat concentration than cattle fed concentrates. In the most extreme situation examined, whereby early-maturing heifers were fed concentrates ad libitum from weaning or grazed grass/conserved grass throughout life, until slaughtered at a similar carcass weight (260 kg) and differed in age by 5months, beef was rated similarly for tenderness and a range of flavours by a trained sensory panel.
