MicroRNAs are non-coding RNAs that act to downregulate the expression of target genes by translational repression and degradation of messenger RNA molecules. Individual microRNAs have the ability to specifically target a wide array of gene transcripts, therefore allowing each microRNA to play key roles in multiple biological pathways. miR-324 is a microRNA predicted to target thousands of RNA transcripts and is expressed far more highly in the brain than in any other tissue, suggesting that it may play a role in one or multiple neurological pathways. Here we present data from the first global miR-324-null mice, in which increased excitability and interictal discharges were identified in vitro in the hippocampus. RNA sequencing was used to identify differentially expressed genes in miR-324-null mice which may contribute to this increased hippocampal excitability, and 3'UTR luciferase assays and western blotting revealed that two of these, Suox and Cd300lf, are novel direct targets of miR-324. Characterisation of microRNAs that produce an effect on neurological activity, such as miR-324, and identification of the pathways they regulate will allow a better understanding of the processes involved in normal neurological function and in turn may present novel pharmaceutical targets in treating neurological disease.Colorectal cancer is a leading cause of death in the western world. The main datum that is employed to guide treatment and prognosis are related to the pathological stage and the genetics of the cancer. Recent electron-microscopic study of the colonic border has suggested a difference between the micro-anatomy of the mesenteric border11, compared to the anti-mesenteric. With colorectal cancer increasing in incidence, the more information that we can employ to guide and tailor patient centred management, the better. A pilot study to test the hypothesis that the circumferential location on the colonic wall, mesenteric or anti-mesenteric, has an impact on the mortality rate associated with right-sided colon cancer. All patients undergoing a right hemicolectomy for non-metastatic adenocarcinoma between 2010 and 2013 were included (155 patients in total). T and N stage were recorded. There was no statistical difference between the groups for age or sex. Survival rates were then calculated according to the location of the cancer and analysed using Kaplan-Meir survival calculations. 100 patients were included in the final analysis. read more 90 patients had cancer on the antimesenteric border. The T and N stage were not statistically different between the two groups. The mean all-cause survival was 44 months for the mesenteric group and 77 for the antimesenteric (P = 0.002). In this study, we sought to investigate the impact of dietary intake of ω3 fatty acid on the development of atherosclerosis, and demonstrate the mechanism of action by identifying anti-inflammatory lipid metabolite. Mice were exposed to a high-fat diet (HFD) supplemented with either conventional soybean oil or α-linolenic acid-rich linseed oil. We found that as mice became obese they also showed increased pulsatility and resistive indexes in the common carotid artery. In sharp contrast, the addition of linseed oil to the HFD improved pulsatility and resistive indexes without affecting weight gain. Histological analysis revealed that dietary linseed oil inhibited foam cell formation in the aortic valve. Lipidomic analysis demonstrated a particularly marked increase in the eicosapentaenoic acid-derived metabolite 12-hydroxyeicosapentaenoic acid (12-HEPE) in the serum from mice fed with linseed oil. When we gave 12-HEPE to mice with HFD, the pulsatility and resistive indexes was improved. Indeed, 12-HEPE inhibited the foamy transformation of macrophages in a peroxisome proliferator-activated receptor (PPAR)γ-dependent manner. These results demonstrate that the 12-HEPE-PPARγ axis ameliorates the pathogenesis of atherosclerosis by inhibiting foam cell formation.Conservation agriculture (CA)-based practices have been promoted and recouped, as they hold the potential to enhance farm profits besides a consistent improvement in soil properties. A 7 years' field experiment consisting of three crop establishment practices viz., zero-till flatbed (ZTFB), permanent beds (PNB), conventional system (CT) along with the three-nutrient management; nutrient expert-based application (NE), recommended fertilization (RDF), and farmers' fertilizer practice (FFP), was carried out from 2013 to 2020. The CA-based practices (ZTFB/PNB) produced 13.9-17.6% greater maize grain-equivalent yield (MGEY) compared to the CT, while NE and RDF had 10.7-20% greater MGEY than the FFP. PNB and ZTFB gave 28.8% and 24% additional net returns than CT, while NE and RDF had 22.8% and 17.4% greater returns, respectively over FFP. PNB and ZTFB had 2.3-4.1% (0.0-0.20 m soil layers) lower bulk density than the CT. Furthermore, microbial biomass carbon (MBC) increased by 8-19% (0.0-0.50 m soil layers) in ZTFB/PNB over the CT, and by 7.6-11.0% in NE/RDF over FFP. Hence, CA-based crop establishment coupled with the NE or RDF could enhance the yields, farm profits, soil properties of the maize-chickpea rotation, thereby, could sustain production in the long run.Schizophrenia is among the most debilitating neuropsychiatric disorders. The aim of this study was to investigate possible alterations in the resting alpha oscillatory activity in normal population high on schizotypy trait, a physiological condition known to be severely altered in patients with schizophrenia. Direct comparison of resting-state EEG oscillatory activity between Low and High Schizotypy Group (LSG and HSG) has revealed a clear right hemisphere alteration in alpha activity of the HSG. Specifically, HSG shows a significant slowing down of right hemisphere posterior alpha frequency and an altered distribution of its amplitude, with a tendency towards a reduction in the right hemisphere in comparison to LSG. Furthermore, altered and reduced connectivity in the right fronto-parietal network within the alpha range was found in the HSG. Crucially, a trained pattern classifier based on these indices of alpha activity was able to successfully differentiate HSG from LSG on tested participants further confirming the specific importance of right hemispheric alpha activity and intrahemispheric functional connectivity.
