ological research.The main purpose of the article was to compare different scenarios of biopolymer production and their impacts on the environment using Life Cycle Assessment. Three alternative polyhydroxyalkanoates (PHA amorphous PHA and poly(3-hydroxybutyrate), P(3HB)) production scenarios were considered to assess its environmental impact Scenario A - Production of mcl-PHA/P(3HB) from crude vegetable oil; Scenario B - Production of P(3HB) with biodiesel by-product; Scenario C - Production of mcl-PHA/P(3HB) from used vegetable oil. Subject to the scenario considered, it was shown that the environmental efficiency of PHA production is highly dependent on carbon sources used, and it is strongly supporting production of mcl-PHA instead of P(3HB). As LCA study shows, due to low yield of P(3HB) in comparison to mcl-PHA production in considered processes, all the P(3HB) production scenarios have higher impacts than the production of mcl-PHA. Production processes based on bacterial fermentation had its impacts related mostly to the raw materials used and to its separation phase. Additionally, using secondary materials instead of raw ones, namely used oil instead of virgin oil, gives significant improvement with regard to environmental impact. The resource efficiency is also the identified as the key factor with sensitivity analysis that indicates the possible increase of biopolymer yield as the most beneficial factor. Preferably production of these microbial polymers should be integrated into biorefinery blocks, where such waste stream arises (e.g. biodiesel production plant).The formation of chemical and biological heterogeneity in lakes can be favored by physical and morphometric constrains. This study describes the results of four whole-lake field campaigns carried out in Lake Como (north Italy) during thermal stratification. The aim was to analyze the distribution of chemical-biological variables in a multi-basin lake as a result of internal and external physical drivers and constrains. Lake Como has a y-like shape encompassing three main sub-basins northern, south-eastern, and south-western. Field data underlined the presence of chemical-biological gradients between the south-western basin and the rest of the lake and the propagation of a fresher water-plume (formed by the two main northern inflows) into the northern basin and then into the south-western closed arm. The use of a three-dimensional hydrodynamic model showed a periodic movement of this plume that tends to enter the south-western basin first and then to return toward north, moving forward and back through the junction of the three arms. The entrance into the eastern basin, instead, occurs only secondarily. Wavelet transform analysis revealed common periodicity between the plume movement and the action of different external and internal lake-stressors, including the discharge of the main inflow (period centered at 1, 3.3, and 6.5 day), the wind intensity (0.5 and 1 day) and the two main basin-scale internal wave motions (3.3 day and 7.1 day). The periodic movement of the fresher water-plume enhances the water exchange and reduces the chemical-biological gradients between the western closed basin and the main lake, playing a crucial role in distributing both inorganic and organic materials at the lake basin-scale.In December 2019, an outbreak of atypical pneumonia (Coronavirus disease 2019 -COVID-19) associated with a novel coronavirus (SARS-CoV-2) was reported in Wuhan city, Hubei province, China. The outbreak was traced to a seafood wholesale market and human to human transmission was confirmed. The rapid spread and the death toll of the new epidemic warrants immediate intervention. The intra-host genomic variability of SARS-CoV-2 plays a pivotal role in the development of effective antiviral agents and vaccines, as well as in the design of accurate diagnostics. We analyzed NGS data derived from clinical samples of three Chinese patients infected with SARS-CoV-2, in order to identify small- and large-scale intra-host variations in the viral genome. We identified tens of low- or higher- frequency single nucleotide variations (SNVs) with variable density across the viral genome, affecting 7 out of 10 protein-coding viral genes. The majority of these SNVs (72/104) corresponded to missense changes. The annotation of the identified SNVs but also of all currently circulating strain variations revealed colocalization of intra-host as well as strain specific SNVs with primers and probes currently used in molecular diagnostics assays. Moreover, we de-novo assembled the viral genome, in order to isolate and validate intra-host structural variations and recombination breakpoints. The bioinformatics analysis disclosed genomic rearrangements over poly-A / poly-U regions located in ORF1ab and spike (S) gene, including a potential recombination hot-spot within S gene. Our results highlight the intra-host genomic diversity and plasticity of SARS-CoV-2, pointing out genomic regions that are prone to alterations. The isolated SNVs and genomic rearrangements reflect the intra-patient capacity of the polymorphic quasispecies, which may arise rapidly during the outbreak, allowing immunological escape of the virus, offering resistance to anti-viral drugs and affecting the sensitivity of the molecular diagnostics assays.Compulsory co-payments limit access and may compromise quality in primary care. Patient Chosen Gap Payments (PCGPs) allow patients to specify a (voluntary) out-of-pocket contribution, creating an incentive for patient-centred care without the need for complex outcomes-based funding formulae. It is not yet known if widespread use of PCGP services is consistent with consumer preferences. We conducted a discrete choice experiment (DCE) in a sample of the adult Australian general population (n = 1457) during April 2019 to simulate patient choice between alternative primary care services and describe preferences for PCGP services. Participants also completed a supplementary valuation task in which participants reported their intended PCGP contribution for PCGP services. Finally, we conducted policy-simulations to predict market shares when PCGP clinics operate alongside the two existing models of primary care funding in Australia. Results suggest that patients prefer shorter wait time, longer consults, lower compulsory copayments, services with higher patient satisfaction ratings, choice of doctor and $0 suggested voluntary contribution for PCGP services.
