The prostate-specific membrane antigen (PSMA) has been demonstrated in numerous studies to be expressed specifically on prostate carcinoma cells and on the neovasculature of several other cancer entities. However, the simultaneous expression of PSMA on both, tumor cells as well as tumor vessels remains unclear, even if such "dual" expression would constitute an important asset to facilitate sufficient influx of effector cells to a given tumor site. Exosomes have recently emerged as a component with great potential involved as mediators in cellular communication. The isolation and identification of exosomes has made it possible for them to be used in cell-free therapies. The purposes of this study are (i) to detect exosomes released into adipose-derived MSC conditioned cell culture medium, (ii) to identify exosome morphology, and (iii) to carry out a complete characterization of said exosomes. Moreover, it is aimed at determining which method for exosome isolation would be best to use. Precipitation has been identified as a highly useful method of exosome isolation since it provides higher efficiency and purity values than other methods. While several signaling pathways are well known to regulate somatic stem cells, the underlying mechanisms remain largely unexplored. Here, we demonstrate a cell-intrinsic role for the OvoL family transcription factor, Shavenbaby (Svb), in balancing self-renewal and differentiation of Drosophila intestinal stem cells. We find that svb is a downstream target of Wnt and EGFR pathways, mediating their activity for stem cell survival and proliferation. This requires post-translational processing of Svb into a transcriptional activator, whose upregulation induces tumor-like stem cell hyperproliferation. In contrast, the unprocessed form of Svb acts as a repressor that imposes differentiation into enterocytes, and suppresses tumors induced by altered signaling. We show that the switch between Svb repressor and activator is triggered in response to systemic steroid hormone, which is produced by ovaries. Therefore, the Svb axis allows intrinsic integration of local signaling cues and inter-organ communication to adjust stem cell proliferation versus differentiation, suggesting a broad role of OvoL/Svb in adult and cancer stem cells.In animal breeding, parents of the next generation are usually selected in multiple stages, and the initial stages of this selection are called preselection. Preselection reduces the information available for subsequent evaluation of preselected animals and this sometimes leads to bias. The objective of this study was to establish the minimum information required to subsequently evaluate genomically preselected animals without bias arising from preselection, with single-step genomic best linear unbiased prediction (ssGBLUP). We simulated a nucleus of a breeding program in which a recent population of 15 generations was produced. In each generation, parents of the next generation were selected in a single-stage selection based on pedigree BLUP. However, in generation 15, 10% of male and 15% of female offspring were preselected on their genomic estimated breeding values (GEBV). These GEBV were estimated using ssGBLUP, including the pedigree of all animals in generations 0-15, genotypes of all animals in generations 13-15 and phenotypes of all animals in generations 11-14. In subsequent ssGBLUP evaluation of these preselected animals, genotypes and phenotypes from various groups of animals were excluded one after another. We found that GEBV of the preselected animals were only estimated without preselection bias when genotypes and phenotypes of all animals in generations 13 and 14 and of the preselected animals were included in the subsequent evaluation. We also found that genotypes of the animals discarded at preselection only helped in reducing preselection bias in GEBV of their preselected sibs when genotypes of their parents were absent or excluded from the subsequent evaluation. We concluded that to prevent preselection bias in subsequent ssGBLUP evaluation of genomically preselected animals, information representative of the reference data used in the evaluation at preselection and genotypes and phenotypes of the preselected animals are needed in the subsequent evaluation.William Henry Perkin, Jr. FRS, the son of the inventor of mauve and other commercial dyes and credited for initiating the industrialization of chemistry, was himself a notable chemist. He was the Professor of Organic Chemistry at Manchester from 1892-1912 and then was the Waynflete Professor of Chemistry at Oxford and the first Head of the Dyson Perrins Laboratory from 1912-1929. One of Perkin's graduate students and research assistants at Manchester was Robert Robinson, subsequently Sir Robert Robinson, FRS and recipient of the 1947 Nobel Prize in Chemistry. Perkin and Robinson had perhaps the most productive and broad collaboration between a professor and one's student. Together, during and after Robinson's student days, they had 71 joint publications, 25 of which involved just the two of them, 17 of which involved the structure determination of strychnine, and eight of which were published after Perkin's death in 1929. Upon Perkin's early death, Robinson succeeded him as the fourth Waynflete Professor of Chemistry at the Dyson Perrins Laboratory, Oxford University.
