Hydrogen sulfide (H2S), an emerging gas transmitter, has been shown to be involved in multiple intracellular physiological and biochemical processes. In this study, the effects of hydrogen sulfide coupled with calcium on cadmium removal and resistance in Phanerochaete chrysosporium were examined. see more The results revealed that H2S enhanced the uptake of calcium by P. chrysosporium to resist cadmium stress. The removal and accumulation of cadmium by the mycelium was reduced by H2S and Ca2+ pretreatment. Moreover, oxidative damage and membrane integrity were alleviated by H2S and Ca2+. Corresponding antioxidative enzyme activities and glutathione were also found to positively respond to H2S and Ca2+, which played an important role in the resistance to cadmium-induced oxidative stress. The effects of hydroxylamine (HA; a hydrogen sulfide inhibitor) and ethylene glycol-bis-(2-aminoethylether)-N,N,N',N'-tetraacetic acid (EGTA; a calcium chelator) toward H2S and Ca2+ and their cross-interactions confirmed the positive roles and the potential crosstalk of H2S and Ca2+ in cadmium stress resistance. These findings imply that the protective effects of H2S in P. chrysosporium under cadmium stress may occur through a reduction in the accumulation of cadmium and promotion of the antioxidant system, and the H2S-regulated pathway may be associated with the intracellular calcium signaling system.Key points• Altered monoterpenoid tolerance mainly related to altered activity of efflux pumps.• Increased tolerance to geranic acid surprisingly caused by decreased export activity.• Reduction of export activity can be beneficial for biotechnological conversions.The abundance and distribution of dissolved Re (DRe) were determined in the freshwater part of the Krka River (Croatia), which drains a karst landscape, and in the salinity gradient of its highly stratified estuary. Due to the low DRe concentration, a batch procedure consisting of a pre-concentration step using an anion exchange resin (Dowex) and analysis of DRe in 8 M HNO3 eluate using high-resolution inductively coupled plasma mass spectrometry (HR ICP-MS) was applied. Due to potentially inconsistent recoveries, which ranged from 60 to 87%, quantification was performed using the isotope dilution technique (ID). DRe concentrations in the Krka River increased downstream, from 6.2 pM at the spring site to 11.9 pM upstream of the estuary region. Weathering of the surrounding carbonate lithology is assumed to be the source of the natural Re. Two specific anomalies were registered a strong increase in DRe concentration due to anthropogenic input near the town of Knin (27.5 pM) and a decrease at a downstream site caused by subsurface input of freshwater from the Zrmanja River, resulting in a relatively low DRe concentration (8.5 pM). In the estuarine region, a near-conservative behavior of DRe was found in the salinity gradient of the upper surface layer, with DRe concentrations ranging from 18 to 38 pM. Anthropogenic input was suspected within the estuarine segment near the urban area, causing a small positive deviation from the conservative line. The participants were asked to count/withhold counting the number of O/X letters in the count/no-count task, and to calculate the sum of all digits (i.e., 1/0) in the digital accumulation task. Taken together, in the count/no-count task, the fronto-central N2 might reflect response inhibition or conflict and the parietal P3 might reflect attentional resource allocation but not response inhibition.Subjective estimates of elapsed time are sensitive to the fluctuations in an emotional state. While it is well known that dangerous and threatening situations, such as electric shocks or loud noises, are perceived as lasting longer than safe events, it remains unclear whether anticipating a threatening event speeds up or slows down subjective time and what defines the direction of the distortion. We examined whether the anticipation of uncertain visual aversive events resulted in either underestimation or overestimation of perceived duration. The participants did a temporal bisection task, where they estimated durations of visual cues relative to previously learnt long and short standard durations. The colour of the to-be-timed visual cue signalled either a 50% or 0% probability of encountering an aversive image at the end of the interval. The cue durations were found to be overestimated due to anticipation of aversive images, even when no image was shown afterwards. Moreover, the overestimation was more pronounced in people who reported feeling more anxious while anticipating the image. These results demonstrate that anxiogenic anticipation of uncertain visual threats induce temporal overestimation, which questions a recently proposed view that temporal underestimation evoked by uncertain threats is due to anxiety.

To study the association of myopia progression with the morphological changes of optic disc and β-peripapillary atrophy (β-PPA) in 8-11years old primary school students.

This study was a prospective, school-based investigation. This study included 610 children (1008 eyes) who were continuously observed and had data available from 2016 to 2017 in the Sanhe Cohort Study of the Risk Factors for Myopia (SCSRFM). The children underwent a comprehensive eye examination including measurement of visual acuity, autorefractometry, and posterior segment of the eye. β-PPA regions and optic disc ovality index were identified and measured on the fundus photographs.

The prevalence of myopia was 72.62% (732/1008) in 2016. In myopic children, the prevalence of the vertical β-PPA, the horizontal β-PPA, and the oval optic disc were 75.68% (554/732), 75.96% (556/732) and, 11.61% (85/732) respectively. From 2016 to 2017, with the progression of vertical β-PPA, horizontal β-PPA, area of β-PPA, and optic disc ovality index, thudy of the third-grade primary school students showed that with the progression of myopia and the growth of AL, β-PPA and optic disc ovality index also changed. There was a positive correlation between the change of β-PPA and optic disc ovality index and the progression of myopia diopter and AL.

To evaluate reliability and repeatability of computer-assisted measurements of cone photoreceptor metrics on Heidelberg Engineering Spectralis™ High Magnification Module (HMM™) Automatic Real-time Tracking (ART™) images.

We analyzed HMM™ images in three separate study arms. Computer-assisted cone identification software was validated using an open-access adaptive optics (AO) dataset. We compared results of the first arm to data from AO and histology. We evaluated intersession repeatability of our computer-assisted cone analysis in the second arm. We assessed the capability of HMM™ to visualize cones in the presence of pathology in the third arm.

We included 10 healthy subjects in the first arm of our study, 5 additional healthy participants in the second arm and 5 patients in the third arm. In total, we analyzed 225 regions of interest on HMM™ images. We were able to automatically identify cone photoreceptors and assess corresponding metrics at all eccentricities between 2 and 9° from the fovea. Cone density significantly declined with increasing eccentricity (p = 4.890E-26, Friedman test). With increasing eccentricity, we found a significant increase in intercell distance (p = 2.196E-25, Friedman test) and nearest neighbor distance (p = 1.997E-25, Friedman test). Cone hexagonality ranged between 71 and 85%. We found excellent automated intersession repeatability of cone density counts and spacing measurements. In pathology, we were also able to repeatedly visualize photoreceptors.

Computer-assisted cone photoreceptor analysis on Spectralis™ HMM™ images is feasible, and most cone metrics show excellent repeatability. HMM™ imaging may be useful for photoreceptor analysis as progression marker in outer retinal disease.

Previously, we reported that the intravenous injection of bone marrow-derived cells (BMDC) infected with lentivirus expressing the human RPE65 gene resulted in the programming of BMDC to promote visual recovery in a mouse model of age-related macular degeneration (AMD). The aim of this study was to characterize the spatial and temporal recruitment of these programmed BMDC to the retinal pigment epithelial (RPE) layer.

C57BL/6J female mice received a subretinal injection of AAV1-SOD2 ribozyme to knock down (KD) superoxide dismutase 2 (SOD2) and induce AMD-like pathology. BMDC were isolated from GFP

mice and infected with a lentivirus expressing RPE65. One month after SOD2 KD, fifty thousand GFP

RPE65-BMDC were injected in the mouse tail vein. Animals were terminated at different time points up to 60min following cell administration, and localization of GFP

cells was determined by fluorescence microscopy of neural retina and RPE flat mounts and tissue sections.
